ニキビ(尋常性ざ瘡)は、主として思春期の男女の顔面、上腕部、背部或いは上腕伸側等に発生する皮膚疾患の一つであり、皮脂分泌の増加や、グラム陽性菌であるプロピオニバクテリウム・アクネス(Propionibacterium acnes)の感染等が原因で起きることが知られている。従って、ニキビの治療には、サルファ剤や抗生物質がその予防・治療薬として利用される場合が多いが、副作用や耐性菌の出現、皮膚常在菌の過剰殺菌等の点で問題があり、これらに代わる安全性の高い抗アクネ菌製剤の開発が望まれている。

一方、不飽和脂肪酸の一つであるシス−6−ヘキサデセン酸は、ヒト皮膚最外層のバリアーを構成する皮脂の主要成分であるが、最近、シス−6−ヘキサデセン酸を含む皮脂由来の脂肪酸誘導体が、多くのグラム陽性菌及びグラム陰性菌に対して抗菌活性を有することが報告されている(特許文献1参照)。また、イソプロピルメチルフェノール及びトリクロロカルバニリドは、殺菌力、抗菌性を持ち、皮脂に増殖しやすい細菌に作用することで、従来から様々な外用殺菌剤に配合されている。

しかしながら、シス−6−ヘキサデセン酸とイソプロピルメチルフェノール又はトリクロロカルバニリドを共に配合した抗菌性製剤は全く知られていない。

【課題を解決するための手段】
本発明者らは、抗生物質に代わる安全性の高い抗菌性物質を探索したところ、シス−6−ヘキサデセン酸又はその塩とイソプロピルメチルフェノール又はトリクロロカルバニリドを組み合わせた場合にアクネ菌に対して極めて優れた抗菌力を示し、ニキビに代表される皮膚疾患の予防又は治療剤として有用であることを見出した。

【発明の実施の形態】
本発明のシス−6−ヘキサデセン酸は、ヒト皮膚最外層のバリアーを構成する皮脂の主要成分であることから、動物に長期投与しても全く無害の物質である。シス−6−ヘキサデセン酸は、常法によりその薬理学的に許容される塩とすることができるが、このような塩としては、ナトリウム塩、カリウム塩等のアルカリ金属との塩、カルシウム、マグネシウム等のアルカリ土類金属との塩、又は各種アミン塩等を含むアンモニウム塩やリジン塩、アルギニン塩等のアミノ酸塩やグアニジン塩、アルミニウム塩や亜鉛塩を挙げることができる。

シス−6−ヘキサデセン酸は、公知のメタセシス反応や微生物を用いる方法(特開平1−144982号公報、特公平4−12718号公報)、又は蔓性植物のやはずかずら(ツンベルギアアラータ)から抽出する方法により得ることができるが、特に、パルミチン酸イソプロピルからロドコッカス属微生物を用いて生産する方法(特公平4−12718号公報)によれば、シス−6−ヘキサデセン酸を工業的に生産することが可能である。

抽出処理は、通常3〜100℃程度の温度で数時間〜数週間、常法によって行うことができ、更に抽出物をゲル濾過、カラムクロマトグラフィー、精密蒸留等で精製して用いることもできる。

斯かる界面活性剤としては、ノニオン系、アニオン系の界面活性剤が好適に挙げられ、ノニオン系に関してはポリオキシエチレン硬化ヒマシ油、イソステアリルグリセリルエーテル、ポリオキシエチレンアルキルエーテル、グリセリン脂肪酸エステル、ポリエチレングリコール、脂肪酸エステル、モノステアリン酸ソルビタン、ポリオキシエチレンモノステアリン酸ソルビタン等が好ましく、モノステアリン酸ソルビタンがより好ましい。また、アニオン系に関してはポリオキシエチレンラウリルエーテルリン酸Na、N−ステアロイル−N−メチルタウリンNa、ラウリルリン酸、リン酸モノミリスチル、リン酸モノセチル、ポリオキシエチレンラウリルエーテル硫酸Na、ラウリル硫酸トリエタノールアミン、ポリオキシエチレンラウリルエーテル硫酸トリエタノールアミン等が好ましく、ポリオキシエチレンラウリルエーテル硫酸Naがより好ましい。

斯くして得られる本発明の抗アクネ菌組成物は、ニキビ、脂漏性皮膚炎等のアクネ菌感染性皮膚疾患の予防又は治療、とりわけニキビの予防又は治療を目的とした医薬又は化粧料として有用であるが、医薬として使用する場合には、錠剤、カプセル剤、軟膏、スティック状剤、水剤、エキス剤、ローション剤、乳剤、パップ剤等の皮膚外用剤とすることができ、中でも外用医薬品としての使用が好ましい。例えば軟膏剤とする場合には、油性基剤をベースとするもの、水中油型又は油中水型の乳化系基剤をベースとするもののいずれであってもよく、油性基剤としては、例えば植物油、動物油、合成油、脂肪酸及び天然又は合成のグリセライド等が挙げられる。
また、当該医薬には、他の薬効成分、例えば鎮痛消炎剤、殺菌消毒剤、収斂剤、皮膚軟化剤、ホルモン剤、抗生物質等を必要に応じて適宜配合することができる。

【実施例】
実施例1 抗アクネ菌性の評価
3% 1,3−ブチレングリコール(和光純薬、98%純度品)、0.1% モノステアリン酸ソルビタン(花王、レオドールスーパーSPS10)、50mMクエン酸緩衝液(pH4.5)からなるベース緩衝液10mLを調製した。このベース緩衝液に、0.01%のシス−6−ヘキサデセン酸と、0.01%のイソプロピルメチルフェノールを組み合わせて添加したサンプル及び比較品として、0.01%のシス−6−ヘキサデセン酸又は0.01%のイソプロピルメチルフェノール(大阪化成、イソプロピルメチルフェノール)を単独で添加したサンプルを調製した。尚、シス−6−ヘキサデセン酸は、特公平4−12718号公報記載の方法に準じて製造したものを用いた。
各サンプル10mLに対し、約1.0×108CFU/mLからなるPropionibacterium acnes(JCM6425)を0.1mL接種して、嫌気条件下、室温にて保持した。0、1、3時間後に、接種サンプルを1mLずつ10mLのLP希釈液(和光純薬)に回収して撹拌後、LP希釈液(和光純薬)を含む変法GAM寒天培地(日水製薬)に塗抹し、37℃、5日間嫌気培養し、出現するコロニー数をカウントして残存するアクネ菌数/mLを算出した。結果を図1に示す。
