Abstract

Crude Vitamin B12 in aqueous solution, e.g. a Streptomyces griseus culture broth elaboration product, is purified by reacting with a substance yielding cyanide ions, e.g. hydrocyanic acid gas, potassium, sodium, ammonium, calcium, or barium cyanide, the purity of the Vitamin B12 being not in excess of about 1 per cent, the concentration of vitamin B12 in the solution being not in excess of about 100 micrograms/ml., the concentration of cyanide ions in the solution being in the range of about 0.2 to 0.6 gram-mols. per litre, and the pH of the solution being not less than 6, extracting the Vitamin B12-cyanide complex so formed into normal butyl alcohol in the presence of a salting-out agent, e.g. sodium sulphate, ammonium sulphate or magnesium sulphate, thereafter decomposing the Vitamin B12-cyanide complex by adjusting the pH value of the solution to an acid value and recovering the Vitamin B12. The normal butyl alcohol and the salting-out agent may be introducing during or after the formation of the Vitamin B12-cyanide complex. The extraction into normal butyl alcohol may be effected in a continuous manner. Specification 727,146 is referred to.
