1315566 Amidino-penicillanic acid derivatives LOVENS KEMISKE FABRIK PRODUKTIONSAKTIES-ELSKAB 5 Nov 1971 [6 Nov 1970 7 May 1971] 53026/70 and 51629/71 Heading C2A The invention comprises amidino-penicillanic acid derivative of the general Formula (I) in which R 1 R 2 and R 3 each represent an aliphatic hydrocarbon radical, a mono- or bicyclic aryl radical, an aralkyl radical, a cycloalkyl radical, a cycloalkyl-alkyl radical, a heterocyclic radical or a heterocyclically substituted alkyl radical; R 1 and R 2 when taken together with the nitrogen atom may represent a ring system; R 1 , R 2 and R 3 are optionally substituted; R 4 represents a hydrogen atom or an unsubstituted or substituted alkyl or aralkyl radical; and pharmaceutically acceptable salts thereof. R 1 , R 2 and R 3 may be substituted with one or more halogen atoms or alkyl, hydroxy, alkoxy, alkylthio, acyl, carboxy, carbalkoxy, carbamyl, carbamido, cyano, sulphonyl, amino or substituted amino radicals, carbocyclic aryloxy, carbocyclic arylthio, or azido radicals. A process for the preparation of the compounds of general Formula I comprises reacting a reactive derivative of an amide or thioamide of general Formula (II) where R 5 is oxygen or sulphur with a 6-aminopenicillanic acid derivative of general Formula (III) in which COOR 4 is an ester group or with a silyl ester of 6-aminopenicillanic acid, whereafter, if desired, the reaction is followed by a cleavage of the ester to form the free acid. In a further process for the preparation of compounds of Formula I, a silyl ester of 6-aminopenicillanic acid is reacted with an amide acetal of Formula (IIa) wherein R 6 is an alkyl radical, whereafter the silyl ester of the reaction product is hydrolysed or alcoholysed to form the compounds of Formula (I) where R 4 is a hydrogen atom. Pharmaceutical compositions containing at least one of the compounds of Formula (I) as active ingredient may be prepared and may be adapted for oral or parenteral administration. [GB1315566A]
