Claims (1)
- 【特許請求の範囲】 1.配列番号26に記載されたアミノ酸配列を有するヒトKVLQT1ポリペ プチドをコードする単離された核酸。 2.配列番号25に記載されたヌクレオチド配列を有するDNA、その相補体 、その対立遺伝子変異体または対応するRNAである請求項1記載の単離された 核酸。 3.長QT症候群を引き起こす配列番号26に記載されたヒトKVLQT1ポ リペプチドの突然変異形態につきコードする単離された核酸、その相補体または 対応するRNA。 4.該突然変異が、欠失突然変異、ナンセンス突然変異、挿入突然変異および ミスセンス突然変異よりなる群から選択される請求項3記載の単離された核酸。 5.該単離されたDNAが、表3に示された1以上の突然変異を含む請求項3 記載の単離された核酸。 6.試料中で検出するためにハイブリダイゼーションプローブとして使用する のに適した核酸配列が(i)配列番号1に記載されたヌクレオチド配列、その対 立遺伝子変異体およびその突然変異形態から選択されるヌクレオチド配列を有す るDNAまたは(ii)該DNAに対応するRNAである請求項1ないし5いず れか1項記載の少なくとも15の連続核酸を有する単離された核酸。 7.突然変異を含む請求項3ないし5いずれか1項記載の少なくとも15の連 続核酸を有する請求項6記載の単離された核酸。 8.請求項1ないし7いずれか1項記載の単離された核酸および宿主細胞で働 くレプリコンを含む複製クローニングベクター。 9.KVLQT1ポリペプチドまたはその修飾形態についてのコーディング配 列が、該ベクターについての宿主細胞において該コーディング配列の発現を指示 できる適当な制御配列に作動可能に連結した請求項1ないし5いずれか1項記載 の単離された核酸を含む発現ベクター。 10.請求項8または9記載のベクターで形質転換した宿主細胞。 11.(i)KVLQT1ポリペプチドの生産に適した条件下、該ポリペプチド をコードする発現ベクターを含有する請求項10記載の宿主細胞を培養し、次い で、(ii)該ポリペプチドを回収することを特徴とする、請求項1記載の配列番 号26に記載されたアミノ酸配列を有するKVLQT1ポリペプチドまたは該ポ リペプチドの修飾形態であるポリペプチドの生産方法。 12.配列番号26に記載されたアミノ酸配列を有する単離されたヒトKVL QT1ポリペプチド。 13.配列番号26に記載されたアミノ酸配列を有するKVLQT1ポリペプ チドの突然変異形態を含む単離されたヒト突然変異体KVLQT1ポリペプチド 。 14.該突然変異が表3に示されたものである請求項13記載の単離されたK VLQT1ポリペプチド。 15.請求項12ないし14いずれか1項記載の1以上のポリペプチドに特異 的に結合できる抗体。 16.対象の組織試料から単離されたKVLQT1遺伝子またはその発現産物 の配列を、野生型KVLQT1遺伝子またはその発現産物と比較することによっ て、KVLQT1遺伝子における突然変異につき、該対象をスクリーニングする ことよりなり、ここに該対象の配列における突然変異が長QT症候群の危険性の 指標であることを特徴とする長QT症候群につきヒト対象における危険性を評価 する方法。 17.該発現産物が、KVLQT1遺伝子のmRNAおよび該KVLQT1遺 伝子によってコードされたKVLQT1ポリペプチドよりなる群から選択される 請求項16記載の方法。 18.以下の手順(a)非変性ポリアクリルアミノゲル上の試料からの一本鎖 DNAの電気泳動移動度のシフトを観察し、 (b)KVLQT1遺伝子に対するKVLQT1遺伝子プローブのハイブリダイ ゼーションに適した条件下、KVLQT1遺伝子プローブを該試料から単離され たゲノムDNAにハイブリダイズさせ、 (c)対立遺伝子特異的プローブの、該試料からのゲノムDNAへのハイブリダ イゼーションを測定し、 (d)該試料からのKVLQT1遺伝子の全部または一部を増幅させて増幅配列 を得、該増幅配列を配列決定し、 (e)該試料における特異的KVLQT1突然変異体対立遺伝子の存在を核酸増 幅によって測定し、 (f)該試料からのKVLQT1遺伝子の全部または一部を分子クローニングし てクローン化配列を得、該クローン化配列を配列決定し、 (g)分子(1)該試料から単離されたKVLQT1遺伝子ゲノムDNAまたは KVLQT1 mRNA、および(2)ヒト野生型KVLQT1遺伝子DNAに 相補的な核酸プローブが相互にハイブリダイズしてデュプレックスを形成する場 合に、それらの分子間にミスマッチがあるか否かを決定し、 (h)該試料中のKVLQT1遺伝子配列を増幅させ、野生型KVLQT1遺伝 子配列を含む核酸プローブに該増幅配列をハイブリダイズさせ、 (i)該組織中のKVLQT1遺伝子配列を増幅させ、突然変異体KVLQT1 遺伝子配列を含む核酸プローブに該増幅配列をハイブリダイズさせ、 (j)欠失突然変異につきスクリーニングし、 (k)点突然変異につきスクリーニングし、 (l)挿入突然変異につきスクリーニングし、 (m)該試料中のKVLQT1遺伝子と、該KVLQT1遺伝子配列または突然 変異体KVLQT1遺伝子配列を含む1以上の核酸プローブとのインサイチュ・ ハイブリダイゼーションを測定し、 (n)免疫ブロッティングを行い、 (o)免疫組織化学を測定し、 (p)該組織から単離されたKVLQT1蛋白質と、KVLQT1突然変異体対 立遺伝子のポリペプチド発現産物に特異的に結合できる結合パートナーおよび/ または配列番号26に記載されたアミノ酸配列を有するKVLQT1ポリペプチ ドについての結合パートナーとの間の結合相互作用につき検定し、次いで、 (q)該結合パートナーの生物化学的活性の阻害につき検定する のうちの1以上を行う請求項16または17記載の方法。 19.KVLQT1ポリペプチドのアミノ酸38−39をコードするDNAも しくはRNAまたはKVLQT1ポリペプチドのアミノ酸38−39を比較する 請求項16ないし18いずれか1項記載の方法。 20.表3に示される突然変異につき、DNAまたはRNAを比較する請求項 16ないし18いずれか1項記載の方法。 21.請求項1または2記載のDNAでトランスフェクトされた細胞。 22.請求項3,4または5記載のDNAでトランスフェクトされた細胞。 23.請求項1または2記載のDNAに相補的なRNAでトランスフェクトさ れた細胞。 24.請求項3,4または5記載のDNAに相補的なRNAでトランスフェト された細胞。 25.当該動物が野生型ヒトKVLQT1を含む非ヒト・トランスジェニック 動物。 26.さらに野生型ヒトmimKを含有する請求項25記載の動物。 27.該動物が、突然変異体ヒトKVLQT1を含む非ヒト・トランスジェニ ック動物。
