Abstract
1480120 N<SP>4</SP> substituted 2,2<SP>1</SP>-cyclocytidine derivatives KOHJIN CO Ltd 29 Nov 1974 [29 Nov 1973 4 Dec 1973 6 Dec 1973] 51914/74 Heading C2C Novel compounds of Formula I wherein Y represents a hydroxyl group of an amino (-NH 2 ) group; and R<SP>1</SP> and R<SP>2</SP>, which may be the same or different, each represents a hydrogen atom or an acyl group having the formula wherein R represents a straight or branched chain alkyl group having from 1 to 19 carbon atoms; an aryl group having from 6 to 10 carbon atoms which may be substituted with at least one substituent selected from a halogen atom, an alkyl group having from 1 to 4 carbon atoms, an alkoxy group having from 1 to 4 carbon atoms and a nitro group; an aralkyl group having 7 to 12 carbon atoms in which the aryl moiety thereof may be substituted with at least one substituent selected from a halogen atom and an alkyl group having from 1 to 4 carbon atoms; a cycloalkyl group having from 4 to 10 carbon atoms; and a polycyclic cage-type hydrocarbon group having from 7 to 14 carbon atoms; with at least one of R<SP>1</SP> and R<SP>2</SP> being the acyl group as defined above, and acid addition salts thereof, with the proviso that R<SP>1</SP> may not represent an acyl group having the formula wherein R represents a methyl group when R<SP>2</SP> represents a hydrogen atom and Y represents a hydroxy group are prepared either by acylation of a compound IVb or a compound IVa followed by removal of the protecting group and optional further acylation. N<SP>4</SP>-aminocytidine is prepared by reaction of cytidine with hydrazine hydrate. N<SP>4</SP>-hydroxy-2,2<SP>1</SP>-cyclocytidine is prepared by reaction of N<SP>4</SP>-hydroxycytidine with phosphorus oxychloride. N<SP>4</SP> - amino - 2,2<SP>1</SP> - cyclocytidine hydrochloride is obtained by HCl/methanol hydrolysis of 3<SP>1</SP> - O - acetyl - N<SP>4</SP>- amino - 2,2<SP>1 </SP>- cyclocytidine obtained by reaction of N<SP>4</SP>-aminocytidine with phosphorus oxychloride in ethyl acetate. Pharmaceutical compositions in conventional forms for oral, intravenous, intraperitoneal, subcutaneous or rectal administration and having anti-leukaemic activity comprise an above novel compound and a carrier therefore. Treatment of leukaemia in non-human animals is also claimed.
