Abstract
An antibiotic, desacetylthiolutin, is produced by reacting thiolutin with an inorganic acid having an ionization constant of at least 10-2 in a two-phase organic solvent-aqueous system, preferably at 50 DEG to 100 DEG C. It is preferred to treat thiolutin in refluxing dioxane with concentrated hydrochloric acid. The desacetylthiolutin, which separates, is dissolved in dilute hydrochloric acid, decolorized with activated carbon and crystallized. Desacetyl thiolutin monochloride monohydrate displays ultraviolet absorption maxima at 226, 309 and 381 mm ; has a pKa value of 2.9 and formula C6H6ON2S2. Desacetylthiolutin base is obtained from the hydrochloride by dissolving the hydrochloride in (a) water and adding a base such as ammonia, pyridine or sodium hydroxide; or (b) chloroform and treating with anhydrous ammonia. Thiolutin is reformed by treating an aqueous or organic solution of desacetylthiolutin with an acetylating agent such as acetyl chloride, acetic anhydride, ketene or acetylbromide. A propionylating agent such as propionyl chloride, propionyl bromide or propionic anhydride converts desacetylthiolutin to aureothricin. Desacetylthiolutin forms salts with acids such as hydrobromic, phosphoric, sulphuric, tartaric and citric. Specification 692,066 is referred to.
