Abstract

The sodium derivative of Warfarin (3-[a -acetonylbenzyl]-4-hydroxy coumarin) (a) is prepared by slowly adding an aqueous sodium hydroxide solution to an agitated slurry of warfarin containing excess warfarin in water containing about 10% by volume of acetone, and (b) is purified by dissolving the alkali metal derivative in warm isopropyl alcohol, stirring the resulting solution while it is cooling to room temperature and recovering the resulting precipitate of crystalline warfarin. sodium. The latter contains isopropyl alcohol which may be removed either by heating to ca. 145 DEG C. at less than one mm. pressure, or by solution in absolute ethanol followed by concentration and drying under reduced pressure at temperatures up to about 50 DEG C. Detailed examples illustrate the various processes.ALSO:Warfarin sodium, having rodenticidal and anticoagulatory activity, is purified and can be used in solution or in tablet form, and may be administered in combination with heparin sodium.
