Abstract
【解決手段】 前処理として、検体としてのヒトの血漿をイソプロピルアルコールと混合し、コエンザイムQ−10およびその2電子還元体をイソプロピルアルコールに抽出する。抽出液は分析するまでの間4℃の温度で保管する。抽出液を分析試料として、送液機構12、切り換え機構14、濃縮カラム16、分離カラム18、還元カラム20、紫外吸収検出器22および電気化学検出器24を備える分析システム10で分析する。

Claims (7)
- 検体に含まれるコエンザイムQ−10およびその2電子還元体を定量する、コエンザイムQ−10およびその2電子還元体の分析方法において、
前処理として該コエンザイムQ−10およびその2電子還元体の少なくともいずれかひとつを含む該検体を水溶性有機溶媒で抽出し、抽出液を分析試料として分析することを特徴とするコエンザイムQ−10およびその2電子還元体の分析方法。 - 前記水溶性有機溶媒がイソプロピルアルコールであることを特徴とする請求項1記載のコエンザイムQ−10およびその2電子還元体の分析方法。
- 前記抽出液を分析するまでの間、該抽出液を該抽出液の融点乃至室温の範囲内の温度で保管しておくことを特徴とする請求項1または2記載のコエンザイムQ−10およびその2電子還元体の分析方法。
- 前記分析試料をカラムスイッチング法により濃縮する予備処理を行うことを特徴とする請求項1乃至3のいずれか1項に記載のコエンザイムQ−10およびその2電子還元体の分析方法。
- 前記コエンザイムQ−10およびその2電子還元体の双方を含む検体からの抽出液を分析試料とし、該コエンザイムQ−10およびその2電子還元体をカラム分離し、さらに還元処理した後、検出器により検出することを特徴とする請求項1乃至4のいずれか1項に記載のコエンザイムQ−10およびその2電子還元体の分析方法。
- 請求項5記載のコエンザイムQ−10およびその2電子還元体の分析方法に用いる分析システムであって、
分析試料を第1の移動相とともに送液する第1系列と第2の移動相のみを送液する第2系列とからなる送液機構と、
該送液機構の2つの系列の移動相の送液経路を切り換える切り換え機構と、
該第1系列の移動相を受け入れて該分析試料を濃縮した後、該第2の移動相を受け入れる濃縮カラムと、
該濃縮カラムから送り出される液を受け入れて分離する分離カラムと、
該分離カラムから送り出された液を受け入れて還元する還元カラムと、
該還元カラムから送り出された液を検出処理する電気化学検出器とを有することを特徴とするコエンザイムQ−10およびその2電子還元体の分析システム。 - 検出器としてさらに紫外吸収検出器を有することを特徴とする請求項6記載のコエンザイムQ−10およびその2電子還元体の分析システム。
