Abstract
An anti-an mia liver preparation is obtained by subjecting to the action of Papain an aqueous solution prepared from liver, said solution containing the water-soluble constituents of liver, but being free from the water-insoluble constituents. Preferably, minced liver is mixed with cold water acidified to a pH of 5.6, the mixture is heated to coagulate heat-coagulatable material, the aqueous extract is separated and is treated with Papain at a temperature of 60 DEG C. for, say, two hours, and the extract is then boiled to destroy the Papain. The Papain employed may be previously activated with sodium sulphide. The resulting preparation may be dried or concentrated (e.g. in vacuo) or fractionated by the following procedures: (1) precipitation with Reinecke acid and subsequent decomposition of the precipitate; (2) adsorption on charcoal followed by subsequent extraction of the adsorbate with 50-60 per cent alcohol; (3) precipitation by ammonium sulphate; (4) solution in 70-80 per cent alcohol; (5) solution in phenolic solvents such as cresylic acid; (6) rejection of the precipitate produced by the addition of lead acetate; (7) precipitation by phosphotungstic acid and subsequent regeneration of the precipitate. The product as well as the anti-an mia factor contains vitamin B1, nicotinic acid, pyridoxin, pantothenic acid and folic acid. Specification 565,986 is referred to.
