1462407 p-lsobutyl hydratopic acid AZIENDE CHIMICHE RIUNITE ANGELINI FRANCESCO ACRAF SpA 22 July 1974 [11 April 1974] 32364/74 Heading C2C The anti-inflammatory, analgesic and antirheumatic drug "Ibuprofen" of formula is made from a compound of formula where R is C 1-5 alkyl by heating the latter at 200-240‹ C. (a) with a strong acid in aqueous medium or (b) in the dry with a salt of a strong acid and an organic base. The compounds of Formula II may be formed in situ (1) by the reaction of an alkyl α-methyl-α<SP>1</SP>-methyl succinate with vinylisobutyl ketone or (2) by reacting an acetoacetic ester with an alkyl α- halopropionate and then with vinyl isobutyl ketone.
