Claims (1)
- 【特許請求の範囲】 1.グラム陰性細菌病原体により引き起こされる疾病を予防するために個体を 免疫するための方法であって、前記グラム陰性細菌病原体のhtrB変異体、前記グ ラム陰性細菌病原体のhtrB変異体から単離された内毒素、前記グラム陰性細菌病 原体のhtrB変異体から単離された、キャリヤータンパク質に接合される内毒素、 及び少なくとも1つの異種ワクチン抗原を発現するよう遺伝子操作された前記グ ラム陰性細菌病原体のhtrB変異体から成る群から選択された活性成分を含んで成 るワクチン調製物の予防的有効量により個体をワクチン処理することを含んで成 り、ここで前記htrB変異体が、グラム陰性細菌病原体のリピドAに比較される場 合、実質的に低められた毒性をもたらす、前記グラム陰性細菌病原体に含まれる リピドAの少なくとも1又は複数の第二アシル鎖を欠いてることを特徴とする方 法。 2.前記個人がヒトであり、そして前記ワクチン調製物が皮内、筋肉内、腹腔 内、静脈内、皮下、眼、鼻腔内、及び経口投与から成る群から選択された投与路 により導入される請求の範囲第1項記載の方法。 3.前記ワクチン調製物が、前記グラム陰性細菌病原体のhtrB変異体から実質 的に成る活性成分を含んで成る請求の範囲第2項記載の方法。 4.前記ワクチン調製物が、前記グラム陰性細菌病原体のhtrB変異体から単離 された内毒素から実質的に成る活性成分を含んで成る請求の範囲第2項記載の方 法。 5.前記ワクチン調製物が、前記グラム陰性細菌病原体のhtrB変異体から単離 された、キャリヤータンパク質に接合された内毒素か ら実質的に成る活性成分を含んで成る請求の範囲第2項記載の方法。 6.前記ワクチン調製物が、微生物病原体から少なくとも1つの異種抗原を発 現するよう遺伝子操作された、前記グラム陰性細菌病原体のhtrB変異体から実質 的に成る活性成分を含んで成る請求の範囲第2項記載の方法。 7.前記ワクチン調製物が、生理学的キャリヤー及びアジュバントをさらに含 んで成る請求の範囲第2項記載の方法。 8.前記個人が動物であり、そして前記ワクチン調製物が皮内、筋肉内、腹腔 内、静脈内、皮下、眼、鼻腔内、及び経口投与から成る群から選択された投与路 により導入される請求の範囲第1項記載の方法。 9.前記ワクチン調製物が、前記グラム陰性細菌病原体のhtrB変異体から実質 的に成る活性成分を含んで成る請求の範囲第8項記載の方法。 10.前記ワクチン調製物が、前記グラム陰性細菌病原体のhtrB変異体から単離 された内毒素から実質的に成る活性成分を含んで成る請求の範囲第8項記載の方 法。 11.前記ワクチン調製物が、前記グラム陰性細菌病原体のhtrB変異体から単離 された、キャリヤータンパク質に接合された内毒素から実質的に成る活性成分を 含んで成る請求の範囲第8項記載の方法。 12.前記ワクチン調製物が、微生物病原体から少なくとも1つの異種抗原を発 現するよう遺伝子的に構築された、前記グラム陰性細菌病原体のhtrB変異体から 実質的に成る活性成分を含んで成る請求の範囲第8項記載の方法。 13.前記グラム陰性細菌病原体のhtrB変異体が動物飼料への添加 物として経口投与される請求の範囲第8項記載の方法。 14.微生物病原体から少なくとも1つの異種抗原を発現するよう遺伝子操作さ れた前記グラム陰性細菌病原体のhtrB変異体が動物飼料への添加物として経口投 与される請求の範囲第12項記載の方法。 15.前記ワクチン調製物が、生理学的キャリヤー及びアジュバントをさらに含 んで成る請求の範囲第8項記載の方法。 16.グラム陰性細菌病原体のhtrB変異体、前記グラム陰性病原体のhtrB変異体 から単離された内毒素、前記グラム陰性細菌病原体のhtrB変異体から単離された 、キャリヤータンパク質に接合された内毒素、及び少なくとも1つの異種ワクチ ン抗原を発現するよう遺伝子操作された、前記グラム陰性細菌病原体のhtrB変異 体から成る群から選択された活性成分を含んで成るワクチン調製物であって、こ こで前記htrB変異体が、グラム陰性細菌病原体のリピドAに比較される場合、実 質的に低められた毒性をもたらす、前記グラム陰性細菌病原体に含まれるリピド Aの少なくとも1又は複数の第二アシル鎖を欠いていることを特徴とするワクチ ン調製物。 17.前記活性成分が、前記グラム陰性細菌病原体のhtrB変異体から実質的に成 る請求の範囲第16項記載のワクチン調製物。 18.前記活性成分が、前記グラム陰性細菌病原体のhtrB変異体から単離された 内毒素から実質的に成る請求の範囲第16項記載のワクチン調製物。 19.前記活性成分が、前記グラム陰性細菌病原体のhtrB変異体から単離された 、キャリヤータンパク質に接合される内毒素から実質的に成る請求の範囲第16項 記載のワクチン調製物。 20.前記活性成分が、微生物病原体から少なくとも1つの異種抗原を発現する よう遺伝子操作された、前記グラム陰性細菌病原体の htrB変異体から実質的に成る請求の範囲第16項記載のワクチン調製物。 21.生理学的キャリヤー及びアジュバントをさらに含んで成る請求の範囲第16 項記載のワクチン調製物。 22.野生型グラム陰性細菌病原体の内毒素に比較される場合、実質的に低めら れた毒性の変異体内毒素をグラム陰性細菌病原体において製造するための方法で あって、前記グラム陰性細菌病原体内のhtrB遺伝子を突然変異誘発することを含 んで成り、ここで前記突然変異誘発が前記グラム陰性細菌病原体に含まれるリピ ドAの1又は複数の第二アシル鎖を欠いている変異体内毒素により特徴づけられ る、得られるhtrB変異体の表現型を引き起こすことを特徴とする方法。 23.請求の範囲第22項記載の方法に従って製造される、実質的に低められた毒 性の変異体内毒素であって、前記実質的に低められた毒性を有する変異体内毒素 がフェノール/水抽出、及びプロテアーゼ消化から成る群から選択された方法に よりhtrB変異体から精製されており;そして実質的に低められた毒性を有する前 記精製された変異体内毒素が内毒素−特異的抗体を生成するために使用されるこ とを特徴とする変異体内毒素。 24.キャリヤータンパク質に対する接合をさらに含んで成る請求の範囲第23項 記載の変異体内毒素。 25.請求の範囲第22項記載の方法に従って製造された、実質的に低められた毒 性の変異体内毒素。 26.キャリヤータンパク質に対する接合をさらに含んで成る請求の範囲第25項 記載の変異体内毒素。 27.野生型グラム陰性細菌病原体に比較される場合、実質的に低められた毒性 を有する、野生型グラム陰性細菌病原体のhtrB変異体 を製造するための方法であって、前記グラム陰性細菌病原体内のhtrB遺伝子を突 然変異誘発することを含んで成り、ここで前記突然変異誘発が前記野生型グラム 陰性細菌病原体に含まれるリピドA上のl又は複数の第二アシル鎖を欠いている 内毒素により特徴づけられる、得られるhtrB変異体の表現型を引き起こすことを 特徴とする方法。 28.請求の範囲第27項記載の方法に従って製造された、実質的に低められた毒 性の変異体グラム陰性細菌病原体であって、ここで前記実質的に低められた毒性 を有するグラム陰性細菌病原体が内毒素−特異的抗体を生成するために使用され ることを特徴とするグラム陰性細菌病原体。 29.診断アッセイへの、及び受動的免疫化のためへの、から成る群から選択さ れた使用のための内毒素−特異的抗血清を生成するための方法であって、前記グ ラム陰性細菌病原体のhtrB変異体、前記グラム陰性細菌病原体のhtrB変異体から 単離された内毒素、及び前記グラム陰性細菌病原体のhtrB変異体から単離された 、キャリヤータンパク質に接合される内毒素から成る群から選択された活性成分 を含んで成るワクチン調製物により個人を免疫化し、そして前記免疫化された個 人から生成される抗体を集めることを含んで成り、ここで前記htrB変異体が、野 生型グラム陰性細菌病原体のリピドAに比較される場合、実質的に低められた毒 性をもたらす、前記野生型グラム陰性細菌病原体に含まれるリピドAの少なくと も1又は複数の第二アシル鎖を欠いていることを特徴とする方法。
