Abstract

Streptomycin is produced by propagating Streptomyces griseus under submerged aerated conditions in an aqueous nutrient medium containing soya bean meal, distillers' solubles and an assimilable carbohydrate, with or without other complex nitrogenous organic materials. The soyabean meal may be present to the extent of 1-5 per cent (preferably 1-2 per cent) and the distillers' solubles 0.1-2 per cent (preferably 1.0-1.5 per cent), expressed as grams per 100 millilitres. The carbohydrate, e.g. dextrose or hydrol (by-product from hydrolysis of starch) may be present in amounts up to 4 per cent (preferably 0.75-2 per cent). The medium may also contain sodium chloride up to 3 per cent, e.g. 0.1-1.0 per cent. The medium is sterilized, inoculated with S. griseus (spores or vegetative inoculum) and fermented. The streptomycin is recovered by adsorption from the culture broth with activated charcoal and elution, e.g. with low-normality alcoholic hydrochloric acid.
