Abstract

Oxytetracycline (terramycin) is purified or isolated from a fermentation broth as a metal complex, which may then be converted into any desired therapeutic form of oxytetracycline, by adding to an aqueous solution of pH 7.0 to 10.0 at least one of the metals barium, strontium and calcium, and at least one of the metals magnesium, zinc, beryllium, cadmium and mercury. Specified complexes are of calcium-magnesium, barium - magnesium and barium - zinc; the barium-magnesium complex having the lowest solubility. In the case of a fermentation broth, which normally contains sufficient calcium and magnesium ions, it is preferred to adjust the pH of the filtered broth to 8.0 to 9.0 and to add barium ions, e.g. barium chloride. Iron, chromium and copper impurities are removed by dissolving the complex in dilute acid, e.g. sulphuric acid, adding a soluble ferrocyanide, e.g. potassium ferrocyanide, to precipitate the iron &c. and removing excess ferrocyanide by the addition of a soluble zinc or zirconium salt. Oxytetracycline is obtained by adjusting the filtrate to pH 7.0 with alkali. The precipitate is dissolved in methanol and concentrated hydrochloric acid added to produce the hydrochloride, which may be given a further purification with activated carbon.
