Description
Claims (1)
- 【特許請求の範囲】 1.形質転換生物における遺伝子調節の特異性を改良する方法であって、生物に おける内在性ターゲット遺伝子を同定する工程、ターゲット遺伝子の複数の発現 部位の位置を決定する工程、ターゲット遺伝子の発現部位の一つにおける発現を 含む、生物において少なくとも2つの発現部位で遺伝子発現を生じさせるプロモ ーターおよびターゲット遺伝子の発現を調節する核酸配列またはその産物である 作用物質を含むキメラ遺伝子を作製する工程、キメラ遺伝子を遺伝子形質転換に よって生物の細胞に安定に組み込む工程、ならびに形質転換細胞から生物を再生 させる工程を含み、形質転換生物はキメラ遺伝子を含み、生物におけるその遺伝 子の発現により、形質転換生物またはその子孫における少なくとも一つの特異的 発現部位でターゲット遺伝子またはその産物が調節されるものである、前記方法 。 2.前記核酸配列がコードまたは非コード配列である、請求項1に記載の方法。 3.前記ターゲット遺伝子の発現がアップレギュレートされる、請求項1または 2に記載の方法。 4.前記ターゲット遺伝子の発現がダウンレギュレートされる、請求項1または 2に記載の方法。 5.前記生物が植物である、請求項1、2、3または4に記載の方法。 6.前記核酸配列がアンチセンス方向のものである、請求項1〜4のいずれか1 項に記載の方法。 7.前記核酸がセンス方向のものである、請求項1、2または4に記載の方法。 8.前記核酸がセンス方向のものである、請求項1、2または3に記載の方法。 9.前記ターゲット遺伝子のアップレギュレーションが、前記ターゲット遺伝子 のプロモーターのアクチベーターの導入から生じるものである、請求項3に記載 の方法。 10.前記キメラ遺伝子の前記作用物質が複数の核酸配列を含み、その配列のそ れぞれは、発現された場合に特定の機能を果たすものである、請求項1〜9のい ずれか1項に記載の方法。 11.前記キメラ遺伝子のプロモーターが、前記内在性遺伝子の複数のオーバー ラップ発現部位で発現される、請求項1〜10のいずれか1項に記載の方法。 12.前記プロモーターがKNT1プロモーターまたはRB7プロモーターであ る、請求項1〜11のいずれか1項に記載の方法。 13.前記プロモーターがそれに作用するその他の物質によって発現されるもの である、請求項1〜11のいずれか1項に記載の方法。 14.前記核酸配列が、RB7またはKNT1プロモーターまたはそのコード配 列のアンチセンス配列の全部または一部である、請求項1〜12のいずれか1項 に記載の方法。 15.前記核酸配列が、1以上のリボザイム、メッセンジャーRNAを分解する ための標的RNアーゼ、または特異的RNAの安定化もしくは不安定化物質であ る、請求項1〜11のいずれか1項に記載の方法。 16.前記核酸が転写のアクチベーターを含む、請求項1〜11および13のい ずれか1項に記載の方法。 17.転写の前記アクチベーターがタンパク質をコードする熱ショック因子であ る、請求項16に記載の方法。 18.前記作用物質が前記遺伝子上で作用する機構が、上記で1〜11と番号付 けされた前記機構の中の1以上である、請求項1〜17のいずれか1項に記載の 方法。 19.受託番号NCIMB 40871の寄託された構築物。 20.プラスミドpATC 37010またはそのプラスミドに含まれるようにプロモータ ー−遺伝子断片−ターミネーターカセットを含む構築物。 21.請求項1〜18のいずれか1項に記載の方法によって形質転換された植物 およびその子孫。
