Claims (14)
1. Postupak za proizvodnju etanola koji obuhvaća fermentaciju lignoceluloznog hidrolizata u šaržno napajanom reaktoru u šaržnom načinu rada sa stanicom kvasca, koja se sposobna za pretvorbu šećera, glicerola i octene kiseline, koji obuhvaća sljedeće korake:
a) inokuliranje razrijeđenog lignoceluloznog hidrolizata sa stanicom kvasca;
c) dodavanje lignoceluloznog hidrolizata koji sadrži glicerol u reaktor;
d) nakon dovoljnog vremena fermentacije, izoliranje etanola iz reaktora,
e) po izboru zadržavanje preostale frakcije nakon izolacije iz koraka d) kao istrošene juhe; i
f) po izboru korištenje istrošene juhe u koraku a) za razrjeđivanje lignoceluloznog hidrolizata, pri čemu se pH smjese u šaržno napajanom reaktoru u šaržnom načinu rada održava konstantnim dodavanjem dovoljne količine lignoceluloznog hidrolizata.
2. Postupak u skladu s patentnim zahtjevom 1, naznačen time što korak c) obuhvaća postupno dodavanje u reaktor lignoceluloznog hidrolizata dopunjenog glicerolom.
3. Postupak u skladu s patentnim zahtjevom 1 ili zahtjevom 2, naznačen time što je dodana količina glicerola takva da je molarna koncentracija glicerola u reaktoru dvostruko veća od molarne koncentracije ili 1,8 do 2,2 puta veća od molarne koncentracije octene kiseline u reaktoru.
4. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 3, naznačen time što dodavanje glicerola započinje kada je koncentracija glukoze u reaktoru 2 g/l ili niža.
5. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 4, naznačen time što je stanica kvasca sposobna trošiti ksilozu u lignoceluloznom hidrolizatu.
6. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 5, naznačen time što je stanica kvasca genetski modificirana, koja obuhvaća:
a) jednu ili više nukleotidnih sekvenci koje kodiraju heterolognu NAD+- ovisnu acetilacijsku acetaldehid dehidrogenazu (E.C. 1.2.1.10);
b) jednu ili više nukleotidnih sekvenci koje kodiraju homolognu ili heterolognu acetil-CoA sintetazu (E.C. 6.2.1.1);
c) jednu ili više nukleotidnih sekvenci koje kodiraju heterolognu glicerol dehidrogenazu (E.C. 1.1.1.6); i
d) jednu ili više nukleotidnih sekvenci koje kodiraju homolognu ili heterolognu dihidroksiaceton kinazu (E.C. 2.7.1.28 ili E.C. 2.7.1.29).
7. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 6, naznačen time što stanica kvasca ima deleciju ili poremećaj jedne ili više endogenih nukleotidnih sekvenci koje kodiraju glicerol 3-fosfat fosfohidrolazu i/ili kodiraju gen glicerol 3-fosfat dehidrogenaze.
8. Postupak u skladu s patentnim zahtjevom 6 ili 7, naznačen time što stanica kvasca eksprimira egzogeni simporter glicerola.
9. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 8, naznačen time što se tijekom postupka u smjesu u reaktoru ne dodaje baza.
10. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 9, naznačen time što je stanica kvasca sposobna metabolizirati organsku kiselinu.
11. Postupak u skladu s patentnim zahtjevom 10, naznačen time što je stanica kvasca sposobna metabolizirati octenu kiselinu.
12. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 11, naznačen time što je postupak anaerobni ili anoksični šaržni postupak fermentacije u kojem je pH fermentacijske juhe u reaktoru viši od pH dovedene sirovine.
13. Postupak u skladu s bilo kojim od patentnih zahtjeva 7 to 12, naznačen time što stanica kvasca može anaerobno fermentirati najmanje jedan C6 šećer i najmanje jedan C5 šećer.
14. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 13, naznačen time što se postupku fermentacije dodaje postupak razmnožavanja.
