- 【特許請求の範囲】 1.哺乳動物身体内への移植後の生物学的組織の石灰化を阻害するために、少 なくとも部分的に固定された生物学的組織を処理する方法であって、以下の工程 : 該生物学的組織を処理溶液に浸漬する工程;および 該溶液を加熱する工程;および 該加熱工程の間、相対的かつ反復的な組織/溶液運動を誘発する工程、 を包含する、方法。 2.前記処理溶液を容器内に収容する工程をさらに包含する、請求項1に記載 の方法。 3.前記誘発工程が、前記容器を振とうする工程を包含する、請求項2に記載 の方法。 4.前記誘発工程が、前記容器内の前記溶液を撹拌する工程を包含する、請求 項2に記載の方法。 5.前記浸漬した組織が前記容器内で大きく動かないようにする工程をさらに 包含する、請求項4に記載の方法。 6.処理容器を有する処理装置を提供する工程をさらに包含し、前記加熱工程 が該容器の外部に熱を加え、該容器内の前記溶液を間接的に加熱する工程を包含 する、請求項1に記載の方法。 7.前記処理容器を囲いの中に置く工程をさらに包含し、前記熱を加える工程 が該囲いを加熱する工程を包含する、請求項6に記載の方法。 8.前記加熱工程が、前記処理溶液に直接熱を加える工程を包含する、請求項 1に記載の方法。 9.前記加熱工程が、前記処理溶液を約45〜55℃の間に加熱する工程を包含す る、請求項1に記載の方法。 10.前記加熱工程が、前記処理溶液を約50℃に加熱する工程を包含する、請 求項9に記載の方法。 11.前記相対的な組織/流体運動を少なくとも30分間誘発する工程をさらに 包含する、請求項1に記載の方法。 12.約15日間と60日間の間の期間、前記相対的な組織/流体運動を誘発する 工程をさらに包含する、請求項11に記載の方法。 13.前記組織が部分的に固定されるのみであり、前記相対的な組織/流体運 動を誘発する前記工程を十分長く続け、該組織を前記処理溶液に浸漬している間 に完全に固定化する、請求項11に記載の方法。 14.前記相対的な組織/流体運動を誘発する前記工程の前に、前記組織を完 全に固定化する、請求項1に記載の方法。 15.前記浸漬工程が、前記組織を、0.2%と0.8%の間の濃度の緩衝化グルタ ルアルデヒドの処理溶液内に浸漬する工程を包含する、請求項1に記載の方法。 16.前記誘発工程が、前記処理溶液を前記組織を横切るように流し、かつ前 記浸漬した組織が大きく動かないようにする工程を含む、請求項1に記載の方法 。 17.哺乳動物身体内への移植後の生物学的組織の石灰化を阻害するために、 少なくとも部分的に固定された生物学的組織を処理する方法であって、以下の工 程: 該組織をフロー容器内に置く工程; 該組織が該容器内で大きく動かないようにする工程; 処理溶液を該フロー容器を通じて流し、該組織と接触させる工程;および 該流す工程中、該溶液を加熱する工程、 を包含する方法。 18.前記動かないようにする工程が、前記組織を平坦な配置で、前記流れる 溶液の流れ方向に対して実質的に平行に設ける工程を包含する、請求項17に記載 の方法。 19.前記流れる溶液の流れ方向に対して実質的に垂直に配向する断面を有す るフロー容器内に前記組織を配置し、バッフルの下流に位置する組織が、その断 面に亘って実質的に均一な下流の流れのプロファイルを作る、請求項17に記載の 方法。 20.処理溶液を前記フロー容器の入り口へ、レザバから供給し、流体を該フ ロー容器の出口から、該レザバへと排出する工程をさらに包含する、請求項17に 記載の方法。 21.前記レザバ内の前記処理溶液を加熱する工程をさらに包含する、請求項 20に記載の方法。 22.前記フロー容器を、ミシン目の付いたバッフルによって一連のセクショ ンに分割し、複数の組織を個々のセクションに配置する工程をさらに包含する、 請求項17に記載の方法。 23. 前記フロー容器内に入り口および出口をさらに有し、前記方法が、処 理流体を該フロー容器を通じて該入り口から該出口へと流し、前記組織と接触さ せる工程を包含する、請求項17に記載の方法。 24.哺乳動物身体内への移植後の生物学的組織の石灰化を阻害するために、 少なくとも部分的に固定された生物学的組織を処理する装置であって、以下: フロー容器; 処理溶液の供給源; 該容器への流体インプット; 該容器からの流体アウトプット; 少なくとも部分的に固定した生物学的組織を該容器内の該インプットおよびア ウトプットの間の配置し、該組織がその中で大きく動かないようにするための、 組織マウント;および 該流体を加熱する手段、 を有する、装置。 25.前記フロー容器が、ミシン目の付いたバッフルによって分離された、少 なくとも2つの一連のセクションを有し、個々のセクション内に少なくとも1つ の組織マウントがある、請求項24に記載の装置。 26.前記フロー容器が長いチューブであって、前記バッフルが円形である、 請求項25に記載の装置。 27.前記組織マウントが、前記組織を平坦な配置で、前記容器を通じて流れ る前記溶液の流れ方向に対して実質的に平行に設けるように配置されている、請 求項24に記載の装置。 28.前記フロー容器内の、前記組織マウントの上流に配置された少なくとも 1つのバッフルをさらに有し、該バッフルが、実質的に均一な下向きの流れを作 り、該フロー容器の断面の輪郭を作るように配置されている、請求項24に記載の 装置。 29.前記バッフルがミシン目の付いたプレートであり、前記容器を通じて流 れる前記溶液の流れ方向に対して実質的に垂直に配向しており、前記フロー容器 が、少なくとも2つの一連のセクションに分割されており、それぞれ2つの隣接 するセクションがバッフルによって分離されており、少なくとも1つの組織マウ ントが個々のセクション内で、該バッフルの1つに取り外し可能に固定されてい る、請求項28に記載の装置。 30.前記流体の供給源がレザバを有し、前記流体を加熱する前記手段が、該 レザバ内の流体を加熱するためのヒーターを有する、請求項24に記載の装置。 31.前記レザバ内の前記流体の温度をモニターするためのセンサー、および 感知した温度に応答性であり、該ヒーターの温度を調節するための、フィードバ ック制御ループをさらに有する、請求項30に記載の装置。 32.前記フロー容器が直立したチューブを有し、前記入り口の開口が該チュ ーブの下端にあり、前記出口の開口が該チューブの上端にあり、前記装置が、該 入り口の開口の上に減速装置を、および該減速装置の上であって前記第一組織マ ウントの下に整流装置を、さらに有する、請求項24に記載の装置。
