Abstract
小胞体を光照射により還元して酸素配位能を回復させる
こと。 【解決手段】ヘモグロビン小胞体の内水相にアミノ酸、
糖類、アルコール類、フラビン類を適量含有させ、ヘモ
グロビンが酸化してメトヘモグロビン含量が増大した時
に光照射することでこれを還元して酸素配位能を回復さ
せて機能を持続させる方法。
Description
モグロビン小胞体分散液において、ヘモグロビンの酸化
により失った酸素結合能を復元させ、酸素輸液の酸素運
搬機能を長時間持続させるための方法に関する。
散液は、医学および薬学において広く応用が可能であ
り、特に、種々の添加物を加えて臨床治療における血液
代替物として利用される。
テムには、次のような問題点が指摘されている:1)感
染(肝炎、エイズウイルスなど)の可能性があること;
2)赤血球の保存期限が3週間であること;3)高齢化
社会の到来で、輸血患者のうち高齢者の割合が高くなる
一方、健康献血者総数が低下し続けていること;4)保
存中に汚染の危険があること;5)信仰上の理由で輸血
を拒否する患者に対して適用できないこと;6)災害時
の緊急需要に対応できないこと。
きる代替物に対する要求が大きく、その一つとして、電
解質輸液、膠質輸液等の輸液製剤が従来広く使用されて
きた。しかし、これらの輸液製剤は血液の最も重要な機
能、即ち酸素を運搬する赤血球機能の代替機能を有して
ないため、この酸素運搬機能をも代替する物質(酸素輸
液)の開発が課題になっている。
(ヒトヘモグロビン、ウシヘモグロビン、および組換え
ヘモグロビン)を用いた酸素輸液の開発も進められてお
り、分子内架橋ヘモグロビン、水溶性高分子結合ヘモグ
ロビン、分子間架橋重合ヘモグロビンなどの臨床試験が
欧米で進行しているが、非細胞型構造に起因する各種の
副作用が指摘されるにつれ、ヘモグロビンをカプセルに
封入した所謂細胞型構造の重要性が明確になってきた。

脂質/コレステロール/脂肪酸からなるリポソームを利
用したヘモグロビン小胞体の検討を開始して以来、出願
人らのグループを含む幾つかのグループで、ヘモグロビ
ン小胞体の研究が精力的に展開されてきた。ヘモグロビ
ン小胞体は、1)ヘモグロビンを修飾せずにそのまま使
えること;2)粘度、膠質浸透圧および酸素親和度を任
意の値に調節できること;3)血中滞留時間が延長され
ること;4)各種添加剤を小胞体内水相に高濃度で封入
できる等の利点を有する。これらのうち、特に4)の利
点は本発明において重要である。発明者等は、これまで
にヘモグロビン小胞体の効率的な調製法を独自に確立
し、諸物性値が血液に極めて近いヘモグロビン小胞体輸
液を得ており、これについては動物投与試験でも優れた
酸素運搬能を確認している(Tsuchida ed. Blood Subst
itutes Present and FuturePerspectives, Elsevier, A
msterdam, 1998)。

になったメトヘモグロビンは酸素を結合できない。ま
た、酸素を結合したヘモグロビンは徐々にスーパーオキ
シドアニオンを遊離してメトヘモグロビンとなる。更
に、このスーパーオキシドアニオンが酸化剤として作用
し、メトヘモグロビンの生成を促進する。赤血球内には
メトヘモグロビン還元系および活性酸素消去系が存在
し、メトヘモグロビン含量が増大しない機構が作用する
が、精製ヘモグロビンを用いるヘモグロビン小胞体で
は、精製工程でこれらの酵素系をすべて除去しているの
で、保存中および投与後にヘモグロビンの酸化が生じて
酸素運搬能力が低下する。この酸化反応を抑制する方法
として、ヘモグロビン精製工程で酵素活性を喪失しない
ような穏和な条件で精製を行う方法(緒方ら、人工血液
2、62-66、1994)、グルタチオン、ホモシステインお
よび/またはアスコルビン酸などの還元剤、並びにカタ
ラーゼおよび/またはスーパーオキシドディスムターゼ
等のような活性酸素を消去する酵素を添加する方法(Sa
kai et al., Bull. Chem. Soc.Jpn., 1994)、および電
子伝達物質としてメチレンブルーを小胞体膜に導入し、
小胞体の外水相に添加したNADHからの電子移動により小
胞体に内包されたメトヘモグロビンを還元する方法(Ta
keoka et al., Bull. Chem. Soc. Jpn, 70, 1171-1178,
1997)等が試みられている。
ムCが光照射により還元される現象が、酸素輸液とは関
係なく、VorkinkおよびCusanovichによって最初に報告
された(Photochem. Photobiol. 19, 205-215, 197
4)。これ以外にも、ミオグロビン、チトクロムオキシ
ダーゼ等でも同様に、光照射により還元反応が進行する
現象が見出されて以来、多くの生化学者によってヘムタ
ンパク質の光還元が研究されてきた(Kitagawa & Naga
i, Nature, 281, 503-504, 1979;Kitagawa et al.,J.
Sm. Chem. Soc. 106, 1860-1862, 1984;Morikis et a
l., J. Biol. Chem.265, 12143-22145, 1990;Sage et
al., J. Chem. Phys. 90, 3015-3032, 1989;Gu et a
l., J. Am. Chem. Soc. 115, 4993-5004, 1993;Pierre
et al., Eur.J. Biochem. 124, 533-537, 1982;Bazin
et al., Eur. J. Biochem. 124, 539-544, 1982)。
ンにメトヘモグロビン溶液に添加して450nm付近の可視
光を照射すると還元型フラビンが生成し、これがメトヘ
モグロビンを還元する現象も良く知られている(Yubisu
i et al., J. Biol. Chem. 255, 11694-11697, 1980;E
verse, Methods Enzymol. 231, 524-536, 1994)。
ヘモグロビン小胞体の従来の還元法には以下に示すよう
な問題がある。
本的問題として、ヘモグロビンの精製工程においてウイ
ルスの不活性化が必要とされるため、アルブミン製剤と
同様に、ヘモグロビンについても60℃で10時間以上の加
熱が望まれる。その際に、赤血球に本来存在するメトヘ
モグロビン還元酵素系も変成して失活する。これら酵素
系の活性を残すために低張溶血法による穏和な精製に止
めれば、得られるヘモグロビン小胞体の酸化は抑制でき
るが、ウイルスの不活性化を達成できない。また、酵素
系は化学的に不安定であるために、保存中に活性が低下
する。
やホモシステインのような比較的穏和な還元剤をヘモグ
ロビン小胞体に内包させれば、三価鉄に酸化されたヘム
鉄が二価鉄へと還元され、総体的に酸化反応は抑制され
る。しかし、これらの還元剤は、メトヘモグロビンが存
在しない場合でも酸素により僅かながら酸化されて次第
に失活する。従って、メトヘモグロビン含量が増大した
ときにだけ還元活性を示し、これをヘモグロビンに還元
する系が望まれている。
ロビン溶液に光照射して還元反応を開始させる現象につ
いて報告があるが、実際には、この現象は均一溶液系で
は効率が極めて低いという問題がある。
で、酸化により可逆的な酸素結合能力を喪失したヘモグ
ロビン小胞体を効率的に復元するための方法および装置
を提供することを目的とする。
酸素輸液の系統的研究を重ね、メトヘモグロビンを生じ
たヘモグロビン小胞体の酸素結合能を復元する方法を開
発すべく鋭意努力を続けた結果、上記の問題を解決でき
る本発明に想到したものである。
ヘモグロビン小胞体分散液の低下した酸素結合能を回復
させる方法であって:リン脂質小胞体内にヘモグロビン
水溶液を内包し且つ前記小胞体の内水相に電子供与体を
含有するヘモグロビン小胞体の分散液に対して、前記ヘ
モグロビンが酸化してメトヘモグロビンとなり酸素結合
能が低下したときに光を照射することにより、メトヘモ
グロビンをヘモグロビンに還元し、その酸素結合能を回
復させることを特徴とする方法である。
方法を実施するための装置であって:請求項1で定義し
たヘモグロビン小胞体分散液を生体内に静脈投与した
後、その酸素結合能がメトヘモグロビンの生起により低
下したときに血液を生体外に取り出すための採血手段
と;該採血手段から得られた血液から、前記ヘモグロビ
ン小胞体を分離するための分離手段と;分離された前記
ヘモグロビン小胞体の酸素結合能を回復するために、該
小胞体に光を照射するための手段と;前記酸素結合能を
回復したヘモグロビン小胞体を、前記生体内に戻すため
の手段とを具備した装置である。
明者らが既に公表した方法(Sakaiet al., Biotechnol.
Progress, 12, 119-125, 1996;Bioconjugate Chem.
8, 23-30, 1997)によって調製することができる。本発
明を適用するためのヘモグロビン小胞体は、この公知の
方法でヘモグロビン小胞体を調製する際に、該小胞体に
内包させる濃厚ヘモグロビン溶液に、予め電子供与体を
含有させておくことにより得ることができる。この電子
供与体としては、例えば1〜300 mMの、水酸基を有する
アルコール類(グリセリン、クエン酸等;更に、マンニ
トール、グルコース、スクロース、マルトース、ヒアル
ロン酸等の糖類を含む)またはアミノ酸(例えばトリプ
トファン、チロシン、ヒスチジン、メチオニン、セリン
等)を用いることができる。或いは、電子移動の中間体
として、1μM〜100 mMのフラビン類(フラビンモノヌク
レオチド:FMN、フラビンアデニンジヌクレオチド:FA
D、ルミフラビン、ジクロロリボフラビン、リボフラビ
ン:ビタミンB2、10-メチルイソアロキサジン)等のイ
ソアロキサジン環を有する物質を添加する。また、その
他の中間体としては、例えばフェナジンメトサルフェー
ト等のフェナジン環を有する物質、メチレンブルー等の
フェノチアジン環を有する物質、1,1’-ジメチル-4,4’
-ビピリジルおよびルテニウムトリ(2,2’-ビピリジン)
などのビピリジル基を有する物質、クレシルブルー等の
フェナゾニウム環を有する物質、インジゴスルホン酸等
のインジゴ基を有する物質、1-ナフトール-2-スルホン
酸インドフェノール等のインドフェノール環を有する物
質、トルイレンブルーなどのインダミン類の物質、アン
トラキノン-1,5-ジスルホン酸等のアントラキノン環を
有する物質、1,2-ナフトキノン-4-スルホン酸等のナフ
トキノン環を有する物質、ベンゾキノン環を有する物
質、N,N,N,N-テトラメチル-p-フェニレンジアミン等の
ベンゾアミン環を有する物質、カルバゾール基を有する
物質、インドール環を有する物質、ポルフィリン環を有
する物質等、いわゆる光増感剤として作動する物質を選
んで添加する。なお、これら電子移動の中間媒質は、還
元体のときにメトヘモグロビンへの電子供与体として作
動するので、この中間媒体への電子供与体としてメチオ
ニン、システイン等のアミノ酸、ニコチン、アスコルビ
ン酸、エチレンジアミンテトラ酢酸、ジメチルアミノプ
ロパノール等を1〜300mMの量で同時に添加する。
モグロビンがメトヘモグロビンに酸化された後に本発明
を適用することにより、これをヘモグロビンに還元して
酸素結合能を回復することができる。例えば、自動酸化
により生じたメトヘモグロビンを含有するヘモグロビン
小胞体分散液、または亜硝酸ナトリウムの添加によりメ
トヘモグロビンに酸化したヘモグロビン小胞体分散液を
用いて、本発明の効果を確認することができる。即ち、
この分散液を生理塩水溶液で所定の成分濃度(例えば、
ヘモグロビン濃度2.5μM)に希釈する。このとき、ヘモ
グロビン小胞体分散液を希釈しても、小胞体内水相の成
分濃度は希釈されずにそのまま維持される。このこと
は、本発明の方法を適用する上で極めて有利である。次
いで、光を透過するガラス製、プラスチック製もしくは
石英製の密封セル、または循環できるセルに収容し、こ
れに紫外・可視領域の光(280〜600 nm)を照射する。
その際、所望の波長領域の光を得るために、フィルター
の組合わせおよびレーザー光源を選択すればよい。これ
により、小胞体中のメト化したメトヘモグロビンは次第
に還元される。
生体内でメト化したヘモグロビン小胞体を、生体外に取
り出して光還元により酸素結合能を回復させ、これを再
度生体内に投与する態様で実施することもできる。例え
ば、出血性ショックの際の蘇生液、血液希釈、体外循環
等の各種の適用において、上記ヘモグロビン小胞体を生
体内に投与すると、これに含まれるヘモグロビンは次第
にメトヘモグロビンに酸化されて酸素運搬能力が低下す
る。このようなときに、カテーテルを経由して血液の一
部を体外に取り出した後、例えば図1に示すような装置
を用いて本発明の方法を実施するのが好ましい。
採取された血液は、ポンプ1により遠心分離ユニット2
に送られる。遠心分離ユニット2では、血液を血球成分
層とヘモグロビン小胞体を含有する血漿層とに分離す
る。ヘモグロビン小胞体の粒径は200〜300 nmであり、
血球成分に比較して1/40と小さいため両者は容易に分離
され、ヘモグロビン小胞体は血漿層に浮遊した状態で回
収される。ヘモグロビン小胞体を含有する血漿層は、ポ
ンプ3により透明な照射装置5に通される。照射装置を
通る際に、血液は光源6から所定波長の光を照射され
て、メトヘモグロビンのヘモグロビンへの還元が行われ
る。このように、血球成分とヘモグロビン小胞体とを分
離し、ヘモグロビン小胞体にのみ光照射を行うので、赤
血球内ヘモグロビンの光吸収による光還元の阻害や、血
球成分自体への光照射の影響を低減することができる。
また、酸素は本発明の光還元を阻害する場合があるの
で、酸素除去装置4(例えば人工肺)等を利用して、溶
液中の酸素を除去することが好ましい。なお、光還元反
応における光照射の効率を高めるために、被照射液を希
釈したり液厚をできるだけ薄くする必要があるので、照
射装置5では、例えば被照射液を中空ファイバー内で循
環させたり、液膜を形成させてこれに光を照射するよう
にするのが好ましい。こうして光還元された血漿層は、
遠心分離ユニット2で分離された血球層と合体され、ポ
ンプ7により体内に返血される。
モグロビン小胞体が酸化されて酸素結合能力が低下した
ときに、光照射をすることによって酸素結合能を回復す
ることができる。例えば長期保存後にメトヘモグロビン
が生じている場合に、光照射により還元してから体内投
与を行えば、酸素運搬能を最大限活用することができ
る。また、体内投与すると血管内を循環するうちに次第
にメトヘモグロビン量が増大するが、このときにも経皮
的にまたは上述の体外循環経路を設けて光照射すれば、
酸化したヘモグロビン小胞体は還元型ヘモグロビン小胞
体に変換され、再度酸素を結合することができ、結果と
して酸素輸液としての機能を持続することができる。
とは、電子供与体としての添加剤とヘモグロビンが濃度
高く小胞体に内包されているので、均一溶液系よりも速
やかな還元が可能になることである。また光照射を停止
すれば還元反応も停止するので、電子供与体の酸素酸化
による消費も抑えられる。

製して得た高純度ストロマフリーヘモグロビン溶液(40
g/dL,6.2mM)に、マンニトールを濃度10mMおよび100mM
となるように添加した。このときのモル比[マンニトー
ル]/[ヘモグロビン]はそれぞれ1.6,16である。Remo
linoTM(日本Millipore社製)を用いて孔径0.22μmのF
Mミクロフィルター(富士フィルム社製)で濾過し、仕込
みヘモグロビン溶液を得た。混合脂質粉末Presone PPG-
I(ホスファチジルコリン/コレステロール/ホスファ
チジルグリセロールの混合物、日本精化社製)を脂質濃
度が4.5wt%となるように少量ずつ添加し、4℃で終夜
撹拌してヘモグロビン内包多重層小胞体を得た。Remoli
noを用いたエクストルージョン法により粒径及び被覆層
数の制御を行った。FMミクロフィルターを孔径3,0.
8,0.65,0.45,0.3,0.22μmの順に使用した。得られ
たヘモグロビン小胞体溶液を生理食塩水で希釈し、超遠
心分離(50,000g,40min)後に上澄みヘモグロビン溶液
を吸引除去した。生理食塩水に溶解させたポリオキシエ
チレン結合脂質[N-(モノメトキシポリエチレングリコ
ール-カルバミル)ジステアロイルホスファチジルエタノ
ールアミン;N-(monomethoxy polyethyleneglycol-carb
amyl)distearoyl phosphatidylethanolamine、ポリエチ
レングリコール鎖の分子量は5300]を、小胞体外表面の
脂質の0.3mol%分を滴下し、25℃で二時間撹拌後に4
℃で終夜撹拌してヘモグロビン小胞体の表面をポリエチ
レングリコールで修飾した。ヘモグロビン濃度を10g/dL
とし、Dismic-25:0.45μmフィルター(ADVANTEC)で濾
過し、ポリエチレングリコールで修飾したヘモグロビン
小胞体を得た。
ウムを添加し、メトヘモグロビン含量をほぼ100%とし
た。超遠心分離により小胞体を沈殿させて上清の亜硝酸
ナトリウムを完全に除去した後、石英製のセルにメトヘ
モグロビン濃度2.5μMになるようにリン酸緩衝生理食塩
水(pH 7.4)入れて、一酸化炭素を通気した。超高圧水銀
ランプ(USH-250D,ウシオ電機)とフィルター(U-36
0,HOYA)を組合わせて、365nmを中心とする光を照射し
た。メトヘモグロビンの最大吸収波長は405nmである
が、これが次第に減少して代わって419nmの一酸化炭素
結合ヘモグロビン小胞体に変換した。還元はマンニトー
ル10mM内包した系では10分で60%が、100mM内包した系
では65%が還元された。またマンニトール10mM内包した
系では50分で、100mM内包した系では30分以内に完了し
た。次に、ハロゲンランプ(500W)を用い、酸素通気下で
3分間可視光照射すると、最大吸収波長は415nmに変化し
たことから、一酸化炭素結合ヘモグロビン小胞体は酸素
を結合したオキシヘモグロビン小胞体に変換された。
するメトヘモグロビン小胞体を石英製のセルにメトヘモ
グロビン濃度2.5μMになるように入れて、アルゴン雰囲
気下、同様の光照射をしたところ、デオキシヘモグロビ
ン由来の430nmの吸収が増大し120分で80%の還元が進行
した。これに酸素を通気すると415nmにピークが出現
し、酸素を結合したオキシヘモグロビン小胞体の生成を
確認した。
4)を濃度2.5μMとし、これにマンニトールをモル比で1
6倍量添加し、ヘモグロビン小胞体と同一の条件で光還
元を実施した。一酸化炭素雰囲気における還元は、120
分で70%進行したに留まった。マンニトールの添加量を
ヘモグロビンに対して40000倍モル(100mM)までに高め
ると50分で還元が完了した。また、アルゴン雰囲気では
還元は進行しなかった。従って、ヘモグロビン溶液では
ヘモグロビン小胞体に比較して還元の効率が劣り、高濃
度のマンニトール添加の必要があることが明らかとなっ
た。
トールの代わりにトリプトファンを内水相ヘモグロビン
に10mMおよび100mM導入した場合について同様の調製法
によりトリプトファン内包ヘモグロビン小胞体を得た。
このときのモル比[トリプトファン]/[ヘモグロビ
ン]はそれぞれ1.6,16である。37℃にてインキュベー
ションを48時間すると、メトヘモグロビン含量が42%に
達した。これを石英製のセルにヘモグロビン濃度2.5μM
になるように入れて、一酸化炭素を通気した。同様の方
法により365nmを中心とする光を照射した。メトヘモグ
ロビンの最大吸収波長は405nmであるが、これが次第に
減少して代わって419nmのピークが増大し、一酸化炭素
結合型ヘモグロビン小胞体に変換した。還元はトリプト
ファン10mM内包した系では10分で33%が、100mM内包し
た系では43%が還元された。またトリプトファン10mM内
包した系では90分で、100mM内包した系では50分以内に
完了した。続いて可視光を照射しながら酸素を通気させ
ると速やかに最大吸収波長は415nmに変化したことか
ら、酸素を結合したオキシヘモグロビン小胞体に変換さ
れたことを確認した。
トールの代わりにフラビンモノヌクレオチドを5 mMと、
メチオニンを200 mM添加した場合について同様の調製法
によりヘモグロビン小胞体を得た。37℃にて遮光してイ
ンキュベーションを48時間すると、メトヘモグロビン含
量が40%に達した。これを硝子製のセルにヘモグロビン
濃度2.5μMになるように入れて、窒素を通気した。ハロ
ゲンランプ(500W)とフィルター(L-39/HA-30,HOYA)を組
合わせて、400−600 nmの可視光を照射した。Q帯スペク
トルは次第に555nmの最大吸収波長を示し、デオキシ型
ヘモグロビン小胞体に還元されたことを確認した。還元
は5分以内に完了した。続いて酸素を通気させるとQ帯の
スペクトルは555nmのピークは消失し、かわって最大吸
収波長541nm,576nmを示したことから、酸素を結合した
オキシヘモグロビン小胞体に変換されたことを確認し
た。
せたのち、頸動脈と頸静脈に挿管した。グルコース100m
M内包ヘモグロビン小胞体分散液(ヘモグロビン濃度10g/
dL,4mL)を頸静脈より1mL/minの速度で投与した。12時
間後に頸動脈から2mLの血液を抜き取り、これをEDT
A添加済の採血管(テルモ)に入れて遠心分離(2000g,10
分)することでヘモグロビン小胞体分散液を上澄みに得
た。下層の血球成分は生理食塩水で希釈して頸静脈より
直ちに投与した。上層のヘモグロビン小胞体溶液は、30
%のヘモグロビンが酸化されていた。このメトヘモグロ
ビン小胞体を石英製のセルに入れて窒素をバブルしたの
ち365nmを中心とする光を照射し、還元を行った。還元
型のデオキシヘモグロビン含量が95%になったところで
光照射を止め、孔径0.45μmの滅菌フィルタを通過させ
た後に頸静脈より投与した。
全身麻酔し、気管内挿管して機械呼吸管理とした。一回
換気量20mL/kg,呼吸回数を12回/minとした。大腿動脈
より240mLを急速脱血すると、体動脈圧は初期値の約50
%にまで低下した。実施例4で調製したフラビンモノヌ
クレオチドを20μMと、メチオニンを100mMを内水相に含
むヘモグロビン小胞体を静注したところ、血圧は脱血前
と同等にまで回復した。12時間経過後にヘモグロビン
小胞体のメト化率は40%に達していたので、血液を10
0mL大腿動脈から採血し、これを生理食塩水で三倍希釈
したのち、限外濾過膜(Millipore社製、ミニカセットDV
PP,公称分画径0.65μm、濾過面積0.1m2)を用いて血球
成分とヘモグロビン小胞体を分離した。血球成分は循環
相側で濃縮され、分離後に直ちに静注した。濾液のヘモ
グロビン小胞体の分散液は、1リットルの硝子容器に入
れた後、撹拌しながら可視光(360W−ナトリウムラン
プ、理工科学産業)を照射した。95%以上の還元率に到
達したことを確認後に照射を止め、限外濾過膜(Millipo
re社製、Biomax-1000,公称限外分子量1000kDa)にて濃
縮後、静注した。
させた後、頚動脈と頚静脈に挿管し、頚動脈から血液を
1 mL/minの速度で3 mLを採取し、出血ショック状態とし
た。30分後に実施例4で調製したフラビンモノヌクレオ
チド100μMを、メチオニン100 mMとを内水相に含むヘモ
グロビン小胞体分散液(ヘモグロビン濃度10 g/dL, 3m
L)を頚静脈より1mL/minの速度で投与した。24時間経過
後に頚動脈から血液を100μL抜き取り、ヘモグロビン小
胞体のメトヘモグロビン含量を測定したところ、48%に
達していた。ヌードラットを硝子板の上に置き、360Wの
ナトリウムランプ(理工化学産業)二本を用いて上下か
ら可視光を照射した。目には光が当たらないように、頭
部全体には黒い布をかぶせた。10分間照射した後、頚動
脈より血液を100μL抜き取り、ヘモグロビン小胞体のメ
トヘモグロビン含量を測定したところ、21%にまで低下
していた。これにより、本発明の方法は光りを経皮的に
照射した場合にも効果が得られることが分かった。

たときの還元率の推移を示した図である。Hb濃度は2.5
μM。

Claims (3)
- 【請求項1】 酸素輸液として使用するヘモグロビン小
胞体分散液の低下した酸素結合能を回復させる方法であ
って:リン脂質小胞体内にヘモグロビン水溶液を内包し
且つ前記小胞体の内水相に電子供与体を含有するヘモグ
ロビン小胞体の分散液に対して、前記ヘモグロビンがメ
トヘモグロビンに酸化されて酸素結合能が低下したとき
に光を照射することにより、メトヘモグロビンをヘモグ
ロビンに還元し、その酸素結合能を回復させることを特
徴とする方法。 - 【請求項2】 前記電子供与体がアミノ酸類、糖類、ア
ルコール類およびフラビン類からなる群から選択される
請求項1に記載の方法。 - 【請求項3】 請求項1に記載の方法を実施するための
装置であって:請求項1で定義したヘモグロビン小胞体
分散液を生体内に静脈投与した後、メトヘモグロビンの
生起によりその酸素結合能が低下したときに血液を生体
外に取り出すための採血手段と、 該採血手段から得られた血液から、前記ヘモグロビン小
胞体を分離するための分離手段と、 分離された前記ヘモグロビン小胞体の酸素結合能を回復
するために、該小胞体に光を照射するための手段と、 前記酸素結合能を回復したヘモグロビン小胞体を、前記
生体内に戻すための手段とを具備した装置。
