Abstract

The invention comprises 5b -pregnanes having the general formula <FORM:0988805/C2/1> where either R1 is a hydrogen atom, in which case R2 is the group (H, b -OH) and R3 and R4 either each is a hydrogen atom or together are the group >C.R5R6, or R1 is an acyl group derived from a lower aliphatic carboxylic acid having 2-7 carbon atoms, in which case R2 is either the group (H1 b -OH) or the oxygen atom of a keto group, and R3 and R4 together are the group >C.R5R6,R5 and R6, which may be the same or different, each represents an alkyl group having 1-4 carbon atoms, and processes for the preparation thereof which comprise treating a 3a -hydroxy-20-carboalkoxy-5b - pregnan - 11 - on - 21 - oic acid alkyl ester having the general formula <FORM:0988805/C2/2> (wherein R7 and R8 are the same or different and are each an alkyl group having 1-6 carbon atoms) with a mixed metal hydride so as to simultaneously reduce the 11-keto group and to both reduce and saponify the carboalkoxy groupings attached at the 20-position, thus yielding 3a ,11b ,21 - trihydroxy - 20 - hydroxymethylene - 5b - pregnane, reacting the latter with a lower aliphatic ketone in the presence of a dehydrating agent to yield the corresponding ketonide of 3a ,11b ,21 - trihydroxy - 20 - hydroxymethylene - 5b - pregnane, reacting the latter with an acylating agent to yield the corresponding ketonide of a 3a -acyloxy-11b ,21-dihydroxy - 20 - hydroxymethylene - 5b -pregnane, oxidizing the latter with a mixture of sulphuric and chromic acid to obtain the corresponding ketonide of a 3a -acyloxy-21-hydroxy-20 - hydroxymethylene - 5b - pregnan - 11 - one and subjecting the latter to hydrolysis to obtain 3a - acyloxy - 21 - hydroxy - 20 - hydroxymethylene - 5b - pregnan - 11 - one.
