Abstract
【解決手段】本発明によれば、新規の可溶性IFN−α/β結合タンパク質、その製造法およびそれを含有する医薬組成物が提供され、過剰量のIFN−αまたはIFN−βが存在する状態をともなう疾患などの予防、治療、診断などに有用である。

Description
患者血清はダン アデルカ(Dan Aderka)博士(イチロフ ホスピタル(Ichilov Hospital)、テル アビブ、イスラエル)より入手した。正常個体の1000-倍濃縮(15)粗尿タンパク質は、セロノ ラボラトリーズ(Serono Laboratories)(ローマ、イタリア)より入手し、アガロース ヒドラジド(シグマ社製)に固定した抗−IFN−α MAb No.74.3は、前に述べられた(22)ように調製しジ−N−ヒドロキシ−サクシンイミジル スベレート(DSS)はピアス社より入手した。
組換えIFN−α2(2×108ユニット/mg)はチューリッヒ大学のシー ワイズマン(C.Weismann)博士により提供された。IFN−α2はクロラミンT法(35)の変法により標識した。手短に述べると、7μgのIFN−α2を1mg/mlのクロラミンT存在下で1mCiのNa125Iを用いて標識し(氷上、20秒間)4×107cpm/μgの比活性とした。IFN−αBはチバ−ガイギー社より入手し、IFN−αCおよびIFN−βはインター−ラボ(Inter-Lab)社、(ネス−ジオナ(Ness-Ziona)、イスラエル)より入手した。
MAb 21.4を用いたヒト血清および尿のウェスタンブロッティング
正常ヒト血清(NHS)、毛様細胞性白血病(HCL)血清および粗尿タンパク質のサンプルを、非還元条件下で(36)SDS−PAGEに付し、ニトロセルロース膜(シュライヒャー アンド シュエル(Schleicher and Schuell)社製、0.45μm)に25mMトリス、1.92mMグリシン、20%メタノール中、電気的に転写した。転写につづいて膜をブロッキング緩衝液(0.05%ツイーン-20および0.02%アジ化ナトリウムを含むPBS中、10%脱脂ミルク)とインキュベートし、組換えIFN−αRAに対して調製した抗−IFN−αRA抗体21.4と室温にて2時間インキュベートした。ニトロセルロース膜を0.05%ツイーン-20含有PBSで洗浄し、125I−ヤギ 抗−マウス抗体(ブロッキング緩衝液中0.7×106cpm/ml)と4℃にて一晩インキュベートした。膜を洗浄後、乾燥してオートラジオグラフィーを行なった。
未処理血清および尿のクロスリンキングおよび免疫沈降
血清または尿のサンプルを、100倍過剰量の非標識IFN−α2の非存在下または存在下で125I−IFN−αB(300,000cpm)と4℃にて1時間インキュベートした。そののちジメチルスルフォキシド(Me2SO)中に溶解したDSSを最終濃度が1mMとなるように添加し、混液を4℃にて20分間放置した。1Mトリス塩酸pH7.5および1M塩化ナトリウムを最終濃度が100mMとなるように添加して反応を停止した。アガロースヒドラジドに固定した抗−IFN−α MAb 74-3(22)(25μl、7mg/ml)を添加して試料を免疫沈降させた。4℃にて一晩インキュベートしたのち、ビーズをPBSで3回洗浄し、2%メルカプトエタノールを含むサンプル用緩衝液中に懸濁して、その上清をSDS−PAGE、続いてオートラジオグラフィーにて分析した。
IFN−α/β結合タンパク質の単離
製造業者の指示に従って、アフィゲル−10(Affigel−10)(1ml、バイオラッド(BioRad)社製)にIFN−α2(5mg)を結合させてカラムにつめた。粗尿タンパク質(1000倍濃縮、10g、250ml)を流速0.25ml/minでカラムに付した。カラムは0.5M塩化ナトリウム含有リン酸緩衝性生理食塩水(PBS)250ml、つぎにPBS(10ml)を用いて洗浄した。そののち結合したタンパク質(75μg)を0.25mMクエン酸pH2.2で溶出し、直ちに1M炭酸ナトリウムで中和した。1mlずつ画分を集めた。その画分をSDS−PAGEおよび銀染色によって分析し(図3)、タンパク量をフルオレサミンを用いて測定した。種々の画分について、実施例2に述べたように125I−IFN−α2とのクロスリンキング、免疫沈降、SDS−PAGEおよびオートラジオグラフィーを行なって分析した。未処理尿中ならびにIFN−α2−アガロースおよびIFN−β−アガロースカラムの両方から溶出した画分中にIFN−α/β結合タンパク質が見出された(図4)。
タンパク質配列分析
実施例3のスペロース12カラムの画分27(4μgタンパク質)をPVDF膜(プロ−スピン(Pro-Spin)、アプライド バイオシステムズ(Applied Biosystems)社、米国)に吸着させ、その膜をモデル475 マイクロシーケンサー(アプライド バイオシステムズ社製)で、タンパク質配列分析に付した。
純粋なIFN−α/β結合タンパク質のクロスリンキングおよび免疫沈降
IFN−α2またはIFN−βカラムで精製した尿のIFN−α/β結合タンパク質またはさらにスペロース12カラムで精製した尿のIFN−α/β結合タンパク質のサンプルを、100倍過剰量の非標識IFN−α2の非存在下または存在下にて125I−IFN−α2(106cpm)と4℃で1時間インキュベートした。そののちジメチルスルホキシド(Me2SO)中に溶解したDSSを最終濃度が1mMとなるように添加し、混液を4℃にて20分間放置した。1Mトリス塩酸pH7.5および1M塩化ナトリウムを最終濃度が100mMとなるように添加して反応を停止した。アガロースヒドラジドに固定した抗−IFN−α MAb 74-3(22)(25μl、7mg/ml)を添加して試料を免疫沈降させた。4℃にて一晩インキュベートしたのち、ビーズをPBSで3回洗浄し、2%メルカプトエタノールを含むサンプル用緩衝液中に懸濁して、その上清をSDS−PAGE(7.5%アクリルアミド)、続いてオートラジオグラフィーにて分析した。分子量約60Kの特異的なクロスリンク産物がすべての画分に見られ、過剰の非標識IFN−α2存在下では完全に消えていた(図7)。
クロスリンキング実験における125I−IFN−α2の各種I型IFNでの置き換え
均一なIFN−α/β結合タンパク質(40ng)を3〜670倍過剰量の非標識IFN−α2、IFN−αB、IFN−αC、IFN−βまたはIFN−γの非存在下または存在下で125I−IFN−α2(106cpm)と4℃にて1時間インキュベートした。そののちジメチルスルフォキシド(Me2SO)中に溶解したDSSを最終濃度が1mMとなるように添加し、混液を4℃にて20分間放置した。1Mトリス塩酸pH7.5および1M塩化ナトリウムを最終濃度が100mMとなるように添加して反応を停止した。アガロースヒドラジドに固定した抗−IFN−α MAb 74-3(22)(25μl、7mg/ml)を添加して試料を免疫沈降させた。4℃にて一晩インキュベートしたのち、ビーズをPBSで3回洗浄し、2%メルカプトエタノールを含むサンプル用緩衝液中に懸濁して、その上清をSDS−PAGE(7.5%アクリルアミド)、続いてオートラジオグラフィーにて分析した(図6)。125I−IFN−α2のIFN−α/β結合タンパク質との結合の阻止において、すべてのI型IFNが等しく有効であることを見出した。この結果より、IFN−α/β結合タンパク質が、前記したIFN−αBのみに特異的なIFN−αRA(10)とは異なることが示された。
IFN−α/β結合タンパク質によるIFN−αおよびIFN−β活性の阻害
実験は、IFN活性測定に用いられる細胞変性効果(CPE)阻害分析(37)に基づいて行なった。抗ウイルス活性は、ヒトIFN−α、IFN−βおよびIFN−γのNIH参照標準に対して検量した。下記の手順を用いた:
96−平底ウェルプレート中、10%FCS添加MEM100μl中に20,000細胞/ウェルを播種してWISH細胞(ヒト羊膜細胞、ATCC CCL−25)のコンフルエントな細胞単層を調製した(1日目)。プレートを5%CO2存在下、37℃でインキュベートした。2日目に標準IFN−β(10μl、1000U/ml)を別のプレート中(65μl)に連続的に2倍希釈した。可溶性IFN−α/βR(10μl、4μg/ml、IFN−αカラムならびにスペロース12カラムにて精製)を標準IFN−β希釈液中に添加した。1000U/mlのIFN−αを中和するに足る抗−IFN−α中和モノクローナル抗体(25μl、No.9−3(22))をIFN−α2アガロースカラムからリークしたIFN−α2を中和するために各ウェルに加えた。混液を37℃にて90分間放置したのち、WISH細胞の入ったプレートに移し、直ちに水疱性口内炎ウイルス(VSV、50μl、220PFU/細胞)を用いて攻撃した。プレートを37℃で一晩インキュベートした。標準IFN−βに対して検量し、分析を行なった。3日目、ウイルスの攻撃から約20時間後に、細胞変性効果を顕微鏡下で評点し、単層をクリスタルバイオレットにて染色した(図9)。
マウスの免疫、細胞融合およびスクリーニング
Balb/cマウスに尿のIFN−α/β結合タンパク質の均一なサンプル(1μg/マウス/インジェクション)を4回注射する。転化sRIA(後述)によって最高の力価を示すマウスを融合用に選択する。脾臓リンパ球をNSO−1ミエローマ変異(NSO)細胞(シー ミルスタイン(C.Milstein)、MRC、ケンブリッジ、英国により提供)と融合する。転化sRIAにより抗−IFN−α/βR抗体の存在に関してハイブリドーマの上清を調べる。96−ウェルプレートをアフィニティで精製したヤギ抗−マウス抗体で被覆し、続いてハイブリドーマの上清さらに125I−IFN−α/βRを添加する。平行して、IFN−α/β抗ウイルス活性の中和に関してハイブリドーマを調べる。抗−IFN−α/βR抗体を分泌していると認められたハイブリドーマを限界希釈法によってクローン化しさらにもう一度クローン化する。
IFN−α/β結合タンパク質の組換え生産
組換えIFN−α/β結合タンパク質を生産するために、ポリペプチドの配列を、配列番号3で示されるペプチド配列(256パーミュテーション)および配列番号4で示されるペプチド配列(192パーミュテーション)に対応する2つのオリゴヌクレオチド配列に逆翻訳する。

Claims (14)
- IFN−α/β結合タンパク質であって、ヒト血清から得ることができ、ヒト血清から得た場合、分子量が約50kDであるIFN−α/β結合タンパク質またはその融合タンパク質もしくは塩。
- ヒト血清をクロマトグラフィーのカラムに通し、引き続き精製を行なうことによりヒト血清からIFN−α/β結合タンパク質を単離することを含んでなるIFN−α/β結合タンパク質の製造法。
- IFN−α2が結合したクロマトグラフィーのカラムを用いる請求項2記載の製造法。
- IFN−βが結合したクロマトグラフィーのカラムを用いる請求項2記載の製造法。
- 抗−IFN−α/β結合タンパク質抗体が結合したクロマトグラフィーのカラムを用いる請求項2記載の製造法。
- 請求項1記載のIFN−α/β結合タンパク質をコードするヌクレオチド配列を含むDNA分子。
- 請求項6記載のDNA分子を含んでなり、形質転換宿主細胞内でIFN−α/β結合タンパク質または同じ生物学的活性を有するタンパク質を発現することのできる複製可能な発現媒体。
- 請求項7記載の発現媒体で形質転換された宿主細胞。
- 原核細胞である請求項8記載の形質転換宿主細胞。
- 真核細胞である請求項8記載の形質転換宿主細胞。
- 請求項8記載の形質転換宿主細胞を培養し、IFN−α/β結合タンパク質を回収することを含んでなる組換え可溶性IFN−α/βレセプタータンパク質の製造法。
- 請求項1記載の組換えIFN−α/β結合タンパク質。
- 請求項1または12記載のIFN−α/β結合タンパク質を含有する医薬組成物であって、糖尿病、自己免疫疾患、移植拒絶およびエイズからなる群より選択される過剰量のIFN−αまたはIFN−βが内在的に合成されたり、外から投与されたような状態を治療するための医薬組成物。
- 状態が自己免疫疾患または炎症である請求項13記載の医薬組成物。
