Abstract

An antitrachoma vaccine is prepared by inoculating a chicken egg with trachomatous material, culturing, suspending the infected yolk sac membranes in an aqueous trypsin suspension in aqueous phosphate buffer solution at pH 7.5 to 8 and 35 DEG to 40 DEG C. to free the virus from the protein. The viral suspension is separated from the protein fraction and further treated with silicious material to absorb impurities. It is then separated from the silicious material and finally treated with a fat solvent such as diethyl ether. The separation of the virus may be by differential centrifuging. The final virus suspension may be used as such or in admixture with aluminium hydroxide or mineral oils.
