本発明は、グルコースセンサに関し、特に血液等の溶液中のグルコース濃度を測定するための光導波路型グルコースセンサ及び発色試薬膜固定化方法に係わる。

【従来の技術】
平面光導波路型グルコースセンサは、基板表面に一対のグレーティング(回折格子)を配置し、一方のグレーティングに光が入射され、もう一方のグレーティングから光が出射される。これらグレーティング間に位置する基板表面に単一の光導波路層を形成し、さらにこの光導波路層上に分子認識機能および情報変換機能を有する膜を形成した構造を有する。

このような構造のグルコースセンサにおいて、検体から抽出した血液等に含まれる生体分子を分子認識機能および情報交換機能を有する膜に接触した状態でレーザ光のような光をグレーティングを通して光導波路層に入射させ、エバネッセント波を発生させ、光導波路層上の膜による血液等に含まれる生体分子との反応に起因するエバネッセント波の変化量をグレーティングから出射される光を受光する受光素子により検出して、血液等に含まれる生体分子を分析する(例えば、特許文献1参照。)。

しかしながら、従来のグルコースセンサは光導波路層が単層で、ここで発生するエバネッセント波の変化量の検出には感度的に限界があり、また光導波路層上の膜構造から、検体から抽出した血液等に含まれる極微量の生体分子分析には不向きであるという問題があった。

本発明は、検体から抽出した、例えば体液中の極微量のグルコースを高感度かつ高精度で分析することが可能な光導波路型グルコースセンサと、その光導波路型グルコースセンサにおける発色試薬膜固定化方法を提供することを目的とする。

【課題を解決するための手段】
上記目的を達成するために、本発明の第1の特徴は、基板と、基板に接して設けられた第1光導波路層と、第1光導波路層に接して、互いに離間して設けられた入射側グレーティング及び出射側グレーティングと、第1光導波路層に接して、入射側グレーティングと出射側グレーティングの間に設けられた第1光導波路層より高屈折率を有する第2光導波路層と、第2光導波路層上に形成された発色試薬分子の単分子層からなる発色試薬膜と、発色試薬膜に接して設けられた酵素と触媒を含有する酵素触媒固定化層とを具備する光導波路型グルコースセンサを要旨とする。

(第1の実施の形態)
本発明の第1の実施の形態に係る光導波路型グルコースセンサは、図1に示すように、基板1と、基板1に接して設けられた第1光導波路層2と、第1光導波路層2の両端部付近の基板1に接して設けられた入射側グレーティング3aおよび出射側グレーティング3bと、第1光導波路層2に接して設けられ、入射側グレーティング3aと出射側グレーティング3bの間に位置する第1光導波路層2より高屈折率を有する第2光導波路層4と、第2光導波路層4上に形成された発色試薬分子の単分子層からなる発色試薬膜8と、発色試薬膜8に接して設けられた酵素と触媒を含有する酵素触媒固定化層9とを具備する。

(イ)まず、図2(a)に示すように、例えばホウケイ酸ガラスからなる基板1の表面を例えば380〜400℃程度の硝酸カリウム溶液塩等のイオン交換溶液に浸漬してカリウム、ナトリウム等の高屈折率元素をイオン交換することにより第1光導波路層2を形成する。その後、化学蒸着(CVD)法等により、第1光導波路層2上に例えばTiO2、ZnO、LiNbO3、GaAs、ITO、Ta2O5またはポリイミド等の第1光導波路層2より屈折率の高い材料の層12を形成する。

(ロ)次に、図2(b)に示すように第1光導波路層2より屈折率の高い材料の層12をフォトエッチング技術等を用いてパターニングすることにより第1光導波路層2の中央付近に外周が傾斜した形状の第2光導波路層4を形成する。

(チ)次に、保護膜5の開口部6から露出した酵素触媒固定化層9上に、例えば有機モノマー、光反応開始剤およびデカン酸メチルのような貧溶媒を含む溶液をスピン塗布し、乾燥させ光重合させた後、洗浄処理することにより図3の(h)に示すように多孔質膜10を形成する。

(リ)最後に、チタンアルコキシドを加水分解して原液を作り、これをスピン塗布し、乾燥後加熱して酸化チタン薄膜を得るゾルゲル方法等によって、図3(i)に示すようにメッシュ状導電性薄膜11を多孔質膜10上に形成する。そして検体100に電界(例えばパルス状の電界)が印加されるように、検体100の電極板101に電源102の陽極を接続し、メッシュ状導電性薄膜11に電源102の陰極を接続する。この結果、図1に示す光導波路型グルコースセンサが完成する。

つづいて本発明の第1の実施の形態に係る発色試薬膜固定化方法について、図4、図5を参照しながら詳細に説明する。図1に示した光導波路型グルコースセンサは、第2光導波路層4上での発色試薬と生体分子との発色反応に起因するエバネッセント波の吸光量を検出することで、その生体分子量を測定しようとするものである。しかしエバネッセント波は第2光導波路層4表面から約100nmの厚さまでしかしみ出さない。そこで本発明の第1の実施の形態では、分析精度を向上させるため発色試薬膜8を第2光導波路層4表面から約100nm以内に設けるように形成する。

(I) シランカップリング剤溶液の調製:
例えば図4(A)に示すようなエポキシ基を有する構造をしたシランカップリング剤を、アルコールと少量の純水からなる溶液に溶かし、シランカップリング剤溶液を調製する。

(II) 発色試薬溶液の調製:
発色試薬として、例えば図4(B)に示すようにアミノ基を有する3,3’,5,5’−テトラメチルベンジジンを使用し、この発色試薬を室温で純水に溶かし、発色試薬溶液を調製する。

(III) 膜構造発色試薬の固定:
1)第2光導波路層4表面にCVD法等により、図4(a)に示す金属酸化膜8aを形成する。なお、第2光導波路層4が金属酸化物で形成されているのであれば、その表面が図4(a)のような状態になっているので特にこの操作は必要としない。
2)(I)で調製したシランカップリング剤溶液を図4(a)に示す金属酸化膜8a表面上にスピン塗布するか、図4(a)に示した構造体を(I)で調製したシランカップリング剤溶液中に30分から60分程度浸漬した後に純水で洗浄する。
3)続いて120℃で焼き付け、図4(b)に示す金属シラン化膜8bを形成する。
4)次に、(II)で調製した発色試薬溶液を金属シラン化膜8b上にスピン塗布するか、図4(b)に示した構造体を(II)で調製した発色試薬溶液中に30分から60分程度浸漬した後に純水で洗浄する。
5)続いて常温で乾燥させ、図4(B)の発色試薬のアミノ基と金属シラン化膜8bのエポキシ基と結合させ、図4(c)に示す発色試薬膜8を形成する。 この固定方法により形成される発色試薬膜8の立体的な構造は、図5に示すように、発色試薬分子がシランカップリング剤によって第2光導波路層4表面に対してほぼ垂直に結合し配列した単分子層となる。そして、発色試薬分子が酵素触媒固定化層9と接する構造となる。この単分層子は高々数nm〜数十nmレベルの厚さしかないため、発色を起こす部位が第2光導波路層表面から100nm以内のところに位置するようになる。そのために、発色すれば、確実にエバネッセント波を吸収することになる。

続いて、検体100のグルコースの測定方法について説明する。図1に示す光導波路型グルコースセンサのメッシュ状導電性薄膜11側に検体100、例えば人体の皮膚の一部を接触させ、さらに検体100の別の部位に電極板101を接触させる。ここで、多孔質膜10の上部に配置されたメッシュ状導電性薄膜11と電極板101の間に電源102から所望の電界(例えばパルス状電界)を印加すると、検体100からグルコースを含む体液が多孔質膜10を通して酵素触媒固定化層9に効率よく抽出される、いわゆる微侵襲作用がなされる。この体液中のグルコースは、酵素触媒固定化層9でグルコースオキシダーゼ(GOD)等との酸化酵素反応により過酸化水素(H2O2)を発生させる。さらに過酸化水素からFe−EDTAを触媒としてラジカル酸素原子(O*)が生成される。このラジカル酸素原子により発色試薬膜8で発色試薬を発色させる。この反応を模式すると(2)、(3)、(4)のようになる。

このような状態で、図1に示すように光源21(例えば波長650nmの半導体レーザ)および受光素子22をそれぞれグルコースセンサの基板1の裏面左側および右側に配置する。そして光源21からレーザ光を偏光フィルタ23を通してグルコースセンサの基板1裏面側に入射する。そのレーザ光は基板1を通して入射側グレーティング3aと第1光導波路層2の界面で屈折されて、その第1光導波路層2を伝播される。第1光導波路層2を伝播されるレーザ光は、この第1光導波路層2より高屈折率の第2光導波路層4との界面で2つのモード(TMモード、TEモード)に分割され、TMモードレーザ光は第1光導波路層2を、TEモードレーザ光は第2光導波路層4を伝播する。このとき、発色試薬膜8における発色試薬の発色に基づく変化(例えば吸光度変化)によりこの発色試薬膜8直下の第2光導波路層4を伝播する光の強度が変化する。第1光導波路層2と第2光導波路層4を伝播した光は、受光素子22側端付近において第1光導波路層2と第2光導波路層4の界面で再び結合、干渉するため、第2光導波路層4を伝播する光の強度変化を増幅される。その結果、第2光導波路層4を伝播する光の極微な変化も偏光フィルタ24を通して受光素子22で検出される。

したがって、光導波路を第1光導波路層2と第2光導波路層4により構成し、本発明の発色試薬膜固定化方法により、発色試薬膜8を第2光導波路層4上に接して100nm以内に設けることで、検体100(人体の皮膚下)から抽出されたグルコースと酵素の反応による発色試薬の発色に基づく第2光導波路層4を伝播する光の極微な変化を第1光導波路層2と第2光導波路層4の界面で検出できる。したがって、検体100から抽出された体液中の極微量のグルコースを一層高感度で分析できる。

また、図1に示すように入射側グレーティング3aと出射側グレーティング3bを含む第1光導波路層2上に保護膜5を設けることによって、第1光導波路層2および入射側グレーティング3aと出射側グレーティング3bに外部圧力が直接加わるのを防止できる。このため、第1光導波路層2および入射側グレーティング3aと出射側グレーティング3bに外部圧力が直接加わることに伴う部材の屈折率変化によって、その第1光導波路層2を伝播する光が外部に漏れるのを防止できる。しかも、保護膜5を第1光導波路層2に比べて低屈折率材料で形成することによって、その第1光導波路層2を伝播する光を第1光導波路層2と保護膜5との界面で効果的に全反射させて第1光導波路層2内に封じ込めることができる。したがって、第1光導波路層2から光が外部に漏れるのを防止できる。その結果、検体100中の極微量のグルコースをより高感度で分析することが可能となる。

さらに、第1光導波路層2を伝播する光が保護膜5との界面から保護膜5側に漏れた場合、迷光トラッピング膜7を保護膜5の表面(開口部6の内面を除く)に設けることによって、漏光を迷光トラッピング膜7でトラップすることができる。すなわち、第1光導波路層2を伝播する光が保護膜5との界面から保護膜5側に漏れると、保護膜5表面と外界(空気)との屈折率の差により漏光は保護膜5表面で全反射して第2光導波路層4に迷光として入射されるため、前述した検体100中のグルコースの検出感度を低下させる。これに対し、迷光トラッピング膜7を保護膜5の表面に設けることによって、漏光が保護膜5表面で全反射することなく迷光トラッピング膜7でトラップできる。したがって、漏光が第2光導波路層4に迷光として入射するのを防止でき、検体100中のグルコースをより高感度で分析することが可能となる。

本発明の第1の実施の形態の変形例として、図6に示すように電源102の陰極を導電性のある金属酸化物でできた第2光導波路層4に接続し、検体100の電極板101に電源102の陽極を接続する。このようにすることでメッシュ状導電性薄膜11は不要となる。そして多孔質膜10側に検体100、例えば人体の皮膚の一部を接触させ、さらに検体100の別の部位に電極板101を接触させる。ここで、この多孔質膜10の後方に配置された導電性の第2光導波路層4と電極板101の間に電源102から所望の電界(例えばパルス状電界)を印加すると、微侵襲作用により、検体100からグルコースを含む体液が多孔質膜10を通して酵素触媒固定化層9に効率よく抽出される。

ここで、図1と図6は微侵襲作用を利用して体液を抽出する場合について示しているが、本発明のその他の実施の形態として、検体100への電界の印加に代えて、検体100への穿刺等によって得られる血液等を直接メッシュ状導電性薄膜11または多孔質膜10上から滴下して分析することも可能であることは勿論である。したがって、本発明の技術的範囲は上記の説明から妥当な特許請求の範囲に係る発明特定事項によってのみ定められるものである。
