【発明の詳細な説明】
生物活性ポリマーナノ粒子-核酸接合体の製造方法
技術分野
本発明は、生物活性ポリマーナノ粒子-核酸接合体の製造方法ならびに遺伝子
転送および/または遺伝子発現制御のための当該接合体の使用に関する。
背景技術
合成オリゴデオキシヌクレオチド及びその誘導体等の核酸および核酸誘導体を
遺伝子発現制御(アンチセンス法(anti-sense strategy))に使用する場合、
またはDNA断片およびプラスミドを遺伝子転送に使用する場合、細胞膜を介し
て効果的な輸送が要求される。さらに、核酸は、酵素分解から保護されると共に
、所望の効果を得るために、細胞内の目標位置(細胞核、細胞質内のmRNA)に充分
な濃度で到達する必要がある。
生物学的および/又は治療学的有効物質の輸送には、過去長期間に亘ってコロ
イドキャリアー方式が使用されている。該キャリアー方式の要件を以下に示す。
− 細胞構造サイズ以下の小粒径(1μm未満の直径)
− 高負荷能(highloading capacity)
− 低毒性
− 表面改質性
− サイズ及び/又は表面特性の変更によるターゲット位置の制御
− 吸着物質の解離の調整
− キャリアー物質、分解生成物または助剤による副作用のないこと
ナノメーター域の合成ポリマー粒子を核酸吸着に使用することは、例えば、フ
ランス公開特許2,649,321号に記載されている。このフランス公開特許に
よれば、ポリ(アルキルシアノアクリレート)は、低分子量疎水性カチオンの吸
着によって正の表面電荷を示し、イオン作用によって負帯電オリゴデオキシリボ
ヌクレオチドを付着させることが可能となる。この系では、低分子量カチオンの
吸
着結合が非常に不安定であり、且つ、ポリマー状原物質の分解生成物と同様に当
該疎水性カチオンが毒性を示すという欠点を有する。
ヨーロッパ公開特許430,517号は、乳化重合法による膜、フィルム又は
粒子状のバイオモザイク(biomosaic)ポリマーの製造方法を記載している。こ
の製造方法おいては、本質的に、生物活性物質(例えば、核酸等)と同様に表面
活性物質(イオン性または非イオン性テンサイド(ionic or non-ionic tenside
))の存在下に重合を行い、前記生物活性物質を不可逆的にポリマー状原物質中
に重合し及び/又はその表面に結合させる。
更に、国際特許公開公報wo96/24377には、カチオン性ポリマーナノ粒子とペ
プチド及び/又は変性または非変性核酸からなる薬剤が記載されている。これに
よれば、アクリル酸、アクリル酸エステル又はメタアクリル酸エステル系の粒子
状混合ポリマーが、ポリマー状原物質として用いられる。活性成分の結合に必要
なカチオン性官能基の導入には、プロトン化および/またはアルキル化可能なア
ミノ官能基を有する変性コモノマーが使用される。これら電荷担持モノマー単位
は、ラジカル又はイオン重合反応において、適当な非変性モノマーといかなる割
合においても反応する。これらモノマー単位の製造が複雑であること以外に、混
合ポリマーの製造時に、これらモノマーの溶解性および反応性の相違により度々
不均質な生成物が生成される欠点を有する。。
従って、本発明の目的は、従来技術における上記欠点を克服すると共に、生物
学的媒体中で充分な安定性を示し、且つ、細胞膜を介する輸送を高効率で行い得
る均質なポリマーナノ粒子-核酸接合体を簡易で、廉価且つ再現可能な方法によ
り製造する方法を提供することにある。
発明の開示
本発明によれば、前記問題点は、カチオン性ラジカル開始剤の存在下に、低水
溶性ビニルモノマーを非乳化剤型乳化重合法により重合し、次いで得られたポリ
マー懸濁液と核酸とを反応させることによって解決される。
発明の実施形態
この方法で製造された物質は、高核酸負荷性および充分な酵素分解安定性を示
す。更に、これら接合体の特性は、例えば、ポリマー状原物質、粒子径、表面改
質度、核酸変性度等の多くのパラメーターを変更することによって、種々変化さ
せることが可能である。本発明に係る接合体は、国際特許公開公報wo96/24377
に記載の接合体と以下の点で相違する。即ち、本発明に係る接合体は、カチオン
性基が、実質的にポリマーナノ粒子を構成する各ポリマー鎖の末端鎖部分に存在
し、ポリマー鎖の中心部に存在しない。
本発明の方法は、通常、2つの工程からなる。第1の工程は、水性分散媒中で
ビニルモノマーを非乳化剤型乳化重合する公知の方法でポリマー状原物質を製造
する。ここで使用するビニルモノマーは、好ましくは20g/l以下の低水溶解
度を有する。かかるビニルモノマーとしては、スチレン、アクリル酸またはメタ
クリル酸誘導体が例示される。好ましいアクリル(メタクリル)酸誘導体として
は、アルキル(メタ)アクリレート(但し、アルキルの炭素数は1〜8である)
、および、例えば、N−ブチルメチルアクリルアミド、N−イソブチルメチルア
クリルアミド又はN−オクチルメチルアクリルアミド等のN−アルキル又はジア
ルキルアクリルアミドが挙げられる。
本発明によれば、上記乳化重合は、好ましくは20〜80℃の温度範囲におい
て乳化剤の添加なしに行われ、且つ、ポリマー状原物質の表面電荷は、イオン性
ラジカル開始剤のみによって引き起こされる。ここで、上記開始剤としては、例
えば、2,2’−アゾビス(2−アミジノプロパン)ジヒドロクロリド(AIB
A)又は2,2’−アゾビス(2−(2−イミダゾリン−2−イル)プロパン)
ジヒドロクロリド(AIBI)等の塩基性末端基含有カチオン性開始剤が例示さ
れる。
乳化剤または安定剤等の添加剤を反応系に添加する必要が無いため、本発明の方
法においては、後の精製工程でこれら添加剤の複雑な分離工程を行う必要がない
。
また、反応を水性分散媒中で行うため、ポリマー状原化合物の製造プロセスは
、特にコスト的に有利である。重合工程は、バッチ法で行うのが好ましい。バッ
チ法は、温度制御および選択された撹拌法のバラツキによる最終生成物の特性へ
の影響が少ないため、反応のスケールアップを行っても問題がない。
バッチ反応のモノマー濃度および開始剤濃度にもよるが、ポリマー粒子の粒径
は、10〜1,000nmの範囲が好ましく、乳化重合後の表面電荷は、0.1
〜10C/ポリマー1gの範囲が好ましい。
また、得られたポリマー懸濁液は、高い貯蔵安定性を示し、室温で数カ月以上
全く凝集すること無く保存することが出来る。更に、かかる本発明のポリマー状
キャリアー物質は、生物学的媒体中で高い安定性を示すと共に、所望の応用分野
で低毒性を示す。
核酸と同様にポリマー懸濁液は、例えば、反応前に、遠心分離または透析濾過
によって精製することが好ましい。透析濾過においては、通常の透析法および透
析膜を用いることが出来る。
更に、本発明によれば、安定剤を重合後のポリマー懸濁液に添加して更に安定
性を高めることが出来る。安定剤の添加量は、懸濁液に対し0.01〜5重量%
である。安定剤として、疎水性部位および親水性部位を有する生物不活性非イオ
ン性ブロックコポリマーを好適に用いることが出来る。かかる安定剤としては、
ポロキサマー類(poloxamers)又はポロキサミン類(poloxamines)が例示され
る。
本発明の方法の第2工程の上記ポリマー懸濁液と核酸の反応は、好ましくは、
10〜30℃の温度で且つ11未満のpH値で行われる。上記反応では、結合す
る核酸は、非プロトン化状態で存在する。ここで、核酸としては、例えば、デオ
キシリボヌクレオチド、リボヌクレオチド、7〜40個のヌクレオチド単位を有
する化学変性デオキシリボヌクレオチド及びリボヌクレオチドが挙げられる。ま
た、プラスミドも、核酸として問題なく使用できる。
好ましい実施態様によれば、核酸部位に起因して過剰の負電荷を有する、上記
反応により得られた本発明のポリマーナノ粒子-核酸接合体は、更に、ペプチド
、7を超える等電点を有するプロテイン又はポリエチレンイミンで、サンドイッ
チ型複合体に変性することが出来る。核酸および/またはペプチド又はプロテイ
ンを有するキャリアー物質の高負荷能(highloadingcapacity)は、安定剤の添
加の影響を受けない。核酸および/またはペプチド又はプロテインの有効結合量
は、ポ
リマーナノ粒子-核酸接合体の遠心分離および上澄み中に存在する過剰の未結合
物質量を測定することによって、容易に求めることが出来る。
本発明に係るポリマーナノ粒子-核酸接合体は、優れた生物活性を示し、その
結果、遺伝子転送および遺伝子発現制御に好適に適用できる。
アンチセンス-オリゴヌクレオチドによって多重薬剤耐性(mdr-)遺伝子が阻害
されているラット肝細胞を試料として用い、本発明に係るポリマーナノ粒子-核
酸接合体の効能をテストした。接合体中の抗多重薬剤耐性ホスホルチオエート-
オリゴヌクレオチド濃度は、遊離オリゴヌクレオチドを用いた場合に比較して、
同一効果を得るために10倍(10分の1に)減少させることが出来る。接合体
形態が酵素分解から保護されるため、本発明の接合体において効果を発揮するヌ
クレアーゼ−不安定ホスホルジエステル-オリゴヌクレオチドを使用した場合、
その差はより明確になる。しかしながら、より高い特異性および生物適合性のた
め、未変性天然DNA断片は、アンチセンス法(antisense strategy)の修正を
もたらす。
プラスミドを用いた本発明のポリマーナノ粒子-核酸接合体は、また、遺伝子
転送に優れた適用性を示す。高負荷能および遺伝子転移の有用性を調べるため、
検出容易な発現コントロールとしてβ-ガラクトシダーゼ遺伝子を含有するプラ
スミドを使用する。高負荷能(DNA1μm/ポリマー1mgにおいて)は、短
オリゴヌクレオチド断片よりも約4倍増加した。
本発明に係るポリマーナノ粒子-核酸接合体は、負帯電核酸がその帯電末端基
により粒子表面に静電的に直接結合するため、接合体の製造に荷電キャリアーと
してイオン性コモノマー及び疎水性カチオンのいずれも使用する必要がないとい
う長所を有する。生物学的分解性キャリアー物質または吸着助剤を用いる場合に
頻繁に起こる生物系、特に細胞培養試験における副作用は、本発明の方法では、
最小限に抑制される。この理由は、結合した核酸および/またはペプチドを除い
て、いかなる生物学的有効物質も遊離されないためである。ラット肝細胞に関す
る生物適性試験で示されるように、ポリマー1g/インキュベイション媒体(in
cubation medium)1リッターの濃度でのポリスチレンの細胞毒性の定量におい
て、LDHの遊離量は、未処理の対照基準(コントロール)との比較において、

0時間後でも僅か5%高いだけであった。
このように、本発明に係るポリマーナノ粒子-核酸接合体およびこれらの塩基
性ペプチド又はプロテインとの複合体は、遺伝子発現生体外制御(アンチセンス
法)の新規且つ有用な系を示している。更に、遺伝子トランスフェクション用の
ベクターとして広範囲に使用することが出来る。有効核酸成分およびその誘導体
の選択および塩基性ペプチド又はポリエチレンイミンによる表面改質によって、
ポリマーナノ粒子の原物質の粒子径、種類および表面電荷を変更することにより
、ポリマーナノ粒子-核酸系を、生物試験システムの各要件に適合させることが
出来る。
以下、実施例により、本発明を更に詳細に説明する。
実施例
実施例1
純水40ml及び保護ガス(アルゴン)下で予め精製されたスチレン2.5m
lを、KPG撹拌器、環流冷却器とアルゴン導入口および排出口装置を備えた1
00ml丸底フラスコに添加し、アルゴンガスの導入下で強く混合した。水5m
l中に溶解した2,2’−アゾビス(2−アミジノプロパン)ジヒドロクロリド
(AIBA)50mgを、上記混合物に添加し、反応フラスコを80℃に調節し
たオイル浴に浸せきし、撹拌回転数360rpmにて撹拌した。約10分後、乳
白濁反応生成物が認められた。24時間後、該反応生成物を室温まで冷却したと
ころ、固形分42g/l及び粒径400〜500nmのポリマー懸濁液が得られ
た。得られた懸濁液は、単分散の粒径分布を有する粒子を含有していた。得られ
た懸濁液を、透析膜(SpectraPor;MWCO:10000;Roth
,Karlsruhe社製)を用いて7日間純水で透析処理し、分散媒中に溶解
したポリマー及び微量残留モノマーを除去した。精製生成物は、固形分40g/
lを有し、導電性測定により求められた粒子電荷は、759mC/ポリマー1g
であった。
ホスホルアミダイト法によって合成した、5’CCC TGC TCC CC
C CTG GCT CC 3’の配列を有するホスホルチオエート-オリゴデ
オ
キシヌクレオチド20量体(DNA−1、重量平均分子量(Mw):6207g
/mol)を、調製用HPLCにより精製し、ジメトキシトリチル保護基を開裂
した後、滅菌透析膜(SpectraPorCE;MWCO:500;Roth
,Karlsruhe社製)を用いて7日間純水で透析処理した。
ポリマー懸濁液(ポリマー含量:8mg)200μlと純水に溶解したホスホ
ルチオエート-オリゴヌクレオチド(DNA−1)100ng(16nmol)
とを12〜24時間熟成処理した。この結果、ポリマー1g当たり1.7μmo
lのオリゴヌクレオチドの核酸部(核酸含量:10.6mg/ポリマー1g)を
有するポリマーナノ粒子-核酸接合体を得た。
高負荷能の定量は、以下の方法により行った。
核酸とナノ粒子とを熟成処理した後、得られた接合体を遠心分離し(1400
0g、2x40分)、上澄み中の未結合物質量を、UV測定法によって求めた。
吸着量は、核酸および/またはペプチドの出発量との差から計算により求めた。
実施例2
スチレンの使用量を1.5mlとした以外は、実施例1と同様にして、重合バ
ッチ法および生成物精製を行った。この結果、固形分23g/lのポリマー懸濁
液を得た。得られた粒子の粒径は、150〜400nmであった。透析後、固形
分は、22g/lであり、粒子電荷は、1080mC/gであった。
実施例1と同様にして、5’TTC TTG TCT GCT CTT 3’
の配列を有するホスホルジエステル-オリゴヌクレオチド15量体(DNA−2
;重量平均分子量(Mw):4491g/mol)23.8ng(5.3mol
)を含有するポリマー懸濁液100μl(ポリマー含量:2.2mg)を処理し
て、ポリマー1g当たり2.0μmolのオリゴヌクレオチドの核酸部(核酸含
量:8.9mg/ポリマー1g)を有するポリマーナノ粒子-核酸接合体を得た
。
実施例3
実施例1と同様の方法で、水10ml中に2,2’−アゾビス(2−(2−イ
ミダゾリン−2−イル)プロパン)ジヒドロタロリド(AIBI)118mgを
溶
解した溶液を、水80ml及びスチレン3mlに添加し、24時間重合させた。
得られたポリマー懸濁液は、固形分23.9g/lを有し、単分散粒径分布を示
す平均粒径150〜200nmの粒子を含有していた。21日間透析した後、固
形分は、22.7g/lであり、粒予電荷は、930mC/gであった。AIB
Iを用いて製造されたポリマー粒子は、より高い表面電荷およびより高い負荷能
(高ローディング能)を有するので、核酸との最良の接合体を形成する。従って
、本発明者の知見では、本発明の最良の実施態様と考える。
0.1%(w/v)ポロキサマー(poloxamer)338(ICIケミカル社製、
マンチェスター、英国)の水溶液で安定化されたポリマー懸濁液(安定化後の密
度:9.56g/l)のラット肝細胞における毒性を、LDH遊離量の測定によ
り求めたところ、(インキュベイション媒体1リッター中ポリマー1g濃度にお
ける)20時間後の遊離量は、未処理対照基準(コントロール)の77単位に対
し、81単位であった。
実施例1と同様にして、72.7ng(16.2mol)のDNA−2と安定
化ポリマー懸濁液200μl(ポリマー含量:4.5mg)とを処理して、ポリ
マー1g当たり3.0μmolのオリゴヌクレオチドの核酸部(核酸含量:13
.5mg/ポリマー1g)を有するポリマーナノ粒子-核酸接合体を得た。
実施例4
ホスホルチオエート-オリゴデオキシヌクレオチドDNA−3(重量平均分子
量(Mw):4716g/mol;実施例2で用いたDNA−2と同様の配列)7
6.4ng(16.2mol)と、実施例3で得られた精製し且つ安定化した懸
濁液200μlとを処理して、ポリマー1g当たり3.1μmolのオリゴヌク
レオチドの核酸部(核酸含量:14.6mg/ポリマー1g:実施例1と同様の
測定法による)を有するポリマーナノ粒子-核酸接合体を得た。
ポリマーナノ粒子-DNA−3接合体の多重薬剤耐性遺伝子の発現の抑制効果
を、ラット肝細胞におけるmRNA定量(ノーザンブロッティング)及び多重薬剤耐
性プロテイン定量(ウエスタンブロッティング)により調べた。ポリマーナノ粒
子-オリゴヌクレオチド接合体は、インキュベイション媒体1リッター当たりポ
リマー0.032、0.096、0.32及び0.96gの濃度(インキュベイシ
ョン媒体1リッター当たり0.1、0.3、1及び3μmolのDNA−3)で使
用した。インキュベイション媒体1リッター当たり1μmolのDNA−3の濃
度において、mRNA含量は、90%減少し、プロテイン含量は、50%減少した。
遊離オリゴヌクレオチドにおいては、インキュベイション媒体1リッター当たり
10μmolのDNA−3濃度においてようやく同一の抑止効果が得られた。基
準(コントロール)オリゴヌクレオチド(DNA−4:配列 5’CCT GTT GT
T TTC TCT 3’及び/又はDNA−5:配列 5’AAG AGC AGA CAA GAA
3’)は、遊離状態およびナノ粒子との接合形状での各濃度において、全く効
果を示さなかった。
実施例5
最終生成物の特性に対する重合バッチの大きさの影響をテストするため、実施
例3の反応バッチの大きさを5倍にした。粒径、粒子分布、表面電荷および核酸
高負荷能に関し精製物質の特性は、実施例3で得られた特性と顕著な差がなかっ
た。(透析後の固形分:23.8mg、粒子電荷:1020mC/ポリマー1g、核
酸含量:3.4μmolDNA−3/ポリマー1g及び16.0mgDNA−3/
ポリマー1g;実施例1と同様の測定法による)
実施例6
実施例3で得られた安定化した懸濁液50μlと、純水100μlに溶解した
ポリマー1g当たり核酸42mgの核酸部を有するpcMVβプラスミド22μ
gとを処理してポリマーナノ粒子-プラスミド接合体を得た。更に、得られた接
合体を、2.6μg(0.95nmol)ペプチド ペネトラチン(類似ドメイン
配列;重量平均分子量(Mw):2720.2g/mol;Derossi,D.
,Joliot,A.H.,Chassaing,G.及びProchiant
z,A.著「J.Biol.Chem.」1994、269(14)、1044
4−10450)と12〜24時間処理した。プラスミド及び/又はペプチドの
高負荷能測定は、実施例1と同様に行った。
実施例7
スチレンの使用量を2mlとした以外は、実施例3と同様にして、重合バッチ
法を行った。粗生成物は、10.4g/lの固形分を有し、粒子径50〜200
nmの多分散系粒子分布を示していた。10日間透析によって精製したところ、
得られた生成物は、8.2g/lの固形分を有し、粒子電荷は、2190mC/
gであった。

Claims (1)
- 【特許請求の範囲】 1.水溶液中で低水溶性のビニルモノマーを重合し、次いで得られたポリマー懸 濁液と核酸とを反応させる工程からなる生物活性ポリマーナノ粒子-核酸接合体 の製造方法において、前記ビニルモノマーの重合を、カチオン性ラジカル開始剤 の存在下に非乳化剤型乳化重合法によって行うことを特徴とする生物活性ポリマ ーナノ粒子-核酸接合体の製造方法。 2.ビニルモノマーが、20g/l未満の水溶解度を有する請求の範囲1に記載 の方法。 3.ビニルモノマーが、スチレン、アクリル酸誘導体、メタクリル酸誘導体また はこれらの混合物である請求の範囲1または2に記載の方法。 4.カチオン性ラジカル開始剤が、2,2’−アゾビス(2−アミジノプロパン )ジヒドロクロリド(AIBA)又は2,2’−アゾビス(2−(2−イミダゾ リン−2−イル)プロパン)ジヒドロクロリド(AIBI)等の塩基性末端基含 有カチオン性開始剤である請求の範囲1〜3の何れかに記載の方法。 5.乳化重合が、20〜100℃の温度範囲で行われる請求の範囲1〜4の何れ かに記載の方法。 6.乳化重合後のポリマー粒子が、10〜1,000nmの粒子径を有する請求 の範囲1〜5の何れかに記載の方法。 7.ポリマー懸濁液が、核酸との反応前に、遠心分離または透析濾過によって精 製される請求の範囲1〜6の何れかに記載の方法。 8.ポリマー懸濁液と核酸との反応が、10〜30℃の温度範囲且つ11未満の pH値で行われる請求の範囲1〜7の何れかに記載の方法。 9.核酸が、デオキシリボヌタレオチド、リボヌクレオチド又はヌクレオチド単 位7〜40を有する化学的変性デオキシリボヌクレオチド及びリボヌクレオチド である請求の範囲1〜8の何れかに記載の方法。 10.核酸が、プラスミドである請求の範囲1〜8の何れかに記載の方法。 11.懸濁液に対して0.01〜5重量%の安定剤を、重合後のポリマー懸濁液 に添加する請求の範囲1〜10の何れかに記載の方法。 12.安定剤が、疎水性部位および親水性部位を有する非イオン性ブロックコポ リマーである請求の範囲11に記載の方法。 13.非イオン性ブロックコポリマーが、ポロキサマー類またはポロキサミン類 である請求の範囲12に記載の方法。 14.生物活性ポリマーナノ粒子-核酸接合体が、ペプチド、7を超える等電点 を有するプロテイン又はポリエチレンイミンにより変性されている請求の範囲1 〜13の何れかに記載の方法。 15.前記各請求の範囲の何れか1つの方法により得られた生物活性ポリマーナ ノ粒子-核酸接合体。 16.請求の範囲15の生物活性ポリマーナノ粒子-核酸接合体の遺伝子転送の ための使用。 17.請求の範囲15の生物活性ポリマーナノ粒子-核酸接合体の遺伝子発現制 御のための使用。
