Abstract
Hydrocortisone is obtained by saponifying hydrocortisone acetate by mixing a substantially oxygen-free solution of 11b , 17a -dihydroxy - 21 - acetoxy - 4 - pregnene - 3,20-dione in a lower alkanol with an aqueous substantially oxygen-free solution of at least a molar equivalent, calculated on the hydrocortisone acetate, of an alkali-metal bicarbonate and containing the reaction at a temperature between about 15 DEG to 30 DEG C. while protecting the mixture from atmospheric oxygen, and thereafter neutralizing the saponified mixture with an aqueous solution of an acid, e.g. acetic acid, and isolating the thus-produced hydrocortisone. As alkali metal bicarbonates there may be used sodium potassium or lithium bicarbonate. Preferably neutralization is effected below 30 DEG C.
