Abstract
The invention comprises the 3,5-dinitrobenzoate of monoepoxycalciferol having a m.p. of 148-149 DEG C., monoepoxycalciferol having a m.p. of 109-110 DEG C. and a process for the production of the 3,5-dinitrobenzoate of monoepoxyprecalciferol, characterized in that precalciferol dinitrobenzoate dissolved in a solvent therefore which is immiscible with water, e.g. ether, is oxidized with an organic peracid, e.g. perphthalic acid at a temperature comprised between - 10 DEG and + 10 DEG C. so as to produce the 3,5-dinitrobenzoate of monoepoxyprecalciferol. Preferably the starting material is dissolved in ether and the reaction effected with perphthalic acid between - 5 DEG to + 5 DEG C. Subsequently the 3,5-dinitrobenzoate may be saponified in an inert solvent, e.g. ether with a solution of potassium hydroxide in methanol to obtain the 3,5-dinitrobenzoate of monoepoxyprecalciferol, or hydrolysed using aqueous sulphuric acid to obtain the 3,5-dinitrobenzoate of dihydroxy-dihydroprecalciferol which in turn yields dihydroxy - dihydroprecalciferol with potassium hydroxide in methanol. In an example, the production of monoepoxyprecalciferol and the 3,5-dinitrobenzoate are described and also the subsequent conversion of the 3,5-dinitrobenzoate to the dinitrobenzoate of dihydroxy-dihydroprecalciferol and to dihydroxy - dihydroprecalciferol. Specification 668,114 is referred to.
