Abstract

1513806 Centrifuging blood CORNING GLASS WORKS 9 Oct 1975 [7 Nov 1974] 41407/75 Heading B2P In a method of separating blood into lighter and heavier phases, a vessel is provided with a quantity of coagulation-enhancing particles of a material selected from a group of siliceous substances including glass, kaolin, bentonite, hydrated aluminium silicate and diatomaceous silica, and is supplied with blood the particles having a size and density such that they are readily suspended throughout the blood without external agitation and remain so suspended until coagulation of the blood is complete and the mixture of blood and particles is centrifuged to separate the mixture into a lighter phase consisting of serum and a heavier phase consisting of cellular and fibrillar matter and the coagulation-enhancing particles. The vessel may be in the form of a glass of plastics tube fitted with a pierceable closure through which blood may be siphoned into the tube by venipuncture with a double-ended needle. The coagulation-enhancing particles 20, Fig. 4, may be applied to the interior surface 34 of a cavity 33 within a member 32 which is initially placed in separating gel 30 positioned at the closed end of the tube. As blood is supplied to the tube the particles 20 become suspended in it and upon application of centrifugal force, are precipitated with the heavier phase toward the closed end of the vessel and away from the lighter phase which is moved toward the open end. The gel 30 is forced upwardly by the member 32 to partition the separated lighter and heavier phases. The coagulation-enhancing particles may be prepared by grinding into a finely divided powder glass having a density greater than that of the serum phase of the blood; mixing the powder with a liquid having a density not greater than that of whole blood; allowing the mixture to stand so as to sediment the heavier particles and separating the latter from the remaining mixture; centrifuging the remaining mixture under a force greater in magnitude and duration than that used in the normal centrifugation of blood to precipitate particle therefrom; and removing and drying the precipitated particles.
