Abstract
A stable suspension for oral therapeutic use comprises a therapeutic agent and coconut oil which has been chemically modified by ester exchange, inter-esterification and/or acidolysis to reduce the setting point below 60 DEG F. (15,5 DEG C.). Up to 20 per cent of a mineral oil, edible oil or injectable oil such as butyl stearate or isopropyl myristate may be present. Other optional additions are (a) surface active agents such as lecithin and polyoxyalkylenesorbitols, (b) fillers such as aluminium hydroxide, talc, magnesium trisilicate, kaolin and calcium carbonate, (c) demulcents such as potassium alginate (d) colours, flavours, sweetening agents such as sugar and saccharin and preservatives. The drugs are preferably milled or micronized. Among those specified are (a) antibiotics such as tetracycline, oxytetracycline, chlortetracycline, streptomycin; penicillin, which may be coated with gums, methyl cellulose, polyvinyl pyrrolidone or sodium carboxymethyl cellulose; chloramphenical, polymycin, neomycin and hacitracin; (b) vitamins, (c) sulphonamides such as sulphapyridine, sulphathiazole, sulphadiazine and sulphamethazine; (d) antihistaminics such as N, N-dimethyl-N-1 benzyl-N1-phenyl ethylene diamine, (e) stimulants such as caffeine and amphetamine, (f) sympathomimetric and adrenergic drugs, e.g. edhedrine and 2-aminohexane, (g) analgesics such as asprin, salicylamide and phenacetin, (h) laxatives, e.g. phenolphthalein, (i) sedatives such as amytal (j) anticonvulsants such as phenobarbitone, (k) antimalarials such as atabrine, (l) anticoagulants such as heparin, (m) hormones such as cortisone, (n) minerals such as salts of iron, copper, zinc and calcium, (o) alkaloids, (p) contrast media such as barium sulphate. The following synergistic combinations are specified (1) penicillin and antihistamine, (2) aspirin and codeine, (3) tetracycline and chlortetracycline, (4) sulphadiazine and sulphamerazine, (5) streptomycin sulphate, levo-primaric-6, 8a-cisendo-succinate and dihydrostreptomycin sulphate, (6) asprin, phenacetin and caffeine, (7) sodium penicillin and procaine penicillin. Specification 665,367 is referred to.
