- 【特許請求の範囲】 1.SEQ ID NO:5およびそのフラグメントに少なくとも70%の同一性を有する 配列からなる群から選択されたアミノ酸配列からなり、プレニルシステインカル ボキシルメチルトランスフェラーゼ分子が単離されるときまたは膜結合するとき プレニルシステインカルボキシルメチルトランスフェラーゼ活性を有する、単離 されたプレニルシステインカルボキシルメチルトランスフェラーゼ分子。 2.SEQ ID NO:5またはそのフラグメントに少なくとも85%の同一性を有する 配列からなる、請求項1記載の単離されたプレニルシステインカルボキシルメチ ルトランスフェラーゼ分子。 3.SEQ ID NO:5またはそのフラグメントに少なくとも90%の同一性を有する 配列からなる、請求項1記載の単離されたプレニルシステインカルボキシルメチ ルトランスフェラーゼ分子。 4.SEQ ID NO:5またはそのフラグメントに少なくとも95%の同一性を有する 配列からなる、請求項1記載の単離されたプレニルシステインカルボキシルメチ ルトランスフェラーゼ分子。 5.SEQ ID NO:5またはそのフラグメントのアミノ酸配列からなる、請求項1 記載の単離されたプレニルシステインカルボキシルメチルトランスフェラーゼ分 子。 6.SEQ ID NO:5に少なくとも85%の同一性を有する配列からなる、請求項1 記載の単離されたプレニルシステインカルボキシルメチルトランスフェラーゼ分 子。 7.SEQ ID NO:5に少なくとも90%の同一性を有する配列からなる、請求項1 記載の単離されたプレニルシステインカルボキシルメチルトランスフェラーゼ分 子。 8.SEQ ID NO:5に少なくとも95%の同一性を有する配列からなる、請求項1 記載の単離されたプレニルシステインカルボキシルメチルトランスフェラーゼ分 子。 9.SEQ ID NO:5のアミノ酸配列からなる、請求項1記載の単離されたプレニ ルシステインカルボキシルメチルトランスフェラーゼ分子。 10.請求項1記載のプレニルシステインカルボキシルメチルトランスフェラ ーゼをコード化するヌクレオチド配列からなる組換えDNA分子。 11.ヌクレオチド配列がSEQ ID NO:5のアミノ酸配列をコード化する、請求 項10記載の組換えDNA分子。 12.ヌクレオチド配列がSEQ ID NO:5のヌクレオチド44ないし895からなる、 請求項11記載の組換えDNA分子。 13.請求項10の組換えDNA分子からなる発現ベクター。 14.請求項13の発現ベクターを用いて形質転換した宿主。 15.単離されるかまたは膜結合するときプレニルシステインカルボキシルメ チルトランスフェラーゼ活性を有するタンパク質をコード化するヌクレオチド配 列からなり、前記ヌクレオチド配列が50%ホルムアミドおよび5×SSCで40℃に てSEQ ID NO:4のヌクレオチド44ないし895の補体にハイブリッド形成する、組換 えDNA分子。 16.請求項15記載の組換えDNA分子からなる発現ベクター。 17.請求項16記載の組換えDNA分子からなる発現ベクター。 18.請求項1記載のプレニルシステインカルボキシルメチルトランスフェラ ーゼに特異的な抗体の抗原結合部位を含む分子。 19.前記分子がポリクローナル抗体である、請求項18記載の分子。 20.前記分子がモノクローナル抗体である、請求項18記載の分子。 21.プレニルシステインカルボキシルメチルトランスフェラーゼ分子が存在 する膜調製品を準備し; 単離したプレニルシステインカルボキシルメチルトランスフェラーゼの基質と 、プレニルシステインカルボキシルメチルトランスフェラーゼのポテンシャル阻 害剤を用いてまたは用いずに、膜調製品をインキュベーションし; ポテンシャル阻害剤の不在でのプレニルシステインカルボキシルメチルトラン スフェラーゼのレベルに関して、ポテンシャル阻害剤の存在でのプレニルシステ インカルボキシルメチルトランスフェラーゼ活性のレベルを決定し; そして 前記ポテンシャル阻害剤の存在でのプレニルシステインカルボキシルメチル トランスフェラーゼ活性のレベルが前記ポテンシャル阻害剤の不在でのそれより も実質的に小さいことを、前記決定の工程が決定するための、任意の前記ポテン シャル阻害剤を阻害剤として単離する 各工程からなる、プレニルシステインカルボキシルメチルトランスフェラーゼ の阻害剤を単離するスクリーニング方法。 22.プレニルシステインカルボキシルメチルトランスフェラーゼが膜調製品 の供給源本来のものではない、請求項21記載の方法。 23.プレニルシステインカルボキシルメチルトランスフェラーゼが、SEQ ID NO:5またはそのフラグメントに少なくとも70%の同一性を有する配列からなる 群から選択されたアミノ酸配列からなる単離されたプレニルシステインカルボキ シルメチルトランスフェラーゼ分子であり、前記分子が単離されるときまたは膜 結合するときプレニルシステインカルボキシルメチルトランスフェラーゼ活性を 有する、請求項21記載の方法。 24.交配受容能のためのプレニルシステインカルボキシルメチルトランスフ ェラーゼを異所的に発現するように、プレニルシステインカルボキシルメチルト ランスフェラーゼをコード化する組換えDNA分子を用いて、官能プレニルシステ インカルボキシルメチルトランスフェラーゼを欠く無菌株から安定に形質転換し た交配受容能の酵母Mata株を準備し; プレニルシステインカルボキシルメチルトランスフェラーゼのポテンシャル阻 害剤の存在または不在で酵母Mata株をインキュベーションし; ポテンシャル阻害剤の不在でインキュベーションした酵母株に関して、交配受 容能のためのポテンシャル阻害剤の存在でインキュベーションした酵母株をアッ セイし;そして アッセイ工程が、ポテンシャル阻害剤の存在での酵母株の交配受容能がポテン シャル阻害剤の不在でのそれよりも実質的に小さいことを決定するための、任意 のポテンシャル阻害剤を阻害剤として単離する 各工程からなる、プレニルシステインカルボキシルメチルトランスフェラーゼ 分子の阻害剤を単離するスクリーニング方法。 25.プレニルシステインカルボキシルメチルトランスフェラーゼが膜調製 品の供給源本来のものではない、請求項24記載の方法。 26.プレニルシステインカルボキシルメチルトランスフェラーゼが、SEQ ID NO:5およびそのフラグメントに少なくとも70%の同一性を有する配列からなる 群から選択されたアミノ酸配列からなる単離されたプレニルシステインカルボキ シルメチルトランスフェラーゼ分子であり、前記分子が単離されるときまたは膜 結合するときプレニルシステインカルボキシルメチルトランスフェラーゼ活性を 有する、請求項24記載の方法。
