本発明は、バイオケミカルセンサに関し、特に水溶液中および生物の生体分子の量、性状を認識するためのバイオケミカルセンサに係わる。

例えば特開平9−61346号公報には平面光導波路型バイオケミカルセンサが開示されている。このバイオケミカルセンサは、基板表面に光が入射、放出される一対のグレーティングを形成し、これらグレーティング間に位置する前記基板表面に単一の光導波路層を形成し、さらにこの光導波路層上に分子認識機能および情報変換機能を有する膜を形成した構造を有する。

このような構造のバイオケミカルセンサにおいて、検体中の血液等の生体分子を前記分子認識機能および情報変換機能を有する膜に接触した状態でレーザ光のような光を前記グレーティングを通して前記光導波路層に入射させ、エバネッセント波を発生させ、前記光導波路層上の膜による前記検体中の生体分子との反応に起因する前記エバネット波の変化量を前記グレーティングから放出される光を受光する受光素子により検出して前記検体中の生体分子を分析する。

【発明が解決しようとする課題】
しかしながら、従来のバイオケミカルセンサは光導波路層が単層で、ここで発生するエバネッセント波の変化量に検出には感度的に限界があり、また前記光導波路層上の膜構造から検体中の極微量の生体分子の分析に不向きであるという問題があった。

本発明に係る光導波路型バイオケミカルセンサは、基板と、

前記第1光導波路層の両端表面にそれぞれ形成されたグレーティングと、

【発明の実施の形態】
以下、本発明の光導波路型バイオケミカルセンサを図面を参照して詳細に説明する。

(第1実施形態)
図1は、この第1実施形態に用いられる光導波路型バイオケミカルセンサを示す断面図である。

例えばガラスからなる基板1は、表面にこの基板1より高屈折率の第1光導波路層2が形成されている。この第1光導波路層2は、例えばカリウム、ナトリウム、銀等の高屈折率元素を前記ガラス成分とイオン交換することにより形成される。2つのグレーティング3は、この第1光導波路層2の両端部(両端部近傍)表面にそれぞれ形成されている。これらのグレーティング3は、例えば酸化チタン、酸化亜鉛、ニオブ酸リチウム、GaAsにより作られ、前記第1光導波路層2より高い屈折率を有する。外周が傾斜した形状の第2光導波路層4は、前記2つのグレーティング3の間に位置する前記第1光導波路層2上に形成されている。この第2光導波路層4は、例えば酸化チタン、酸化亜鉛、ニオブ酸リチウム、GaAsにより作られ、前記第1光導波路層より高い屈折率を有する。

次に、前述した光導波路型バイオケミカルセンサを製造方法の一例を図2、図3を参照して説明する。

まず、図2の(a)に示すように例えばホウケイ酸ガラスからなる基板1の表面を例えば380〜400℃の溶融硝酸カリウム、溶融硝酸ナトリウムのようなイオン交換溶液に浸漬してカリウム、ナトリウム等の高屈折率元素をイオン交換することにより第1光導波路層2を形成した後、この第1光導波路層2上に例えば酸化チタン、酸化亜鉛、ニオブ酸リチウム、GaAsのような第1光導波路層より屈折率の高い材料の層11を例えばCVD法等により形成する。

次いで、図2の(b)に示すように前記層11をフォトエッチング技術でパターニングすることにより前記第1光導波路層2の中央付近に外周が傾斜した形状の第2光導波路層4を形成するとともに、前記第1光導波路層2上に2つのグレーティング形成層12を第2光導波路層4の両側に隣接するように形成する。つづいて、これらグレーティング形成層12をフォトエッチング技術でパターニングすることにより図2の(C)に示すように前記第1光導波路層2の両端付近上に2つのグレーティング3を形成する。

次いで、前記グレーティング3および第2光導波路層4を含む第1光導波路層2上に例えば感光性フッ素系樹脂のような前記グレーティング3に比べて低屈折率を有する材料の被膜を塗布した後、露光、現像処理を施すことにより図2の(d)に示すように前記第2光導波路層4の表面に対応する箇所に矩形状の開口部6を有する感光性フッ素系樹脂からなる保護膜5を形成する。つづいて、図3の(e)に示すように保護膜5の表面(前記開口部6の内面を除く)に例えば顔料、色素入り樹脂からなる迷光トラッピング層7をCVD法、スピンコート法、真空蒸着法により形成する。

次いで、図3の(f)に示すように前記保護膜5の開口部6から露出した前記第2光導波路層4表面部分に生体分子認識機能および情報変換機能を有する機能膜8を形成する。つづいて、図3の(g)に示すように前記保護膜5の開口部6から露出した前記機能膜8上に例えばゲルから作られる多孔質膜9を形成する。この後、パルス状の電界が印加されるメッシュ状導電性薄膜10を前記多孔質膜9に直接接触するか、または所望の間隔をあけて対向配置することにより前述した図1に示す光導波路型バイオケミカルセンサを製造する。

バイオケミカルセンサの生体分子認識機能および情報変換機能を有する機能膜8に設けられた多孔質膜9側に生体分子を含む検体を接触させ、この多孔質膜9の前方に配置されたメッシュ状導電性薄膜10に外部から所望のパルス状電界を印加すると、前記検体中の生体分子が前記多孔質膜9を通して前記機能膜8に効率よく抽出される、いわゆる微浸襲作用がなされる。この生体分子は、前記機能膜8との間でバイオケミカル反応がなされる。このような状態で、図1に示すように光源21(例えば波長650nmの半導体レーザ)および受光素子のような検出器22をそれぞれバイオケミカルセンサの基板1の裏面左側および右側に配置し、前記光源21からレーザ光を偏光フィルタ23を通して前記バイオケミカルセンサの基板1裏面側に入射すると、そのレーザ光は基板1を通してグレーティング3と第1光導波路層2の界面で屈折されてその第1光導波路層2を伝播される。第1光導波路層2を伝播されるレーザ光は、この第1光導波路層2より高屈折率の第2光導波路層4との界面で2つのモード(Tモード、TEモード)に分割され、各光導波路層2,4を伝播する。このとき、生体分子認識機能および情報変換機能を有する機能膜8における前記検体中の生体分子のバイオケミカル反応に基づく変化(例えば吸光度変化)によりこの機能膜8直下の第2光導波路層4を伝播する光の強度が変化する。このように第1、第2の光導波路層2,4を伝播した光は、検出器22側端付近においてそれら光導波路層2,4の界面で再び結合、干渉するため、前記第2光導波路層4を伝播する光の強度変化を増幅できる。その結果、前記機能膜8における検体中の生体分子のバイオケミカル反応に基づく第2光導波路層4を伝播する光の極微な変化も偏光フィルタ24を通して前記検出器22で検出することが可能になる。

したがって、図1に示す構造の第1実施形態の光導波路型バイオケミカルセンサによれば、生体分子認識機能および情報変換機能を有する機能膜8の前方にメッシュ状導電性薄膜10を配置し、この導電性薄膜10に外部から所望のパルス状電界を印加することにより検体中の生体分子を前記機能膜8に効率よく抽出する、いわゆる微浸襲作用を図ることができ、かつ光導波路を第1、第2の光導波路層2,4により構成して前記機能膜8における検体中の生体分子のバイオケミカル反応に基づく第2光導波路層4を伝播する光の極微な変化もそれらの層2,4界面での検出できるため、前記検体中の極微量の生体分子を高感度で分析することができる。

また、図1に示すようにグレーティング3を含む前記第1光導波路層2上に保護膜5を形成することによって、前記第1光導波路層2およびグレーティング3を外部圧力が直接加わるのを防止できる。このため、前記第1光導波路層2およびグレーティング3に外部圧力が直接加わることに伴うそれら部材の屈折率変化によって、その第1光導波路層2を伝播する光が外部に漏れるのを防止できる。しかも、保護膜5を前記第1光導波路層2に比べて低屈折率材料で形成することによって、その第1光導波路層2を伝播する光を第1光導波路層2と保護膜5との界面で効果的に反射させて第1光導波路層2内に封じ込めることができるため、前記第1光導波路層2から光が外部に漏れるのを防止できる。その結果、検体中の極微量の生体分子をより高感度で分析することが可能になる。

さらに、迷光トラッピング層7を前記保護膜5の表面(開口部6の内面を除く)に形成することによって、第1光導波路層2を伝播する光が前記保護膜5との界面から保護膜5側に漏れた場合、その漏光を迷光トラッピング層7でトラップすることができる。

すなわち、第1光導波路層2を伝播する光が前記保護膜5との界面から保護膜5側に漏れると、前記保護膜5表面と外界(空気)との屈折率の差により前記漏光は保護膜5表面で全反射して第2光導波路層4に迷光として入射されるため、前述した検体中の生体分子の検出感度を低下させる。これに対し、前記迷光トラッピング層7を前記保護膜5の表面に形成することによって、前記漏光が保護膜5表面で全反射することなくトラッピング層7でトラップできるため、その漏光が第2光導波路層4に迷光として入射するのを防止でき、検体中の生体分子をより高感度で分析することが可能になる。

さらに、生体分子認識機能および情報変換機能を有する機能膜8を多孔質膜9で覆うことによって、前記機能膜8に対する検体中の不純物の影響、つまり機能膜8における検体中の生体分子のバイオケミカル反応に基づく変化によりこの機能膜8直下の第2光導波路層4を伝播する光の強度変化に対して作用する外乱を低減でき、検体中の極微量の生体分子をより一層高感度で分析することが可能になる。

なお、前述した第1実施形態では単一の機能膜を用いたが、図4に示すように第2光導波路層4上に形成された情報変換機能を有する第1機能膜81とこの第1機能膜81上に形成された生体分子認識機能を有する第2機能膜82とに分割して構成してもよい。

図5は、この第2実施形態に用いられる光導波路型バイオケミカルセンサを示す断面図である。なお、図5において前述した第1実施形態で説明した図1と同様な部材は同符号を付して説明を省略する。

図5に示す光導波路型バイオケミカルセンサは、2つのグレーティング3の間に位置する第1光導波路層2上にこの第1光導波路層2より高屈折率の導電性材料、例えば酸化錫(SnO2)、インジウム錫酸化物(ITO)から作られる第2光導波路層13を形成し、かつこの第2光導波路層13に所望のパルス状の電界を印加する構造を有する。ただし、パルス状電界に代えて、直流電流を印加することも可能である。

このような構成の光導波路型バイオケミカルセンサにおいて、その生体分子認識機能および情報変換機能を有する機能膜8に設けられた多孔質膜9側に生体分子を含む検体を接触させ、この多孔質膜9の後方に配置された導電性材料から作られる第2光導波路層13に外部から所望のパルス状電界を印加すると、前記検体中の生体分子が前記多孔質膜9を通して前記機能膜8に効率よく抽出される、いわゆる微浸襲作用がなされる。この生体分子は、前記機能膜8との間でバイオケミカル反応がなされる。このような状態で、図5に示すように光源21(例えば波長650nmの半導体レーザ)および受光素子のような検出器22をそれぞれバイオケミカルセンサの基板1の裏面左側および右側に配置し、前記光源21からレーザ光を偏光フィルタ23を通して前記バイオケミカルセンサの基板1裏面側に入射すると、そのレーザ光は基板1を通してグレーティング3と第1光導波路層2の界面で屈折されてその第1光導波路層2を伝播される。第1光導波路層2を伝播されるレーザ光は、この第1光導波路層2より高屈折率の第2光導波路層13との界面で2つのモード(Tモード、TEモード)に分割され、各光導波路層2,4を伝播する。このとき、生体分子認識機能および情報変換機能を有する機能膜8における前記検体中の生体分子のバイオケミカル反応に基づく変化(例えば吸光度変化)によりこの機能膜8直下の第2光導波路層4を伝播する光の強度が変化する。このように第1、第2の光導波路層2,13を伝播した光は、検出器22側端付近においてそれら光導波路層2,13の界面で再び結合、干渉するため、前記第2光導波路層4を伝播する光の強度変化を増幅できる。その結果、前記機能膜8における検体中の生体分子のバイオケミカル反応に基づく第2光導波路層13を伝播する光の極微な変化も偏光フィルタ24を通して前記検出器22で検出することが可能になる。

したがって、図5に示す構造の第2実施形態の光導波路型バイオケミカルセンサによれば、生体分子認識機能および情報変換機能を有する機能膜8の後方に位置する第2光導波路層13を導電性材料から作り、この第2光導波路膜13に外部から所望のパルス状電界を印加することにより検体中の生体分子を前記機能膜8に効率よく抽出する、いわゆる微浸襲作用を図ることができ、かつ光導波路を第1、第2の光導波路層2,13により構成して前記機能膜8における検体中の生体分子のバイオケミカル反応に基づく第2光導波路層13を伝播する光の極微な変化もそれらの層2,13界面での検出できるため、前記検体中の生体分子が極微量であっても、その量を高精度で測定することができる。

An optical waveguide-type biochemical sensor, wherein a mesh-like conductive thin film to which an electric field is applied is disposed to face the functional film at a desired interval.
