Abstract
Alkaloidal extracts of Rauwolfia species are treated with a mixture of halogenated aliphatic hydrocarbon and an aqueous acid of pH below 2.5, the two phases separated and the alkaloids recovered therefrom. The starting material can be made by treating the ground wetted plants with benzene and concentrating the resulting extract. Preferred solvents for the separation are chloroform and aqueous orthophosphoric acid of pH 0.6-1.5. Reserpine can be obtained from the chloroform phase by washing with aqueous alkali, evaporating to dryness and recrystallizing from a lower alcohol such as methanol. Examples show the separation of reserpine from R. serpentine and R. micrantha, and it is stated that R. heterophylla, R. vomitoria, R. perakensis and R. hirsuta may also be used. Other suitable solvents are carbon tetrachloride, ethylene dichloride, trichlorethylene and tetrachlorethylene on the one hand and hydrochloric, sulphuric and acetic acids on the other.
