Abstract

Tetracycline is prepared by treating bromtetracycline in an inert organic solvent with hydrogen in the presence of a catalyst consisting of palladium or platinum or mixtures thereof. Tetracycline hydrobromide is produced and may be recovered after filtering off the catalyst by evaporating the reaction solution to dryness or by adding a non-solvent to precipitate the tetracycline hydrobromide. If tetracycline base is desired, or if the hydrochloride or hydrobromide of bromtetracycline is the substance hydrogenated, the reaction is carried out in the presence of a tertiary amine base such as triethylamine or trimethylamine. Specified solvents are aliphatic alcohols such as methanol and ethanol containing not more than eight carbon atoms, dioxane, methoxyethanol, ethoxyethanol or mixtures thereof. The reaction is carried out at 10 DEG to 50 DEG C. and preferably 25 DEG to 35 DEG C. and at pressures of slightly above atmospheric or of 3 to 5 atmospheres.
