del brevetto per invenzione industriale avente titolo: ?Composizione di sostanze di tipo fenolico per uso medico e\o cosmetico?.
Campo tecnico dell?invenzione La presente invenzione si riferisce ad una formulazione di sostanze di tipo fenolico comprendenti: resveratrolo, catechine, epicatechine, procianidina B2, acido caffeico e quercitina.
Il campo tecnico individuato ? quello delle composizioni di sostanze di tipo fenolico, utilizzate in ambito medico e\o cosmetico, con spiccata attivit? trofica sulla pelle, anti-age e protettiva ed efficaci nella prevenzione e cura delle malattie cardiovascolari e neurodegenerative.
In particolare la presente invenzione concerne una composizione di sostanze di tipo fenolico ad uso topico, da utilizzare con additivi e\o veicoli farmaceuticamente tollerabili o che sono comunemente usati in prodotti cosmetici e/o dermatologici per curare patologie cutanee sostenendo e risollevando la pelle del viso, o da utilizzare con additivi e\o veicoli farmaceuticamente tollerabili o che sono comunemente usati in integratori alimentari per la somministrazione orale nella cura e prevenzione di patologie neurodegenerative e cardiovascolari.
Descrizione dello stato della tecnica Come ? noto, il resveratrolo (3,5,4'-triidrossitrans-stilbene) ? prodotto naturalmente da numerose piante in risposta agli attacchi da agenti patogeni quali batteri o funghi. Le principali fonti alimentari di resveratrolo sono la buccia dell'uva, i mirtilli, i lamponi, il gelso e le arachidi. L'azione antiossidante del resveratrolo ? comune alla classe chimica dei polifenoli a cui appartiene. Il resveratrolo si mostra efficace nelle patologie cutanee a eziologia multifattoriale, specie se presente una componente infiammatoria, per la sua capacit? di inibire la perossidazione lipidica, di chelare gli ioni metallici e di agire contro i radicali liberi (Si veda:

Le catechine sono forti antiossidanti. L'accumulo di radicali liberi di ossigeno provoca danni alla pelle attraverso la degradazione del collagene nella matrice extracellulare della cute. La Catechina previene l'accumulo di ROS indotto da TNF-? catturando i radicali liberi attraverso HO-1. La Catechina sopprime anche le citochine proinfiammatorie, tra cui l'interleuchina IL-1?e IL-6, che sovraregolano le reazioni infiammatorie e promuovono le reazioni legate all'invecchiamento, incluso l'invecchiamento cutaneo. Le catechine aumentano la penetrazione e l'assorbimento di cosmetici biologici nella pelle, migliorando cos? la loro utilit?. L'epicatechina((2R,3R-2-(3,4-dihydroxyphenyl)-3,4 dihydro-2H-chromene3,5,7-triolo) ? uno dei flavonoidi pi? abbondanti presenti in diversi frutti come mele, more, fave, ciliegie, uva, pere, lamponi, cacao e foglie di t?. La procianidina o proanticianidina B2, ? un tipo di proantocianidina contenuta in varie fonti vegetali che sta riscuotendo un enorme successo negli ultimi anni in svariarti ambiti salutistici. ? inoltre indiscussa la sua azione antiossidante e quindi benefica per l?azione di contrasto ai radicali liberi, molecole distruttive responsabili di invecchiamento in generale, capace di danneggiare le strutture cellulari (membrana, mitocondri) e il DNA. Le fonti pi? ricche per tale principio attivo sono la mela annurca campana IGP, l?uva rossa, l?orzo e molti altri. L'acido caffeico ? una sostanza di natura fenolica contenuta in molti alimenti di origine vegetale. Presenta propriet? antinfiammatorie, dovute alla loro capacit? di inibire le ciclossigenasi 1 e 2.si ? dimostrato utile anche nel proteggere la pelle dagli effetti dannosi delle radiazioni UV ed in particolare di quelle dotate della lunghezza d'onda pi? pericolosa (UVB ed UVC). Da uno studio nei topi ? emerso che l?acido caffeico dell?aglio applicato localmente sulla pelle dei topi ha prevenuto con successo la formazione di rughe indotta dall?irradiazione UVB ed ha anche inibito lo stress ossidativo. La quercetina dimostra di possedere diverse interessanti azioni fisiologiche sulla pelle. Ha una forte attivit? antiossidante. Protegge i cheratinociti dagli agenti ossidanti esogeni e elimina i radicali liberi, e inibisce la perossidazione lipidica dopo l'esposizione ai raggi UV. La quercetina presenta anche importanti azioni anti infiammatorie. La quercitina inibisce il NFKB e il rilascio di diverse citochine proinfiammatorie. Queste propriet? antiossidanti e antiinfiammatorie possono avere un ruolo importante nella terapia delle ulcere croniche. La quercetina ha azioni anti-invecchiamento sui cheratinociti e azioni ringiovanenti sulle cellule senescenti terminali. La quercetina dimostra di possedere diverse interessanti azioni fisiologiche sulla pelle. Essa ha una forte attivit? antiossidante; protegge i cheratinociti dagli agenti ossidanti esogeni e elimina i radicali liberi, previene l'esaurimento antiossidante endogeno e inibisce la perossidazione lipidica dopo l'esposizione ai raggi UV.
Nell?ambito delle applicazioni topiche le diverse sostanze nutraceutiche sono state utilizzate come singole molecole; tuttavia i complessi processi patofisiologici che regolano il metabolismo cellulare nell?invecchiamento e nella riparazione dei danni indotti dai fattori esterni richiedono la sinergia di diverse molecole.

Il documento dell?arte nota IT202000006493A rivendica un integratore alimentare adatto a contrastare la degenerazione maculare derivante da un estratto di vinacce ricco di polifenoli e contenente Resveratrolo, Acido Gallico, Catechina, Acido Ferulico, Quercetina, Procianidina B2. L?acido gallico presente in questa composizione pu? indurre uno stato allergico che ne preclude l?utilizzo nelle applicazioni topiche (si veda: Bai, J. et al. Gallic acid: Pharmacological activities and molecular mechanisms involved in inflammationrelated diseases. Biomed Pharmacother 133, 110985 (2021).
Difatti, nonostante i benefici mostrati dall?acido gallico rispetto all'invecchiamento della pelle, sono stati riportati alcuni casi di allergia cutanea negli esseri umani giustificando approcci volti a migliorare la biodisponibilit? e la biodegradabilit? di questo composto fenolico scarsamente solubile in acqua e non biodegradabile ( New Perspectives on the Efficacy of Gallic Acid in Cosmetics & Nanocosmeceuticals. Contrariamente, la presente invenzione non comporta alcun rischio nell?ambito delle applicazioni topiche e per l?assunzione orale. In questa formulazione innovativa le componenti sono state bilanciate al fine di generare la migliore azione antiossidante e di riduzione del processo di aging delle cellule (

L?attivit? di ricerca e gli studi effettuati hanno dimostrato che le sostanze rivendicate dalla presente invenzione, combinate tra loro in proporzione determinata, svolgono una particolare azione sinergica ed un innovativo potenziamento reciproco, non presentando profili di tossicit?, n? di allergenicit?, non essendovi nessuna incompatibilit? tra i componenti.
L?utilizzo combinato delle sostanze di cui alla presente invenzione produce, in modo innovativo, benefici nella cura delle patologie cutanee ovvero nella prevenzione e cura di malattie cardiovascolari e/o neurodegenerative.
Soluzione proposta dalla presente invenzione Scopo generale della presente invenzione ? quello di realizzare una composizione di sostanze fenoliche da utilizzare nell?ambito medico, ad esempio in ambito vulnologico, dermatologico, ginecologico e\o in ambito cosmetico, per la cura di differenti patologie cutanee, neurodegenerative e cardiovascolari. Scopo della presente invenzione ? quello di realizzare una composizione di sostanze di tipo fenolico ad azione nutriente, protettiva, antinfiammatoria e anti-age sulla pelle del viso.
Scopo della presente invenzione ? quello di realizzare una composizione di sostanze di tipo fenolico da utilizzare con additivi e\o veicoli farmaceuticamente tollerabili o che sono comunemente usati in ambito cosmetico e/o dermatologico per uso topico avendo gli effetti sopra descritti.
Scopo della presente invenzione ? quello di realizzare una composizione di sostanze di tipo fenolico da utilizzare, con additivi e\o veicoli farmaceuticamente accettabili o che sono comunemente usati in ambito medico, mediante somministrazione orale nella prevenzione e cura delle malattie cardiovascolari e neurodegenerative.
Ulteriore scopo della presente invenzione ? quello di fornire una composizione di sostanze di tipo fenolico che, stante l?assoluta compatibilit? tra i componenti, non presenti alcun profilo di allergicit? n? tossicit? e dunque possa essere utilizzata su qualsiasi soggetto.
In vista di questi scopi viene fornita una prima forma di realizzazione della presente invenzione in particolare:
- una composizione di sostanze di tipo fenolico per uso topico utilizzata con veicoli farmaceuticamente tollerabili comunemente usati in ambito cosmetico e dermatologico.
In una prima forma di utilizzo la composizione di sostanze oggetto della presente invenzione ? stata applicata sulla pelle del viso, immediatamente dopo la somministrazione transcutanea di PRP, mostrando innovativi effetti protettivi e di potenziamento degli effetti legati al PRP.
In particolare, la pelle del viso, infiammata in seguito alla somministrazione del PRP, gi? dopo la prima applicazione della composizione, veicolata attraverso una crema viso al 3%, risultava meno rossa, meno edematosa e pi? sana, rispetto al controllo. L'uso quotidiano della composizione per via cutanea, nei soggetti sottoposti a trattamento, ha evidenziato un potenziamento dei risultati estetici legati al PRP.
Il questionario: l?efficacia ? stata dimostrata utilizzando il Wrinkle Severity Rating Scale (WSRS)e il questionario di soddisfazione somministrato alle 30 utilizzatrici della crema di et? compresa tra 20 e 70 anni. Il grado di soddisfazione ? stato elevato per tutti i soggetti.

1.Composizione di sostanze di tipo fenolico per uso medico e\o cosmetico per l?impiego nella cura delle patologie cutanee e nella cura e prevenzione di malattie neurodegenerative e cardiovascolari comprendente resveratrolo, catechine, epicatechine, procianidine B2, acido caffeico e quercitina.
2. Composizione di sostanze secondo la rivendicazione 1 comprendente: Resveratrolo 700-800 gr/gr (10%), Catechine 3500-4000 gr/gr. (50%), Epicatechine 700-800 gr/gr. (10%), Procianidine B2 400-500 gr/gr. (6,6%), Quercitina 700-800 gr/gr. (10%), Acido caffeico 700-800 gr/gr (10%).
