Claims (1)
- 【特許請求の範囲】 1. 少なくとも1つの核酸を含有しその少なくとも一部分が多重ラメラ小胞の 内部に含まれている組成物であって、前記小胞は、少なくとも1つの界面活性剤 からなる二重層より構成されるものであり、前記二重層は、同心円状であって、 かつ、前記小胞に玉ねぎ構造を付与するものであることを特徴とする組成物。 2. 小胞は、1μm未満、好ましくは0.1〜1μmの範囲の大きさを有する ものである請求項1記載の組成物。 3. 小胞の二重層を構成する界面活性剤は、非カチオン性界面活性剤である請 求項1又は2記載の組成物。 4. 非カチオン性界面活性剤は: −水素化された又は水素化されていないリン脂質、 −酸、又は、アルカリ金属塩、アルカリ土類金属塩若しくはアミン塩の形態を採 る、飽和の又はモノ−若しくはポリ不飽和の、直鎖状又は分枝状のC6〜C18の 脂肪酸、 −上記脂肪酸と ・ショ糖の ・ソルビタンの ・マンニトールの ・グリセロール又はポリグリセロールの ・グリコールの エトキシ化された又はエトキシ化されていないエステル、 −上記脂肪酸のモノ−、ジ−若しくはトリグリセリド又はグリセリドの混合物 −飽和の又はモノ−若しくはポリ不飽和の、直鎖状又は分枝状のC6〜C18の脂 肪アルコール、 −上記脂肪アルコールと ・ショ糖の ・ソルビタンの ・マンニトールの ・グリセロール又はポリグリセロールの ・グリコールの エトキシ化された又はエトキシ化されていないエステル、 −水素化された又は水素化されていないポリエトキシ化植物油、 −ポリオキシエチレンとポリオキシプロピレンのブロックポリマー[ポロキサマ ー(poloxamer)]、 −ポリエチレングリコールヒドロキシステアレート、 −コレステロール又はシトステロール等のステロール骨格のアルコール からなる群より選択されるものである請求項3記載の組成物。 5. 小胞の二重層は、少なくとも2つの界面活性剤からなるものであって、そ のうちの1つは、1〜6の範囲の親水性親油性比(HLB)を有し、その他は、 3〜15の範囲の親水性親油性比(HLB)を有する請求項1〜4のいずれか1 項に記載の組成物。 6. 核酸の10%以上、好ましくは40%以上が、多重ラメラ小胞の内部に含 まれる請求項1〜5のいずれか1項に記載の組成物。 7. 核酸の60〜100%が、小胞に含まれる請求項1〜6のいずれか1項に 記載の組成物。 8. 核酸は、DNA又はDNA由来のヌクレオチド配列である請求項1〜7の いずれか1項に記載の組成物。 9. 核酸は、特定の遺伝子又は前記遺伝子由来の配列、特に所定のタンパク質 をコードする配列である請求項1〜7のいずれか1項に記載の組成物。 10. 核酸は、RNA又はRNA由来のヌクレオチド配列である請求項1〜7 のいずれか1項に記載の組成物。 11. 核酸は、センスオリゴヌクレオチド及び/又はアンチセンスオリゴヌク レオチドである請求項1〜7のいずれか1項に記載の組成物。 12. 小胞は、更に、核酸とともにカプセル封入されたカチオン性アジュバン ト、特にポリエチレンイミンを含有する請求項1〜11のいずれか1項に記載の 組成物。 13. DNA凝集剤、特に、ヒストン、組込み又は組換え酵素、トポイソメラ ーゼ又はヘリカーゼ等の、カプセル封入された核酸の複製を最適化するための酵 素、からなる群より選択された少なくとも1つのカプセル封入された化合物を含 有する請求項1〜12のいずれか1項に記載の組成物。 14. 小胞は、生体内のターゲットに特異的に認識されることが可能な化合物 の添加、特に、モノクローナル抗体又はFabフラグメントの添加によって、表 面が修飾されたものである請求項1〜13のいずれか1項に記載の組成物。 15. 核酸を配合したラメラ液晶相を製造し、剪断力を用いて前記液晶相を多 重ラメラ小胞に再配列させることからなることを特徴とする、請求項1〜14の いずれか1項に記載の組成物の製造方法。 16. 薬理学的に許容される媒体中の懸濁液の状態で、請求項1〜14のいず れか1項に記載の小胞を含有する医薬用組成物。 17. カプセル封入された核酸は、完全遺伝子、特に、欠失又は欠損している 遺伝子を補うことを目的とした遺伝子、である請求項16記載の医薬用組成物。 18. 核酸は、特定の遺伝子の過剰発現又は活性の低減を誘発することを目的 としたオリゴヌクレオチドである請求項16記載の医薬用組成物。 19. 核酸は、抗原タンパク質をコードするDNAである、ワクチンとしての 請求項16記載の医薬用組成物。 20. 特にガン学若しくはウイルス学の分野又は骨の欠陥の修復において、欠 失若しくは欠損した細胞機能を修復し若しくは改善すること、又は、細胞に新し い機能を付与することを目的とした医薬用組成物の製造における、請求項1〜1 4項のいずれか1項に記載の玉ねぎ構造の多重ラメラ小胞の使用。 21. 請求項1〜14項のいずれか1項に記載の組成物で細胞系を処理するこ とからなることを特徴とする、前記細胞系の生体外形質転換方法。
