Abstract

1434044 Resin-drug composites BIOREX LABORATORIES Ltd 2 July 1974 [17 July 1973] 34040/73 Headings C3A C3P C3R and C3U [Also in Divisions C2 and D1] The invention comprises pharmaceutically active pentacyclic triterpenes which have been immobilized by chemical attachment to a polymeric organic or inorganic carrier; and their preparation by reacting the triterpene with the carrier, either of which may be chemically activated as appropriate. Specified triterpenes are glycyrrhetinic, 3-dehydroglycyrrhetinic, oleanolic, 11-oxo-oleanolic and ursolic acids and the hemisuccinates, hemiglutarates and amides thereof, and hederagenin. The examples relate exclusively to glycyrrhetinic acid hemisuccinate which may be activated by reaction with isobutyl chloroformate. The carrier may be a polysaccharide, e.g. agarose, a cross-linked dextran, starch, cellulose or cellulose acetate, these being activated by successive reactions of the diol system thereof with cyanogen bromide and 1,6-hexanediamine (R = hexamethylene). The carrier may also be chloromethylated "hydrocarbon" beads (activated by reaction with 1,6 - hexanediamine) chloromethylated polystyrene beads (unactivated), nylon (activated by reaction with 3-(dimethylamino)- propylamine), polyacrylamide (activated by diazotization followed by reaction with 1,6- hexanediamine), polyethylene terephthalate or ethylene-maleic anhydride copolymer (activated by reaction with 1,6-hexanediamine and cyclohexylcarbodiimide) or polyvinyl alcohol (activated by reaction with p-nitrobenzoyl chloride followed by reduction of the nitro group to amino with sodium dithionite).
