Description
The invention relates to a method of making coagulation activating particles for use in a process for coagulating and separating blood into its components.
J. P. Soulier und O. Prou-Wartelle, in British Journal of Heamatology, Band 6, S. 88-101.As is known, blood coagulation in vitro and the enzymatic conversions that take place can occur through Contact with substances containing silica such as glass, kaolin, bentonite, hydrated aluminum silicate, diatomaceous earth or kieselguhr are promoted; see J. P. Soulier and O. Prou-Wartelle, in British Journal of Heamatology, Volume 6, pp. 88-101.
That is to say, coagulation-activating particles which can be produced according to the invention are for example for use with the one from the publication Clinical Lab. Products, September 1974, pages 36 and 37 known methods intended for the coagulation and separation of blood into its components.
The use of plastic tubes has already been proposed to stimulate coagulation contain individual glass or kaolin particles and are moved vigorously. This mixing for coagulation prior to centrifugation as part of the process for coagulating and separating blood into its components however, it is quite cumbersome and unfavorable for the course of the procedure.
The object of the invention is the simple production of coagulation-activating particles in a process-favorable manner Use in the process mentioned at the beginning.
Erfindungsgemäß wird diese Aufgabe durch die im Kennzeichen des Patentanspruchs 1 genannten MerkAccording to the invention, this object is achieved by the features mentioned in the characterizing part of claim 1
Preferred further embodiments of the invention Process are described in the subordinate claims.
The inventive design of the method allows problem-free and without great effort Produce coagulation-activating particles, their use in the above-mentioned method leads to a substantial acceleration of the process flow. It is now superfluous as an introduction the blood coagulation to stir the contents of a vessel or to shake a vessel or to move it into a horizontal position because the particles go into suspension by themselves and such a rapid coagulation and transformation of the total fibrinogen content in the blood sample to fibrin cause this conversion at the start of centrifugation has been completed and in the separate serum phase there is no disturbance to the analysis of the serum phase or even damaging fibrin formation takes place.
The density of the coagulation-activating particles is greater than that of the serum phase, and their size is so small that they are evenly suspended in the blood when blood is poured into or drawn into a container and remain in suspension without stirring at least during the onset of coagulation. On the other hand, the particles are so large and dense that they can be precipitated by centrifugation without difficulty, e.g. B. in the case of a borosilicate glass with a density in the range of 2.16-2.43 g / cm 3 . By choosing the size, centrifugation that is too vigorous or too long can be avoided, which can lead to hemolysis of the red blood cells and increase the proportion of milk dehydrogenase, potassium or hemoglobin in the serum.
The maximum size of the particles is expediently chosen so that they remain in the first 2-5 minutes of coagulation remain in suspension without stirring.
A fine powder that remains suspended and in the powder that is removed from the walls of the vessel is particularly beneficial Share its activating effect, or a larger total surface area is available for activation but at the same time less strongly or frequently during centrifugation because of the low particle mass collides with red blood cells. With fine powder, the risk of hemolysis is also not so great.
Sedimentation von der je-To produce the coagulation-activating particles, for example, borosilicate glass Corning 7740 with a density of approx. 2.23 g / cm 3 is ground in a ball mill and passed through a -325 mesh sieve to remove larger particles. The finely divided glass is then mixed with water and left to stand for 5 minutes until the heavier particles have settled. The protruding part is then poured off and centrifuged at up to 1100 G for 7 minutes. The centrifugate is then dried at 100 ° C. in vacuo. The particle size obtained is 0.4-20 μm, preferably an average of 4 μm.
This gives uniform suspensions that are not restricted to the edge parts and remain without stirring, with the possibility of damage-free separation by centrifugation. The size distribution of the particles depends on their density. Since the particle surfaces are irregular or uneven, the frictional forces exerted by the liquid cannot be precisely determined. A numerical size range of the suitable particles cannot therefore be given, since the precipitation or sedimentation of each
