Abstract

1431705 N-substituted benzomorphan derivatives BRISTOL-MYERS CO 26 Oct 1973 [26 Oct 1972] 50039/73 Heading C2C [Also in Division A5] Novel compounds L: (in which R<SP>1</SP> is CH#C-CH 2 -; CH 2 =CH-CH 2 -, (CH 3 ) 2 C=CH-CH 2 -, ClCH=CH-CH 2 - or a radical of formula in which R<SP>6</SP> is H or CH 3 ; R<SP>2</SP> is H, 1-6C alkyl, 2-6C acyl, 3- or 4-pyridyl-carbonyl, N-oxidopyridyl-3-carbonyl, #-morpholinoethyl, methoxymethyl, benzylcarbonyl, orN-oxido-pyridyl- 4-carbonyl; R<SP>5</SP> is H, 2-6C acyl, trichloroacetyl or cinnamoyl; R<SP>3</SP> is H, CH 3 , C 2 H 5 , n-C 3 H 7 , allyl or propargyl; and R<SP>4</SP> is 1-6C alkyl, 3-7C alkenyl or alkynyl or a radical in which n is 1-6 and R<SP>10</SP> is H, Cl, Br, F, NO 2 , 1-6C alkyl or alkoxy or NH 2 ) and salts thereof are prepared by (A) reacting a compound XXXXIII: (in which R<SP>7</SP> is 1-6 alkyl) with a compound (in which X is Cl, Br or iodo, Z is CO or CH 2 and W is as defined for R<SP>1</SP> above but in which the alpha-methylene group is missing) in the presence of a base to give a compound XXXXIV: (B) selectively reducing the carbonyl group of compound XXXXIV if Z is CO to produce a compound XXXXV: and when it is desired to produce compounds L in which R<SP>2</SP> is other than R<SP>7</SP> (C) cleaving the ether function of compound XXXXV to give a compound L where R<SP>2</SP> is H and, if desired, converting, by known methods, the compound L where R<SP>2</SP> is H to a compound L where R<SP>2</SP> is other than H.
