Abstract

1475015 Centrifuge tubes J K and SUSIE L WADLEY RESEARCH INSTITUT & BLOOD BANK 22 Aug 1974 [30 Jan 1974] 36933/74 Heading B2P [Also in Division G1] A device for use in the treatment of a liquid sample with another liquid and the centrifugation of the product comprises an enclosed elongated vessel having a first end and a second end each end having an injectable closure means, a chamber 12 which is at less than atmospheric pressure, an enclosed end chamber 16a containing the other liquid and communication with one of the injectable closure means 16, and injection needle means 34 positioned within end chamber 16a for passing fluid from the end chamber through closure means 16 as shown and into the evacuated space. In use for the determination of microbial pathogens in blood, the evacuated space may initially contain a "liquid filter medium" which may comprise a sugar in concentration at least 40% in a pH 6.0 to 7.0 buffer containing a gel material, and the liquid in end chamber 16a may be a lysing agent which is non-toxic to micro organisms, for example a non-toxic solution of saponin. In use, the vessel is chilled to solidify the filter medium, push button member 32 at the end of stylus 34 is pressed to force the end of the stylus through closure means 16 to allow the lysing liquid to flow into the low pressure chamber 12, a blood sample is injected through closure means 14 using syringe 50, the vessel is warmed to a temperature which will liquify the filter medium but not destroy microbial pathogens then centrifuged, when the microbial pathogens pass into the filter medium layer. Either the filter medium layer or the remaining sample layer may then be removed using syringe inserted through closure means 14. Specification 1458147 is mentioned.
