- 【特許請求の範囲】 1.(a)配列番号2のアミノ酸1から69を含むポリペプチドをコードするポリヌ クレオチドと少なくとも70%の同一性を持つポリヌクレオチド; (b)配列番号4に示すアミノ酸1からアミノ酸69を含むポリペプチドをコードす るポリヌクレオチドと少なくとも70%の同一性を持つポリヌクレオチド; (c)配列番号6に示すアミノ酸1からアミノ酸74を含むポリペプチドをコードす るポリヌクレオチドと少なくとも70%の同一性を持つポリヌクレオチド; (d)(a),(b)または(c)のポリヌクレオチドに相補的なポリヌクレオチド;および (e)(a),(b),(c)または(d)のポリヌクレオチドの少なくとも15塩基を含むポリヌ クレオチド からなる群より選択されるメンバーを含む、単離されたポリヌクレオチド。 2.前記ポリヌクレオチドがDNAである、請求項1に記載のポリヌクレオチド。 3.前記ポリヌクレオチドがRNAである、請求項1に記載のポリヌクレオチド。 4.前記ポリヌクレオチドがゲノムDNAである、請求項1に記載のポリヌクレオ チド。 5.配列番号2のアミノ酸1〜69を含むポリペプチドをコードする、請求項2に 記載のポリヌクレオチド。 6.配列番号4のアミノ酸1〜69を含むポリペプチドをコードする、請求項2に 記載のポリヌクレオチド。 7.配列番号6のアミノ酸1〜74を含むポリペプチドをコードする、請求項2に 記載のポリヌクレオチド。 8.(a)ATCC寄託番号97401に含まれるヒトcDNAによって発現されるアミノ酸配列 を持つ成熟ポリペプチドをコードするポリヌクレオチド; (b)ATCC寄託番号97402に含まれるヒトcDNAによって発現されるアミノ酸配列を持 つ成熟ポリペプチドをコードするポリヌクレオチド; (c)ATCC寄託番号97403に含まれるヒトcDNAによって発現されるアミノ酸配列を持 つ成熟ポリペプチドをコードするポリヌクレオチド; (d)(a),(b)または(c)のポリヌクレオチドに相補的なポリヌクレオチド;および (e)(a),(b),(c)または(d)のポリヌクレオチドの少なくとも15塩基を含むポリ ヌクレオチド からなる群より選択されるメンバーを含む、単離されたポリヌクレオチド。 9.配列番号1のヌクレオチド106からヌクレオチド312を含む、請求項1に記載 のポリヌクレオチド。 10.配列番号3のヌクレオチド103からヌクレオチド309を含む、請求項1に記 載のポリヌクレオチド。 11.配列番号5のヌクレオチド109からヌクレオチド330を含む、請求項1に記 載のポリヌクレオチド。 12.請求項2に記載のDNAを含むベクター。 13.請求項12に記載のベクターを含む宿主細胞。 14.ポリペプチドを産生するプロセスであって、前記DNAによってコードされ るポリペプチドを請求項13に記載の宿主細胞から発現する工程を含む、プロセ ス。 15.ポリペプチドを発現する細胞を産生するためのプロセスであって、細胞が ベクター内のヒトcDNAによってコードされるポリペプチドを発現するように、請 求項12に記載のベクターで細胞を遺伝的に操作する工程を含む、プロセス。 16.(a)配列番号2のアミノ酸1〜69を含むポリペプチド; (b)配列番号4のアミノ酸1〜69を含むポリペプチド; (c)配列番号6のアミノ酸1〜74を含むポリペプチド; (d)(a)、(b)または(c)のポリペプチドと少なくとも70%が同一であるポリペプチ ド からなる群より選択されるメンバーを含む、ポリペプチド。 17.前記ポリペプチドが配列番号2のアミノ酸1からアミノ酸69を含む、請求 項16に記載のポリペプチド。 18.前記ポリペプチドが配列番号4のアミノ酸1からアミノ酸69を含む、請求 項16に記載のポリペプチド。 19.前記ポリペプチドが配列番号6のアミノ酸1からアミノ酸74を含む、請求 項16に記載のポリペプチド。 20.請求項16のポリペプチドの活性化を阻害する化合物。 21.hESF I、IIまたはIIIを必要とする患者を処置するための方法であって、 請求項16に記載のポリペプチドの治療的に有効な量を患者に投与する工程を含 む、方法。 22.前記治療的に有効な量のポリペプチドが、該ポリペプチドをコードするDN Aを患者に投与し、そしてインビボで該ポリペプチドを発現することにより投与 される、請求項21に記載の方法。 23.hESF I、IIまたはIIIポリペプチドを阻害することが必要である患者を処 置するための方法であって、治療的に有効な量の請求項20に記載の化合物を患 者に投与する工程を包含する、方法。 24.請求項16に記載のポリペプチドの過小発現に関連する疾患またはその疾 患に対する罹患性を診断するためのプロセスであって、該ポリペプチドをコード する核酸配列中の変異を決定する工程を包含する、プロセス。 25.診断プロセスであって、宿主由来のサンプル中の請求項16に記載のポリ ペプチドの存在について分析する工程を包含する、プロセス。 26.請求項16に記載のポリペプチドに結合し、そして該ポリペプチドの活性 化を阻害する化合物を同定するための方法であって: その表面上でポリペプチドのためのレセプターを発現する細胞を、分析的に検 出可能なhESF I、IIまたはIIIポリペプチドおよび化合物と、レセプターへの結 合を許容する条件下で接触させる工程、該レセプターは、化合物のレセプターへ の結合に応答して検出可能なシグナルを提供し得る第2の成分に関連する;およ び、 化合物がレセプターに結合し、そして該レセプターを阻害するかどうかを、hE SF I、IIまたはIIIとレセプターとの相互作用から生じるシグナルの不在を検出 することにより決定する工程、 を包含する、方法。
