約100単位から約500,000単位の範囲、好ましくは約1000単位から
約100,000単位の範囲、最も好ましくは約10,000から100,00
0単位の範囲であり得る。1mlあたりの活性単位の量および曝露期間は、感染
の性質、および、担体によって修飾溶菌酵素(群)が接触できる量に依存する。
修飾溶菌酵素(群)は、液体環境で最善に作用することに注意する必要がある。
従って、修飾溶菌酵素(群)の効力は、一部、担体により捕獲される湿気の量に
関連した。敗血症の処置では、治療剤を血流に連続的に静脈内に流すべきである
。敗血症の処置における修飾溶菌酵素(群)の濃度は、感染の重度に依存する。

ジム、セフチゾキシム、セフトリアクソン、セフリアクソンマクソラクタム、セ
フロキシム、セファレキシン、セファロスポリンC、セファロスポリンCナトリ
ウム塩、セファロチン、セファロチンナトリウム塩、セファピリン、セフラジン
、セフロキシムアクセチル、セファロチン二水和物、マクソラクタム、ロラカル
ベフ、マフェート、キレート剤およびその任意の組合せであり得る。

1965))。米国特許第3,278,378号は、後にS.シムランスと再度
命名した、S.スタフィロリティカスの培養培地からリソスタフィンを産生する
発酵法を記載する。リソスタフィンを産生する他の方法は、さらに、米国特許第
3,398,056号および第3,594,284号に記載する。リソスタフィ
ンの遺伝子は、続いて、クローニングおよびシークエンスされた(Recsei
ら、Proc.Natl.Acad.Sci.USA、84:1127〜113
1(1987))。組換え粘液溶解殺菌タンパク質、例えばr−リソスタフィン
は、その標的特異性、低毒性および生物学的に活性な残基の還元の可能性から、
現在の抗生物質療法に関連した問題を回避できる可能性がある。さらに、リソス

染することを可能とする。生体を時に重複感染する細菌の1つは、黄色ブドウ球
菌である。黄色ブドウ球菌の多くの株は、ペニシリナーゼを産生し、よって、ス
タフィロコッカス、ストレプトコッカス、および他のグラム陽性細菌株は、標準
的な抗生物質により死滅しないだろう。結果として、修飾溶菌酵素(群)および
リソスタフィンの、おそらく抗生物質と組合わせた使用は、大半の迅速で効果的
な細菌感染の処置として使用できる。さらに別の好ましい実施形態において、本
発明は、ムタノリシンおよびリゾチームを含み得る。

Claims (62)
- 【請求項1】 非経口的に有効な量の治療剤を投与することを含む、細菌感
染の処置法であって、前記治療剤は、細菌に特異的なバクテリオファージに感染
した前記細菌により産生される少なくとも1つの酵素、ここでの前記少なくとも
1つの酵素は、溶菌酵素、修飾溶菌酵素、およびその組合せからなる群から選択
する;および 感染部位に前記溶菌酵素を送達するための担体を含む、前記処置法。 - 【請求項2】 前記修飾溶菌酵素は、キメラ溶菌酵素、シャッフルド溶菌酵
素、およびその組合せからなる群から選択される、請求項1に記載の方法。 - 【請求項3】 少なくとも1つのホリン酵素をさらに含む、請求項1に記載
の方法。 - 【請求項4】 前記少なくとも1つのホリン酵素はシャッフルド酵素である
