Claims (1)
- 【特許請求の範囲】 1. 血漿を形成するために全血試料から赤血球を分離するためのフィルター であって、 固相支持体;および 赤血球のための凝集素であって、全血試料中では不溶であり、固相支持体上に さらに固定化されている凝集素; を含むフィルター。 2. フィルターが第2の固相をさらに含み、固相支持体が第2の固相の中に 固定化されていて、それによって固相支持体上に固定化されている不溶性凝集素 が第2の固相の中にも固定化されている、請求の範囲第1項記載のフィルター。 3. 固相支持体が非多孔性粒子であり、第2の固相が多孔性物質である、請 求の範囲第2項記載のフィルター。 4. 非多孔性粒子が約0.4μmの直径を有する、請求の範囲第3項記載の フィルター。 5. 固相支持体および固定化された凝集素が、約0.1重量%から約0.2 5重量%/容量の濃度で第2の固相に適用される、請求の範囲第2項記載のフィ ルター。 6. 固定化された固相支持体が第2の固相の中に物理的に封入されている、 請求の範囲第2項記載のフィルター。 7. 凝集素が固相支持体上に約10:1の質量比を使用して固定化されてい る、請求の範囲第1項記載のフィルター。 8. フィルターが、全血試料の流動路の中に固相支持体を物理的に保持する ための手段をさらに含む、請求の範囲第1項記載のフィルター。 10. 不溶性凝集素が、固相支持体に直接的に化学的に結合している、請求 の範囲第1項記載のフィルター。 11. 固相支持体が、セルロース、ガラス繊維、ガラス繊維と混合した合成 物質を有する複合材料、ニトロセルロース、ポリエチレン、ナイロン、およびポ リエステルから成る群から選択される織物またはキャスト材料である、請求の範 囲第1項記載のフィルター。 12. 凝集素が、ヒト赤血球に対するウサギIgG抗体、大豆凝集素、小麦 胚芽凝集素、およびレクチンから成る群から選択される、請求の範囲第1項記載 のフィルター。 13. フィルターが約20μLの全血から約10μLの血漿を形成する、請 求の範囲第1項記載のフィルター。 14. 血漿が3分以内に形成される、請求の範囲第13項記載のフィルター 。 15. フィルターが全血の試料から約40容量%から約50容量%の血漿を 形成する、請求の範囲第1項記載のフィルター。 16. 固相支持体上に固定化された凝集素の量が、全血の試料中の赤血球の 数とほぼ同一数の結合部位を提供する、請求の範囲第1項記載のフィルター。 17. 血漿を形成するために全血試料から赤血球を分離するためのフィルタ ーであって、 非多孔性粒子から作られた固相支持体; 赤血球のための凝集素であって、全血試料中では不溶であり、固相支持体に直 接的に化学的に結合することによって固相支持体上にさらに固定化されていて、 固相支持体上に固定化された凝集素の量が全血の試料中の赤血球の数とほぼ同一 数の結合部位を提供し、そして全血試料の容量の少なくとも40%が血漿を形成 する、凝集素;および 多孔性物質から作られた第2の固相であって、固相支持体が第2の固相の中に 固定化されていて、それによって固相支持体の上に固定化された不溶性凝集素が 第2の固相の中にも固定化されている、第2の固相; を含むフィルター。 18. 血漿を形成するために全血試料から赤血球を分離するためのフィルタ ーであって、 内部空間を画定するハウジングであって、内部空間とハウジングの外部との間 に流体伝達を提供するための入口と出口とを有するハウジング; 内部空間の中に固定化された固相支持体; 全血試料の流動路の中に固相支持体を物理的に保持するための手段;および 赤血球のための凝集素であって、全血試料中では不溶であり、固相支持体上に さらに固定化されている凝集素; を含むフィルター。 19. 保持手段が、固相支持体に不透過性で血漿に透過性であり、出口を覆 う膜を含む、請求の範囲第18項記載のフィルター。 20. 保持手段が、固相支持体に不透過性であり、入口を覆う膜をさらに含 む、請求の範囲第19項記載のフィルター。 21. 固相支持体が非多孔性粒子である、請求の範囲第18項記載のフィル ター。 22. 凝集素が固相支持体上に約10:1の質量比を使用して固定化されて いる、請求の範囲第18項記載のフィルター。 23. 不溶性凝集素が、固相支持体に直接的に化学的に結合している、請求 の範囲第18項記載のフィルター。 24. フィルターが約20μLの全血から約10μLの血漿を形成する、請 求の範囲第18項記載のフィルター。 25. 血漿が3分以内に形成される、請求の範囲第18項記載のフィルター 。 26. フィルターが全血の試料から約40容量%から約50容量%の血漿を 形成する、請求の範囲第18項記載のフィルター。 27. 固相支持体上に固定化された凝集素の量が、全血の試料中の赤血球の 数とほぼ同一数の結合部位を提供する、請求の範囲第18項記載のフィルター。 28. 全血の試料中の複数の分析物の少なくとも1つの存在を測定するため の装置であって、 外部表面を有するハウジング; その存在を測定するために選択された分析物を含む全血の試料を受容するため の試料受容手段であって、ハウジングの外部表面上に位置している試料受容手段 ; 試料受容手段からの全血の試料を受容するためのフィルターであって、固相支 持体、赤血球のための凝集素であって全血試料中では不溶であり、固相支持体上 にさらに固定化されていて、それによって選択された分析物を含む血漿の試料が 形成される凝集素を有するフィルター;および 血漿の試料中の選択された分析物の量と相関する物理的に検出可能な変化を産 生させるための少なくとも1つの試薬と血漿の試料とを反応させるための試料処 理手段であって、ハウジングの中にあり、試料受容手段と流体伝達している試料 処理手段; を含む装置。 29. フィルターが第2の固相をさらに含み、固相支持体が第2の固相の中 に固定化されていて、それによって固相支持体上に固定化されている不溶性凝集 素が第2の固相の中にも固定化されている、請求の範囲第28項記載の装置。 30. 固相支持体が非多孔性粒子であり、第2の固相が多孔性物質である、 請求の範囲第29項記載の装置。 32. 固相支持体および固定化された凝集素が、約0.1から約0.25重 量%/容量の濃度で第2の固相に適用される、請求の範囲第29項記載の装置。 33. 凝集素が固相支持体上に約10:1の質量比を使用して固定化されて いる、請求の範囲第28項記載の装置。 34. フィルターが約20μLの全血から約10μLの血漿を形成する、請 求の範囲第28項記載の装置。 35. 血漿が3分以内に形成される、請求の範囲第34項記載の装置。 36. フィルターが全血の試料から約40容量%から約50容量%の血漿を 形成する、請求の範囲第28項記載の装置。 37. 固相支持体上に固定化された凝集素の量が、全血の試料中の赤血球の 数とほぼ同一数の結合部位を提供する、請求の範囲第28項記載の装置。 38. 全血の試料中における1以上の選択された分析物の存在を測定するた めの方法であって、 容器の外部表面上に全血の試料を導入する工程; 固相支持体、並びに赤血球のための凝集素であって全血試料中に不溶であり、 固相支持体上にさらに固定化されている凝集素で全血の試料を濾過し、それによ って選択された分析物を含む血漿の試料が形成される工程; 試料をハウジング内の反応領域に移す工程;そして 血漿試料中の対応する選択された分析物の量と相関する物理的に検出可能な変 化を産生させるための少なくとも1つの試薬と試料とを化学的に反応させる工程 : を含む方法。 39. 濾過工程が、凝集素を非多孔性粒子に化学的に結合させて結合体を形 成することを含む、請求の範囲第38項記載の方法 40. 固相支持体を全血試験の流動路の中に物理的に保持することをさらに 含む、請求の範囲第38項記載の方法。 41. 濾過工程が、約20μLの全血から約10μLの血漿を形成する、請 求の範囲第38項記載の方法。 42. 凝集素:赤血球の結合を切断してフィルターから赤血球を除去するこ とによって、赤血球に結合する凝集素の能力を再生する工程をさらに含む、請求 の範囲第38項記載の方法。 43. 濾過工程が、全血の試料から約40容量%から約50容量%の血漿を 形成する、請求の範囲第38項記載の方法。
