Abstract

Nitrogenous bases of the morphine series are extracted from an aqueous phase by means of an organic phase which is a mixture of (1) phenol, an alkylphenol or phenylalkanol containing not more than six carbon atoms in the aliphatic chain; (2) a C3-C6 aliphatic alcohol or an ester derived from a C1-C4 fatty acid and a C1-C6 aliphatic alcohol (organic phase lighter than aqueous) or a C6-C9 aromatic nitro compound or C1-C6 aliphatic nitro compound (organic phase heavier than aqueous); and (3) a diluent miscible with (1) and (2) of S.G. less than 0.9 or greater than 1.2 according to the solvent used for (2), the diluent being in such proportion that the two phases differ in S.G. by 0.10-0.28. The first liquid has a high solvent power for the bases and can be phenol, an alkylphenol, benzyl alcohol or phenethyl alcohol. The second liquid reduces the total solvent power but facilitates phase separation; it may be propanol, isopropanol, a butanol, amyl alcohol, isoamyl alcohol, ethyl acetate, propyl acetate or isopropyl acetate. Specified diluents are petroleum ether, benzene, toluene, xylene, cyclohexane, methyl isobutyl ketone, di-isopropyl ketone, di-isopropyl ether, di-butyl ether, ethylene dichloride, trichlorethylene, chloroform and carbon tetrachloride. Examples are given of the extraction of (1) morphine using cresol +n-butanol+cyclohexane; (2) dihydromorphinone using benzyl alcohol+ethyl acetate +benzene; and (3) morphine using cresol+ nitrobenzene+ethylene dichloride. The process is also applicable to the extraction of dihydromorphine, pseudomorphine, nor - morphine and N-alkyl and N-alkenyl derivatives thereof. The bases are separated from the organic phase by concentration and precipitation with alkali or by extraction with aqueous acid, preferably one of those used in Specification 750,360, e.g. oxalic acid. According to the Provisional Specification the process is applicable to nitrogenous bases of the 3-hydroxyhydrophenanthrene series in general.
