Abstract

Compounds of the general formula <FORM:0974891/C1/1> in which R1 and R2 are alkyl groups and R3 is an alkyl or alkenyl group are prepared by a process in which a compound of the general formula <FORM:0974891/C1/2> is reduced with the aid of a metal hydride reducing agent. The starting material may be griseofulvin or a compound of the second formula above prepared in any convenient way, e.g. by etherification of non-griseofulvic acid followed if necessary by selective hydrolysis and re-etherification with a further alkylating agent, or by reacting a 4-methoxy grisan such as griseofulvin with anhydrous magnesium iodide to produce the corresponding 4-hydroxy grisan which may then be etherified with an appropriate alkylating agent. The reducing agent may be sodium, potassium or calcium bromohydride or lithium aluminium hydride, the reaction being preferably carried out in an inert solvent. The product may be converted into an ester thereof with an inorganic or organic acid, esters specified being the acetates, hemisuccinates and hemiphthalates. Pharmaceutical preparations comprise compounds of the first formula above or esters thereof together with a pharmaceutical carrier or diluent in forms such as tablets, pills, capsules, granules, powders, oral liquid preparations of aqueous or oily nature, suppositories, or as solutions or dispersions in sterile parentally acceptable liquids.
