The invention comprises compounds of the general formula: and their non-toxic acid addition salts where R represents a C 1-4 alkyl group and the methyl group in the 3-position may have the α- or #- configuration and the preparation thereof either by reacting a compound of the formula: where X represents a halogen atom with a compound of the formula: or by dehydration of a compound of the formula: The process of the invention may be combined with one wherein the compound IV is prepared either by reacting a compound of the formula: with a compound III or by reduction of a compound of the formula: which in turn may be prepared by condensing formaldehyde, 2-acetyl-thiophene and a compound III whereby a Mannich reaction takes place. Piperidines of formula III are prepared by reacting a #-haloethyl- #-halopropyl-N-tosyl amine with benzyl cyanide followed by saponification and esterification of the compound thus obtained. Pharmaceutical compositions comprise the compounds I or their salts together with a pharmaceutical carrier or excipient. The compositions, which have analgesic properties, may be in the form of capsules, tablets, granules, coated tablets, emulsions, syrups, suspensions, drops or suppositories.
