Abstract

An ACTH preparation comprises a substantially water-insoluble complex of ACTH and an electrolytically prepared alpha-zinc hydroxide or basic zinc salt. The preparation is for subcutaneous or intramuscular injection and is characterized by its ease of suspension and resistance to breakdown when subjected to alternate freezing and thawing. The alphazinc hydroxide may be prepared in an electrolytic cell having a zinc anode, a platinum or other inert cathode, e.g. silver, gold or graphite, and an electrolyte which may have an initial pH from 2 to 5, containing aqueous sodium chloride or other pharmaceutically acceptable electrolyte, which may have a normality of 0.01 to 2.0 N. The ACTH may be present in amount of 20 to 80 u.s.p. units for each 0.2 to 3.0 mg. of elemental zinc, and may be added to the zinc compound within a few minutes of the preparation thereof, or may be present in the cell when the zinc compound is formed. Other compatible materials such as preservatives, e.g. phenol or benzyl alcohol, isotomicity producing substances, e.g. sodium chloride, glycerol, buffers or local an thetics, e.g. procaine hydrochloride.
