Abstract

A process for extracting a leucocytosis factor from aqueous liver extracts comprises adjusting the aqueous extract to pH5 to 10 with alkali, forming a precipitate containing the active factor by the addition of barium, calcium or lead acetate and alcohol, separating this precipitate and dissolving in water, thereafter treating the solution with an acid ion-exchanger whereby metals and impurities are absorbed, the solution containing the active factor is then treated with a basic ion-exchanger, which is then eluted with an alkaline solution to liberate the absorbed factor and evaporating and drying the eluate to produce an active factor in concentrated form. As a modification of the process, the precipitate formed by alcohol and acetate may be treated with dilute sulphuric acid to remove the metal ions by the formation of insoluble sulphates which are removed and the solution then treated with the basic ion-exchanger as described above. The active factor may be further purified by electrophoresis in which the factor is dissolved in water, an alkaline buffer added and the active factor migrates towards the anode. The active substance which is obtained by freeze-drying the eluate from the basic ion-exchanger is yellowish-brown, a 1 per cent solution of which gives an extinction E1 per cent 1 cm.=30 at a wavelength of 260 millimicrons. The factor was found to contain 1 per cent phosphorus and has an acid reaction.
