Claims (1)
- 【特許請求の範囲】 1.数種の微生物混合集団における個々の微生物種またな下位系統発生亜種の 相対的存在量の定量方法であって、以下の工程からなる方法: 1)標識したインシチュー(in situ)ハイブリッド形成用クラスターオリゴ ヌクレオチドプローブのセットを調製する工程; 2)当該プローブを当該混合集団の標本とハイブリッド形成する工程; 3)標識された微生物数を定量分析する工程。 2.当該プローブが蛍光物質で標識してある請求項1の方法。 3.当該プローブが標的微生物の16S rRNAに基づくものである請求項1また は2の方法。 4.当該定量分析を顕微画像分析にて実施する前記請求項のいずれかの方法。 5.当該プローブが微生物の系統発生的クラスターに特異的である前記請求項 のいずれかの方法。 6.当該系統発生的クラスターが属または種である請求項5の方法。 7.当該微生物がバクテリアである前記請求項のいずれかの方法。 8.混合集団が特に哺乳類、とりわけヒトの常在腸管フローラ由来のものであ る前記請求項のいずれかの方法。 9.混合集団における個々の微生物種の相対的存在量動態を分析する方法であ って、前記請求項のいずれかの方法を、一つの集団から異なった時期に採取した 標本につき少なくとも2度実施することを特徴とする方法。 10.標本を前処理および/または精製処理する前記請求項のいずれかの方法 。 11.少なくとも二種の異なったプローブに異なった標識物を備える前記請求 項のいずれかの方法。 12.クラスターに特異的なプローブであって、前記請求項のいずれかの方法 に使用する少なくとも一種の標識物を備えているプローブのセット。 13.バクテリア16S rRNAから誘導される請求項12のプローブのセット。 14.標識物が蛍光物質である請求項12または13のプローブのセット。 15.異なった標識物を備える少なくとも二種の異なったプローブを含む前記 請求項12−14のいずれかのプローブのセット。 16.数種の微生物混合集団における個々の微生物種の相対的存在量を定量す るためのキットであって、請求項12〜15のいずれかのプローブのセットと、 標本を前処理するのに適当な物質および/またはハイブリッド形成を実施するの に適当な物質および/またはハイブリッド形成の結果を分析するための適当な物 質とを含むキット。
