Abstract

The invention comprises 3b ,11a ,21-triesters of 3b ,11a ,17a ,21 - tetrahydroxy - pregnane - 20-one and the preparation thereof by subjecting a 3b ,11a ,21-triacyloxy-pregnane-17-yl-cyanide-20 or a mixture of isomers thereof to hydroxylation by an oxidizing agent, e.g. osmium tetroxide or potassium permanganate, followed if necessary by reductive splitting of the oxidation product, e.g. with sodium sulphite, to obtain a 3b ,11a ,21-triacyloxy - 17a - hydroxypregnane - 20 - one, which may be saponified or re-esterified, e.g. to the benzoyloxy ester, trifluoroacetic ester, and the carbomethoxy, carboethoxy and carbobenzoxy compounds. The 3b ,11a ,21 - triacyloxy - pregnene - 17 - yl - cyanide - 20 or a mixture of isomers thereof may be prepared from a triester of 3b ,11a ,21 - trihydroxy - pregnane - 20-one by the addition of hydrocyanic acid and splitting off of water. An example describes the preparation of 3b ,11a ,17a -triacetoxy-17-hydroxy - pregnane - 20 - one. Specifications 653,480 and 768,123 are referred to.
