Abstract
The invention comprises 4-chloro-testosterone cyclopentylpropionate and a process for the preparation of 4-chloro-3-keto-D 4-steroids by reacting 3-keto-D 4-steroids with chlorine in an organic solvent which is inert to chlorine such as carbon tetrachloride, preferably in the presence of a tertiary organic base such as pyridine. The chlorine may be provided by a substance which releases chlorine under the reaction conditions, e.g. by means of acetic acid as the reaction solvent and t-butyl-hypochlorite. The preferred reactants for the above process have the general formula: <FORM:0864609/IV (b)/1> wherein R represents =(H)OR111, =O, =(CH3)OH, =(C2H5)OH, =(H)COCH3, =(H)OCOCH3, =(H)COCH2OCOCH3, or =(OH)COCH2OCOCH3, R1 represents =(H)H, =(H)OH or =O, R11 represents -H or -CH3 and R111 represents an acyl group. In the examples 4-chloro-testosterone acetate, 4-chloro-testosterone b -cyclopentylpropionate, 4-chloro-19-nortestosterone acetate, 4-chloroprogesterone, and 4-chloro-desoxycorticosterone acetate are prepared. Specifications 838,773, 864,608 and 864,613 are referred to. Reference has been directed by the Comptroller to Specification 792,727.ALSO:A pharmaceutical composition contains 4-chloro-testosterone cyclopentylpropionate and a compatible solid or liquid carrier therefor.
