Abstract

Claims (1)
- 【特許請求の範囲】 1. ベクター内の内部リボソームエントリー部位(IRES)としてのおよ び/またはレトロウイルスベクターのキャプシド粒子形成を可能または改善する ための、Friend(FMLV)およびMoloney(MoMLV)ネズミ 白血病ウイルスを除くC型レトロウイルスのゲノムRNAの5’末端の全部また は一部から誘導されるヌクレオチド配列の使用。 2. 請求項1に記載のC型レトロウイルスがREV、MSV、MHV、ME V、FMOV、AMLV、MEELV、SFFV、RASV、FLV、FSV、 EFLV、SSV、GALVおよびBAEVウイルスから選択される、請求項1 に記載の使用。 3. 請求項2に記載のヌクレオチド配列が細網内皮症ウイルスのゲノムRN Aの5’末端の全部または一部から誘導される、請求項2に記載の使用。 4. 請求項3に記載のヌクレオチド配列がトリ細網内皮症ウイルス、特にA 型のゲノムRNAの5’末端の全部または一部から誘導される、請求項3に記載 の使用。 5. 請求項1〜4のいずれか1項に記載のヌクレオチド配列が、配列番号1 に記載の配列の全部または一部に実質的に相同または同一である、請求項1〜4 のいずれか1項に記載の使用。 6. 請求項5に記載のヌクレオチド配列が、配列番号2の (i)ヌクレオチド番号1で開始しヌクレオチド番号578で終了する、 (ii)ヌクレオチド番号265で開始しヌクレオチド番号578で終了する、 または (iii)ヌクレオチド番号452で開始しヌクレオチド番号578で終了する 配列に実質的に相同または同一である、請求項5に記載の使用。 7. 請求項6に記載のヌクレオチド配列が、配列番号2に記載の (i)ヌクレオチド番号1で開始しヌクレオチド番号578で終了する、 (ii)ヌクレオチド番号265で開始しヌクレオチド番号578で終了する、 または (iii)ヌクレオチド番号452で開始しヌクレオチド番号578で終了する 配列に同一である、請求項6に記載の使用。 8. 請求項1〜7のいずれか1項に従って使用されるヌクレオチド配列を含 む1つ以上の目的の遺伝子の発現用ベクター。 9. ポックスウイルス、アデノウイルス、バキュロウイルス、ヘルペスウイ ルス、アデノ随伴ウイルスおよびレトロウイルス群から選択されるウイルスから 誘導されるプラスミドベクターまたはウイルスベクターであることを特徴とする 、請求項8に記載のベクター。 10. レトロウイルスから誘導され、かつ機能的様式において関連する少な くとも以下のエレメント:レトロウイルス5’LTRおよびレトロウイルス3’ LTR、1つ以上の目的の遺伝子、ならびにウイルス粒子内へのベクターのキャ プシド粒子形成を可能または改善するための請求項1〜7のいずれか1項に記載 のヌクレオチド配列および/または前記ヌクレオチド配列の下流に位置する目的 の遺伝子の発現を可能または促進するためのIRES部位としての前記ヌクレオ チド、を含む請求項8または9に記載のベクター。 11. 前記ヌクレオチド配列がIRES部位として存在し、さらに、前記ヌ クレオチド配列に相同であるキャプシド粒子形成領域を含む、請求項10に記載 のレトロウイルスベクター。 12. 少なくとも (a)レトロウイルス5’LRT、 (b)キャプシド粒子形成領域、 (c)場合によっては、前記レトロウイルス5’LTRの起源とは異なる起源の 内部プロモーター領域が後に続く第1の目的の遺伝子、 (d)第2の目的の遺伝子 (e)IRES部位 (f)第3の目的の遺伝子、および (g)レトロウイルス3’LTR、 を含む請求項10または11に記載のレトロウイルスベクターであって、 キャプシド粒子形成領域およびIRES部位のうちの少なくとも1つが請求項1 〜7のいずれか1項に従って使用されるヌクレオチド配列からなるレトロウイル スベクター。 13. 内部プロモーター領域、第2の目的の遺伝子、IRES部位および第 3の目的の遺伝子が、レトロウイルス5’および3’LTRに対して逆方向にあ る、請求項12に記載のレトロウイルスベクター。 14. 前記キャプシド粒子形成領域がネズミレトロウイルス、特にMoML V、またはVL30型レトロトランスポゾンから誘導され、前記IRES部位が 請求項6に記載のヌクレオチド配列を含む、請求項12または13に記載のレト ロウイルスベクター。 15. 前記キャプシド粒子形成領域がMoMLVから誘導され、前記IRE S部位がヌクレオチド番号265で開始しヌクレオチド番号578で終了するか 、またはヌクレオチド番号452で開始しヌクレオチド番号578で終了する配 列番号2に記載の配列に同一なヌクレオチド配列を含む、請求項14に記載のレ トロウイルスベクター。 16. REVウイルス、特にSNV、任意の起源のレトロウイルス3’LT R、1つ以上の目的の遺伝子、およびキャプシド粒子形成領域としてヌクレオチ ド番号1で開始しヌクレオチド番号578で終了するか、またはヌクレオチド番 号265で開始しヌクレオチド番号578で終了する配列番号2に記載の配列に 実質的に相同または同一なヌクレオチド配列から誘導されるレトロウイルス5’ LTRを含む、請求項10に記載のレトロウイルスベクター。 17. 第VIII因子、第IX因子、CFTRタンパク質、ジストロフィン 、インスリン、α−、β−またはγ−インターフェロン、インターロイキン(I L)、特にIL−2から選択される発現産物をコードする目的の遺伝子および選 択マーカーを含む、請求項8〜16のいずれか1項に記載のベクター。 18. 請求項8〜17のいずれか1項に記載のウイルスベクターから作製さ れるウイルス粒子。 19. 請求項8〜17のいずれか1項に記載のベクターを含むか、または請 求項18に記載のウイルス粒子を感染させた細胞。 20. 遺伝子治療によって治療可能な疾患の治療および/または予防用医薬 組成物を製造するための請求項8〜17のいずれか1項に記載のベクター、請求 項18に記載のウイルス粒子あるいは請求項19に記載の細胞の使用。 21. 組換え経路による1つ以上の目的のポリペプチドの製造のため、また はトランスジェニック動物の保護のための請求項8〜17のいずれか1項に記載 のベクター、請求項18に記載のウイルス粒子あるいは請求項19に記載の細胞 の使用。 22. 治療用または予防用薬剤として、請求項8〜17のいずれか1項に記 載のベクター、請求項18に記載のウイルス粒子、請求項19に記載の細胞、ま たは請求項21に記載の使用に従って得られる目的のポリペプチドを、薬学的に 受容可能なビヒクルと組み合わせて含む医薬組成物。 23. 104〜1014pfu、好ましくは106〜1011pfuの請求 項18に記載のウイルス粒子を含むことを特徴とする、請求項22に記載の医薬 組成物。 24. 多分化能性細胞、特に中枢神経系の多分化能性細胞のトランスフェク ションまたは感染のための請求項8〜17のいずれか1項に記載のベクター、請 求項18に記載のウイルス粒子、あるいは請求項22または23に記載の医薬組 成物の使用。
