Abstract

1476881 Penicillins BRISTOL MYERS CO 27 Jan 1975 [28 Jan 1974] 3540/75 Heading C2C Synthetic penicillins are prepared from penicillin G or penicillin V by (a) forming solutions of silyl esters of said penicillin G or penicillin V in anhydrous unreactive solvents in the presence of weak tertiary amines, (b) adding halogenating agents at temperatures below 0‹ C. to form solutions of the imino-halides, (c) mixing said solutions with alcohols at below - 20‹ C. to form solutions of the iminoethers, (d) adding to said solutions of the iminoethers at - 50‹ C. volumes of water no greater than 10% by volume of the imino-ether solutions to produce 6-aminopenicillanic acid in a single phase system, and (e) after the addition of weak tertiary amines acylating said 6-aminopenicillanic acid in the same solutions at about - 40‹ C. with carboxylic acid chlorides.
