The black rice extract has a large content of available components, no odor and high solubility in water, further facilitates the production of skin lotion, beverage, food and medicine with reduced costs of raw materials and products. 本発明は、有用な食品である黒米又は黒米糠から、いやな臭いがなく、水溶性に優れた黒米エキス又は黒米色素を得る方法に関する。
本発明は、化粧品、医薬品、医薬部外品、飲食品等に適用される黒米エキス又は黒米色素の製造方法に関する。

【産業上の利用分野】
ここで言う黒米は、稲Oryza sativa linne(Gramineae)の種子であり、中国原産の古代米の一種で、紫黒米、紫米、とも呼ばれる。黒米玄米の表面は黒くて、中は白く、玄米種皮の部分に紫黒色系色素(アントシアニン)を含んでいる。
古来、中国では不老長寿、滋養強壮、貧血に効くとされ、中国は漢の時代、張騫(ちょうけん)がこの米を発見して、出世したという故事から、「縁起の良い出世米」として宮廷献上米となり、以来、宮廷料理として常食されてきた。楊貴妃も美容食として黒米を愛用したと伝えられている。
「本草綱目」によれば、黒米は、精力増進、脾臓、肝臓を強め、胃腸を丈夫にし、造血作用により肺機能を強化させる。「造血米」「薬用米」と呼ばれ、現在まで「補血黒米粥」などの薬膳料理として、伝わっている。中国民間の言い伝えとして、慢性病疾患、回復期の病人、幼児、虚弱体質者にとって滋養強壮作用があるとされる。また、黒米を食べ続けると、肌がなめらかになり、髪の毛が黒くなって若返ると言われている。
黒米又は黒米の一部である黒米糠およびそのエキス又は色素を医薬品、食品、化粧品等に配合すると皮膚の老化を防ぎ、はり艶のある皮膚とし、美肌、美白効果があり、骨や結合組織の成長促進、コラーゲンの合成を高め、インフルエンザの予防・治療、糖尿病の予防・緩和および肥満防止に有効で、免疫機能を高め、肺気腫、アテローム性動脈硬化、リュウマチ性関節炎等の治療及び予防に有用である医薬品、医薬部外品、健康食品、食品、化粧品を提供することができるようになった。

【従来の技術】
従来用いられているものとして、黒米抽出法の文献を検索すると、次のような文献や記載がある。特開平7−250647に黒米の色素による水飴製造法。特開平9−238646に黒米の食品着色料及びその製造法。特開平11−196789に黒米の加工食品への着色料として使用。特開平11−332508に黒米色素の酒類、飲料、加工食品。特開2000−63692に黒米から天然色素アントシアン抽出技術。など多くの特許公開があるが、今回の新規な抽出方法で黒米を抽出し、いやな臭いがなく、水溶性に優れた黒米エキス又は黒米色素を得る方法は、見当たらない。

【発明が解決しようとする課題】
黒米又は黒米糠から抽出物を製造すると、嫌な臭いが付いて、色素が十分に抽出されないなどの問題があった。また、仮に抽出を効率よく出来た場合でも、水溶性が悪くなって、使いにくい抽出物となっていた。
今回発明された抽出法は、嫌な臭いもなく、色素成分やポリフェノール成分の損失もなく、水溶性に優れた抽出物を得ることに成功した。医薬品、医薬部外品、食品、化粧品に黒米エキスを容易に配合できるようにした。

【課題を解決するための手段】
前記課題を解決するための本発明の抽出法は、無極性溶媒又は弱極性溶媒である酢酸エチル、エーテル、ヘキサン、二酸化炭素を用いて、脱脂操作を行い、油分を中心とした物質を取り除く。次に、含水エタノール、含水メタノール、含水アセトン、含水酢酸等で抽出を行いエキス又は色素を得る。
例えば、黒米糠にヘキサンを加え、ヘキサンを取り除いた後、30%エタノールで抽出すると、臭いもなく、水に簡単に溶解するエキスを得ることが出来る。

【効果】
本発明法で黒米又は黒米糠からエキスを製造すると、嫌な臭いを取ることが簡単で、エキスの収量も良い。また、ポリフェノールやアントシアニン色素の収量も良い。得られたエキスは、有効性もよく、経済的で、効率的な、廃棄物も少ない抽出法とすることが出来た。さらに、嫌な臭いがなく、水溶性に優れた黒米エキスを得ることに成功した。

【実施例】
本発明の製造法をする場合、抽出温度は、抽出溶媒が液体状態で有ればよく、そのつど温度は任意に決められる事になる。抽出時間は、好ましくは、1時間以上必要であるが温度条件によっては、もっと短い時間でも可能である。

(1)黒米または黒米糠100gにエーテル500ml加え室温放置する、エーテルを取り除き脱脂操作とする。脱脂された黒米または黒米糠に100gに60%エタノール500mlを加え加熱抽出する。抽出後ろ過し、ろ液を得る。このろ液の溶媒を除き、黒米エキス15g得る。
(2)黒米または黒米糠100gに酢酸エチル500ml加え室温放置する、酢酸エチルを取り除き脱脂操作とする。脱脂された黒米または黒米糠100gに70%エタノール700ml加え室温抽出する。24時間後ろ過し、ろ液を得る。このろ液の溶媒を除き、黒米エキス15g得る。
(3)黒米または黒米糠100gに1,3−ブチレングリコール溶液(50%)1l加え加熱抽出する。抽出後ろ過し、ろ液を得る。このろ液を黒米エキスとする。
(4)黒米または黒米糠100gにヘキサン500ml加え室温放置する、ヘキサンを取り除き脱脂操作とする。脱脂された黒米または黒米糠に30%エタノール1000ml加え室温抽出する。24時間後ろ過し、ろ液を得る。このろ液の溶媒を除き、黒米エキス15g得る。
(5)黒米または黒米糠100gにヘキサン500ml加え室温放置する、ヘキサンを取り除き脱脂操作とする。脱脂された黒米または黒米糠に50%エタノール800ml加え室温抽出する。24時間後ろ過し、ろ液を得る。このろ液の溶媒を除き、黒米エキス15g得る。
(6)黒米または黒米糠100gにヘキサン500ml加え室温放置する、ヘキサンを取り除き脱脂操作とする。脱脂された黒米または黒米糠に10%エタノール1000ml加え室温抽出する。24時間後ろ過し、ろ液を得る。このろ液の溶媒を除き、黒米エキス15g得る。
(7)黒米または黒米糠100gにヘキサン500ml加え室温放置する、ヘキサンを取り除き脱脂操作とする。脱脂された黒米または黒米糠100gに1,3−ブチレングリコール溶液(70%)1l加え加熱抽出する。抽出後ろ過し、ろ液を得る。このろ液を黒米エキスとする。

【発明の効果】
以上詳しく示したように、黒米又は黒米糠から抽出物を得る場合、今回の抽出法を用いれば、有効成分を逃がすことなく得られ、より有用な皮膚外用剤、飲食品または薬品を作ることが可能となった。
